Characterization of differentiation antigens of the mouse mammary epithelial cell (MME antigens) carried on the mouse milk fat globule.

The characterization of mouse mammary epithelial (MME) antigens from the mouse milk fat globule (MMFG) is described. Heterologous antisera against fat-free MMFG material identifies by indirect immunofluorescence techniques, cell-type-specific MME antigens on the surface of mouse mammary epithelial cells and MMFGs and not on cells of other tissues. Mammary epithelial cells of all mouse strains tested were positive. However, the intensity of staining differed. Specificity of anti-MME for MME antigens is demonstrated by competition and cross-absorption experiments. The MMFG is composed of 5 main proteinaceous components, 3 of which contain carbohydrate and which are shown to contain the specific MME antigens.

Immunologic methods for the identification of cell types. II. Expression of normal mouse mammary epithelial cell antigens in mammary neoplasia.

Mouse mammary epithelial (MME) cell antigens were cell type-specific and were retained, to a large extent, by MME cells after neoplastic transformation. These MME cell antigens were expressed in mammary tumors of BALB/c, C3H, GRS/A, RIII, and Is/Bi mice tested and were not expressed in tumors whose normal counterpart was other than mammary tissue. They were not dependent on cell culture conditions or on the presence of murine mammary tumor virus; therefore, they can be used in an ubiquitous cell type marker for MME cells in both normal and neoplastic tissues.

Response to prolactin and ovarian steroids of normal mammary epithelial cell cultures.

Mouse mammary epithelial cells in confluent primary monolayer cultures retain responsiveness to the specific hormones that induce mammary growth in vivo. Simultaneous stimulation by prolactin, progesterone and estrogen, in the presence of 5 percent fetal calf serum, is required to induce an increase in both thymidine uptake into DNA and in cell replication (as judged by mitotic indexes) over the hormone-free control. This increase in mitogenic response could not be elicited in either mouse fibroblasts or in mouse mammary tumor cells, the latter known to be hormone insensitive.

Surface differentiation antigens of human mammary epithelial cells carried on the human milk fat globule.

Rabbit antibodies against components of the human milk fat globule bind specifically to normal human breast epithelial cells and cell lines derived from breast carcinomas, as well as to the outer surface of the human milk fat globule. Variation in indirect immunofluorescence staining in both intensity per cell and percentage of cells stained is observed for the different brest cell lines. Cells derived from other epithelial and other ectodermal tissues, fetal fibroblasts, cells of the blood buffy coat, and even fibroblasts of the breast itself do not bind the antibodies. This suggests that these antibodies are detecting cell-type-specific antigens. These normal breast epithelial cell antigens are on the cell surface and their expression is stable in long-term cultured cell lines, even after much chromosomal variation in a given line. By affinity chromatography, three distinct antigenic components can be isolated from the milk fat globule, one of which contains carbohydrate. These differentiation antigens of the human breast epithelial cell are not only useful as specific cell-type markers, but also can provide a tool to study the role of the cell surface in normal and neoplastic mammary development.

Hormone induction of specific protein synthesis in midpregnant mouse mammary cell culture.

Monolayer primary cell cultures of midpregnant mouse mammary cells were subjected to hormone stimulation under strictly defined conditions. Hormonal response was measured in terms of increase in rate of synthesis of mouse casein, using a double antibody precipitation technique. Cells stimulated by insulin plus prolactin plus cortisol plus estradiol plus progesterone showed a marked increase in rate of mouse casein synthesis over the controls. This specific product synthesis, which remains inducible in these cells for at least ten days, was detected either by labelled phosphoric acid or labelled amino acid incorporation. The mouse casein synthesized was identical, as judged by the identification techniques used, to that of mouse milk. Mouse midpregnant mammary explants in organ culture require insulin plus prolactin plus cortisol to express their full lactogenic capabilities. Nevertheless, when the same cells are dispersed and grown in monolayers they require ovarian steroids to elicit a lactogenic response as shown in this study. Ovarian steroids, therefore, are necessary in lactogenesis, although the fundamental nature of their action remains to be established.

Immunologic methods for the identification of cell types. I. Specific antibodies that distinguish between mammary gland epithelial cells and fibroblasts.

Antibodies were produced against intact mouse mammary epithelial cells and cleared fat pad fibroblasts; after appropriate absorption, two specific antibody preparations were obtained. The antimammary epithelial cell preparation did not appear to be strain- or species-specific. The antimammary fibroblast preparation recognized both mammary and fetal fibroblasts. These findings suggested that each cell type possessed distinct immunogenic components. These components were characteristic of each cell type and could be used to identify the respective cells by an immunofluorescence technique. Characterization of the antigenic component of mouse mammary epithelial cells demonstrated that this antigen was released during enzymatic cell dissociation and was regenerated underin vitro culture.
