The effect of HAART on humoral immune response in primary HIV-1 infected patients.

The effect of Highly Active Antiretroviral Therapy (HAART) on binding and neutralizing antibody responses to human immunodeficiency virus type-1 (HIV-1) during primary and chronic infection was investigated. Seven patients HAART treated during primary infection, six HAART treated during chronic infection and five patients treated only with ZVD (Zidovudine) were analysed. HAART inhibited the development of anti env antibodies during primary infection. Administering HAART during primary infection usually did not substantially affect the development of weak neutralizing antibody responses against autologous virus. However, we demonstrated that very early treatment, during seroconversion, induce in some cases, a strong neutralizing antibodies against autologous virus. These results may be relevant for understanding how HAART may elicit a strong protective responses and may be useful in developing new strategies designed to achieve a long term control of the HIV infection.

[Attempts to characterize dexamethasone receptors present in rat kidney cytosol].

Density gradient centrifugation analysis has been used to study rat kidney receptors for dexamethasone. Density gradients performed with cytosol incubated with 3H-dexamethasone in a Tris 10 mM, EDTA 1.5 mM, glycerol 5%, monothioglycerol 12 mM (pH 7.5) buffer never gave any good evidence of a 85 radioactivity peak probably due to instability of the complex. The stability of the complex for at least 30 h was achieved by homogenizing the tissue and incubating cytosol in a distilled water, glycerol 5% monothioglycerol 12 mM solution. Density gradient centrifugation analysis in this way showed radioactivity peak at the bottom of the tube probably due to the aggregation of the complex. Since this drawback is inherent in the technique it must be concluded that density gradient centrifugation analysis cannot be used to study the dexamethasone-receptor complex of the kidney.

[Use of a new amino acid mixture in the diet therapy of patients with classical phenylketonuria].

The treatment of classic PKU is based on the dietary restriction of phenylalanine. The present study was planned to evaluate the effectiveness of a new amino acid mixture (ASP-Nestlé) in the dietary treatment of ten PKU patients (age 2 to 9 years, mean 5 years). The children were given this product for a minimum of 3 and a maximum of 12 months (mean 8 months). The aminoacid mixture, diluted in water or various flavored drinks, was favorably accepted by the patients. No clinical signs of hypophenylalaninemia were noticed and the children grew regularly in height and weight. The biochemical parameters, evaluated before during and after the treatment, showed a good nutritional balance. The plasma phenylalanine control was excellent, when compared to the plasma phenylalanine range obtained with other dietary regimens.
