[Relation between intravenous glucose tolerance tests and oral glucose tolerance tests in severely obese patients. II].

OGTT (75 g glucose) and IVGTT (25 g glucose) were compared in 20 extra-obese patients, both glycaemia and insulin levels being calculated. In only 2 out of 7 cases was the glucose intolerance revealed by OGTT confirmed by IGVTT and IVGTT was never able to identify the excessive insulin output revealed by OGTT. It can therefore be confirmed that in the extra-obese even more than in normal patients, IVGTT and OGTT investigate different functions of glucose tolerance and acute insulin output.

Effects of fatty acids on activity and calmodulin binding of Ca2+-ATPase of human erythrocyte membranes.

Activation and inhibition of Ca2+-ATPase of calmodulin-depleted human erythrocyte membranes by oleic acid and a variety of other fatty acids have been measured. Low concentrations of oleic acid stimulate the enzyme activity, both in the presence and in the absence of calmodulin. Concomitantly, the affinity of the membrane bound enzyme to calmodulin progressively decreases due to competitive interactions of calmodulin and oleic acid with the enzyme. Removal of oleic acid from the membrane by serum albumin extinguishes the activating effect of oleic acid and restores the ability of the enzyme to bind calmodulin with high affinity. High concentrations of oleic acid induce an almost complete and irreversible loss of enzyme activity which cannot be abolished by removal of oleic acid. Despite a complete loss of enzyme activity, binding of calmodulin to membranes is approximately normal after removal of oleic acid. Activities of (Na+ + K+)-ATPase, Mg2+-ATPase and acetylcholine esterase, as well as the total protein content, show no gross changes upon treatment of membranes with increasing amounts of oleic acid, which seems to exclude that membrane solubilisation by oleic acid causes an inactivation of the enzyme.

[Fructose versus glucose. The effects on metabolic parameters in patients with "severe obesity"].

In 15 subjects affected by "severe" obesity, some functional parameters were measured: glycemia, insulinemia, lacticidemia, uricemia, triglyceridemia, following oral load of fructose (100 gms), and separately, of glucose (100 gms). The analysis of the results obtained revealed a trend significantly different in the two cases in the glycemic and insulinemic profiles, whereas no statistically significant variations were observed with reference to the trend of the other parameters taken into consideration.

Endogenous calmodulin and Ca2+-ATPase activity of human erythrocyte membranes.

Treatment of calmodulin-depleted erythrocyte membranes with oleic acid solubilizes only a minor fraction of membrane proteins which, however, contains more than 90% of the total endogenous calmodulin. Concomitantly, Ca2+-ATPase is completely and irreversibly inactivated. By treatment with oleic acid in the presence of Ca.ATP the inactivation of Ca2+-ATPase is abolished to a great extent, the loss of endogenous calmodulin, however, is not affected. Thus, a major participation of endogenous calmodulin in the regulation of Ca2+-ATPase of erythrocyte membranes seems to be improbable.

[Low-calory dietetic treatment of gross obesity].

Reference is made to 96 grossly obese subjects classed as "resistant" to medical treatment and examined as possible candidates for surgery. 31 were eventually subjected to jejunal-ileal bypass. The main difference between these patients and those with slight or average spontaneous obesity was not so much their weight, but its self-maintenance. Weight patterns did not reflect calorie intake, though they pointed to a change in the energy balance when taken as a whole. In some cases that proved resistant both to severe hypocalorific treatment and the by-pass, no reason could be found for the discrepancy between calorie intake and the maintenance of weight. Some correlations between weight and certain endocrine, biohumoral and genetic features offer an idea of the metabolic orientation in gross obesity, but they fail to provide a satisfactory pathogenetic explanation.

[Determination of the concentration of free magnesium ions in hemolysates of oxygenated and deoxygenated packed human erythrocytes].

Concentrations of ionized magnesium in hemolyzates of oxygenated and deoxygenated human erythrocytes were measured directly by means of an ion exchange method. At total concentrations of hemoglobin of 8 mmol/l H2O and total magnesium of 3.6 mmol/l H2O, mean values of [Mg2+] were found to be 0.45 in the oxygenated state and 0.57 mmol/l H2O in the deoxygenated state. The difference of 0.12 +/- 0.1 mmol/l H2O is considerably smaller than corresponding literature data obtained from equilibrium calculation.

[Animal experimental studies of positive tumor demonstration with radio-iodine labelled albumin macroaggregates].

131I-labeled macroaggregates were injected into the femoral artery of rats bearing a Yoshida sarcoma at the lower extremity and of healthy control animals. The radioactivity was measured in tissue samples taken from the tumour and in unaffected muscle tissue. The tumour tissue showed a considerably higher uptake of radioactivity than the not-neoplastic tissue of control animals. The difference was increased when more time elapsed between the injection and the measurement. After 48 hours the ratio was 25:1 in favour of tumour tissue. In contrast to the transplanted Yoshida sarcoma, hepatomas in Wistar rats induced by N-nitroso-morpholin showed a considerably smaller increase of radioactivity than the control group when injected with 131I-MAA. The fixation ratio between tumour and normal liver tissue was 1:5.9. In contrast to autochtone liver tumours rats with transplanted tumours are considered as a suitable model for testing labeled albumin particles like 131I-, 111In- or 99mTc-labelled albumin microspheres developed for angioscintigraphic purposes.
