Apple II computer software for DNA and protein sequence data.

In our attempt to make the aid of a computer for the molecular biology laboratory available on a low-cost basis, we have improved the programs that we published recently. We have experienced in our laboratory, and have learned from others, that the average lab has an increasing demand to use a time-sharing-independent computer for the bulk of data handling. A microcomputer can give this independence. At the same time, a microcomputer can serve as a terminal for the few computer jobs, such as data communication or extensive search programs, which have to be conducted on a time-sharing basis. In this paper, we describe the programs which can be executed on a Apple II microcomputer using the Apple Language System. This system gives the researcher the opportunity to learn to handle sequence data, and it also includes a word processing system useful in writing letters and manuscripts and storing text.

Characterization of virus-specific RNA synthesized in bovine cells infected with bovine viral diarrhea virus.

Infection of bovine kidney cells with bovine viral diarrhea virus resulted in the synthesis of a single species of virus-specific RNA. Electrophoresis of this RNA on agarose-urea and agarose-formaldehyde gels indicated that it had a molecular weight of 2.9 X 10(6), corresponding to 8,200 bases (8.2 kilobases). This 8.2-kilobase RNA was resistant to RNase A treatment at 1 microgram/ml but was digested at higher concentrations of RNase (10 micrograms/ml). Sedimentation on neutral sucrose gradients indicated that the majority of this RNA (98%) sedimented at 21S, with a small amount sedimenting at 33S. Sedimentation on formaldehyde-containing sucrose gradients resulted in the conversion of all of the RNA to the faster-sedimenting form. At no time after infection were we able to detect virus-specific RNA species of lower molecular weight than the 8.2-kilobase RNA. The implications of these findings with respect to the means of replication of various togaviruses are discussed.

Apple II software for M13 shotgun DNA sequencing.

A set of programs is presented for the reconstruction of a DNA sequence from data generated by the M13 shotgun sequencing technique. Once the sequence has been established and stored other programs are used for its analysis. The programs have been written for the Apple II microcomputer. A minimum investment is required for the hardware and the software is easily interchangeable between the growing number of interested researchers. Copies are available in ready to use form.

Bulimia: an analysis of moods and behavior.

An innovative sampling method was used to compare normal-weight bulimic patients to normal controls on four dimensions: (1) overall moods, (2) extent of mood fluctuating, (3) degree of social isolation, and (4) extent of food-related behavior. In addition, a sequence analysis was conducted to explore the impact of binging and purging episodes on various affective states among the bulimics. Results indicated that the bulimics experienced significantly more dysphoric and fluctuating moods than normal controls and that they spent much more time alone and in food-related behavior than the control group. The discussion presents a hypothetical integration regarding the etiology and maintenance of the bulimic behavior based on the results of the data and post-trial interviews with the bulimic patients.

Parathyroid hormone acutely increases polyphosphoinositides of the rabbit kidney cortex by a cycloheximide-sensitive process.

Parathyroid hormone(PTH) rapidly increases the concentrations of diphosphoinositide and triphosphoinositide in rabbit kidney cortex. Cycloheximide pretreatment abolishes these effects of PTH. These findings are similar to those reported for adrenocorticotropin and cyclic AMP action in the adrenal cortex, and suggest a common mechanism. Cycloheximide-sensitive effects of PTH, e.g., phosphaturia, may require polyphosphoinositides and/or other phospholipids.

Thirty years of research on the subjective well-being of older americans.

Thirty years of research among older Americans on life satisfaction, morale, and related constructs has yielded a consistent body of findings. Parallel results for measures of these contructs and high intercorrelations justifies considering them in terms of a single summary construct, subjective well-being. As this research has relied almost exclusively on survey measures, interpretations are limited to the social-psychological level of people's day-to-day verbal behavior. This research shows reported well-being to be most strongly related to health, followed by socioeconomic factors and degree of social interaction, for the general population of Americans over 60. Marital status and aspects of people's living situations are also conclusively related to well-being. Age, sex, race, and employment show no consistent independent relation to well-being. In addition to indicating that negative life situation exigencies, such as poor health and low income are related to lower well-being, the results tentatively indicate that these exigencies create a greater vulnerability to the impact of other negative conditions.

Clinical applications of flow cytometry in hematology and immunology.

OBJECTIVE: To review the major applications of flow cytometry in the diagnosis of hematologic and immunologic disease. DATA SOURCES: Professional literature and authors' experience. DATA SYNTHESIS: Flow cytometry is evolving as a major diagnostic tool for evaluating hematologic disease, monitoring HIV infection and assessing peripheral blood and bone marrow for CD34 positive stem cells in marrow transplantation. This review highlights the opportunities and pitfalls of this area of diagnostic laboratory medicine. CONCLUSION: The applications of flow cytometers in diagnostic hematology and immunology are expanding, providing new opportunities for enhanced precision in diagnosis. As in any relatively new field much work remains in order to optimize accuracy and efficiency while minimizing cost.

Aspergillus and lung transplantation.

BACKGROUND: Aspergillus infection is a known complication of transplantation. METHODS: We describe our experience with 37 patients who received lung transplants over 2 years at Loyola University Medical Center. All patients who had evidence of aspergillus on culture of clinical specimens or had biopsies with hyphal forms consistent with aspergillus were categorized according to the clinical manifestations. Important risk factors were analyzed in comparison with other lung transplant recipients during the same period. RESULTS: The incidence of invasive aspergillosis was high (16%). No patient with disseminated disease survived. Locally invasive disease responded well to treatment with amphotericin B and itraconazole. CONCLUSIONS: Lung transplantation patients may have a higher incidence of aspergillosis as compared with other transplantation groups. Prophylactic measures need to be explored.
