Ribonuclease P: an enzyme with an essential RNA component.

The activity of ribonuclease P on precursor tRNA substrates from Escherichia coli can be abolished by pretreatment of this enzyme with micrococcal nuclease or pancreatic ribonuclease A, as well as by proteases and by thermal denaturation. Highly purified RNase P exhibits one prominent RNA and one prominent polypeptide component when examined in polyacrylamide gels containing sodium dodecyl sulfate. The buoyant density in CsCl of RNase P, 1.71 g/ml, is characteristic of a protein-RNA complex. The activity of RNase P is inhibited by various RNA molecules. The presence of a discrete RNA component in RNase P appears to be essential for enzymatic function. A model is described for enzyme-substrate recognition in which this RNA component plays an important role.

Novel activity of potato nucleotide pyrophosphatase.

The classical Kornberger-Pricer procedure for purification of potato nucleotide pyrophosphatase (EC 3.6.1.9) has been modified to yield a preparation purified 2500-fold. In addition to the known activity against pyrophosphate linkages in pyrophosphates located at the 5'-OH of nucleosides, and phosphodiester linkages in aryl esters of nucleoside-5'-phosphates, the enzyme has now been shown to catalyze the cleavage of: (a) aryl esters of nucleoside-3'-phosphates and orthophosphates, (b) nucleotide pyrophosphate linkages of the type (3')-pp-(3'), and (c) pm7G from m7GpppGm-terminated fragments of viral mRNA. Activities against aryl esters of nucleoside-3'- and 5'-phosphates, and NAD, were shown to be due to the same protein by three criteria: (a) constant ratio of activities during purification and gel electrophoresis, (b) identical chromatographic properties in various systems, and (c) similarities in pH-dependence, heat inactivation, and the effects of cations and other substances. Since potato nucleotide pyrophosphatase does not exhibit exonuclease or phosphatase activities against natural substrates for the latter enzymes, but does cleave synthetic aryl esters of nucleotide-3'- and 5'-phosphates and of orthophosphate, it follows that these substrates are not suitable for detection of such activities in higher plants.

Mung bean nuclease: mode of action and specificity vs synthetic esters of 3'-nucleotides.

Mung bean nuclease hydrolyzes synthetic esters of 3'-nucleotides to nucleosides and phosphate esters; esters of 2'-nucleotides, and 2'--> 5' internucleotide linkages, are resistant. Esters of ribonucleotides are cleaved at 100-fold the rate for deoxyribonucleotides, the increased rate being due to presence of the 2'-hydroxyl and not to differences in conformation. Introduction of a 5'-substituent leads to a 3-fold increase in rate. The rates of hydrolysis vary up to 10-fold with the nature of the base, in the order adenine > hypoxanthine > uracil; and up to 6-fold with the nature of the ester radical. This form of cleavage of esters of 3'-nucleotides is also characteristic for nuclease-3'-nucleotidase activities from potato tubers and wheat, suggesting that one type of enzyme is responsible for all these activities.
