Distinct differentiation-inducing activities of gamma-interferon and cytokine factors acting on the human promyelocytic leukemia cell line HL-60.

The human promyelocytic leukemia cell line HL-60 and monoblastic leukemia cell line U937 undergo differentiation when induced by lymphokine and cytokine preparations. Growth inhibition, acquisition of immunoglobulin Fc receptors, increased expression of monocyte-related surface antigens, and an increase in lysosomal enzyme contents accompany maturation induced by gamma-interferon and other cytokine factors tested. Additionally, increased receptors for chemotactic peptide (fMLPR), increased hydrogen peroxide release in response to phorbol myristic acetate stimulation, and the release of prostaglandins (PGE2 and 6-keto-PGF1a) follow exposure to lymphokine and cell line sources of myeloid colony-stimulating activity (CSA). Gamma-Interferon (gamma-IFN) induced fMLPR in HL-60 (only at 1000 units/ml) but not in U937. Additionally, gamma-IFN did not induce prostaglandin release in either cell line. These myeloid colony-stimulating activity-associated differentiation-inducing factors were obtained from the human hepatoma++ cell line SK-Hep and bladder carcinoma cell line 5637, which were free of interferon activity. The 2-day phytohemagglutinin-induced lymphokine contained no detectable CSA and was a good source of differentiation activity. A simple, rapid assay for a new human CSA with pluripotent hematopoietic stimulating activity (pluripoietin) is described based on stimulation of [3H]glucosamine incorporation. Cell line conditioned media containing pluripoietin, purified pluripoietin, and gamma-IFN are active in this assay. These myeloid leukemia cell line differentiation factors are thus different from interferon and conventional CSA. These results suggest that endogenous human cytokines may have a role in the differentiation of leukemic as well as normal myeloid cells.

Effect of interferon on chemiluminescence and hydroxyl radical production in murine macrophages stimulated by PMA.

Considerably augmented chemiluminescence (CL) occurred when murine peritoneal resident macrophages (MPs), pretreated with murine interferon (MuIFN)-alpha within 24 hr, were stimulated by 4-beta-phorbol, 12-beta-myristate, 13-beta-acetate (PMA). Augmentation of CL generation ceased when incubation in the presence of MuIFN was continued for 48 hr. As 12 hr preincubation with MuIFN procured optimal CL generation, the various reactive oxygen species (OH, O2-., H2O2) were measured at that point. The hydroxyl radical (OH.) level in MuIFN-treated MPs was 19.44 times higher than in MuIFN-untreated MPs. However, the levels of O2-. and H2O2 generation were the same in both MuIFN-treated and untreated MPs. Moreover, by using the inhibitors lipoxygenase and cyclo-oxygenase, we established clearly that CL and OH. generation in MuIFN-treated MPs is due to the lipoxygenase pathway of arachidonic acid metabolism.

Elevated levels of prostaglandin E2 and thromboxane B2 in depression.

Plasma prostaglandin E2 (PGE2) and thromboxane B2 (TXB2) levels were measured in 30 depressed outpatients. Plasma PGE2 was increased in all but one of these patients (p less than 0.001) and all had increased levels of TXB2 (p less than 0.001). Since monamine oxidase inhibitors inhibit brain prostaglandin synthesis, and tricyclic antidepressants may antagonize prostaglandin actions, these observations suggest new approaches to depression research.

Severe acute pancreatitis associated with peliosis hepatis in a patient with systemic lupus erythematosus.

Acute pancreatitis is an unusual complication of systemic lupus erythematosus but can also stem from immunosuppressive therapy. Although abnormal liver tests are commonly seen in SLE, peliosis hepatis is very rarely described. We report here the first case of SLE associating a severe acute pancreatitis with peliosis hepatis who responded well to the immunosuppressive therapy. As suggested by the favourable outcome in this case, the presence of peliosis hepatis in SLE cannot not be held as a strong argument against an aggressive immunosuppressive therapy.
