Bcl-2 oncoprotein expression in acute myeloid leukemia.

BACKGROUND: In lymphoproliferative diseases the expression of Bcl-2, a mitochondrial oncoprotein capable of blocking apoptosis, is well-documented, while little research has been carried out on its distribution in myeloproliferative conditions. METHODS: Using immunocytochemical methods, 63 cases of acute myeloid leukemia (AML) at onset and 10 relapses were studied to investigate Bcl-2 expression and any possible correlations with subtypes of the FAB classification, sex, age or white cell peripheral blood count at onset. RESULTS: Bcl-2 is present in 87.3% of AML cases at onset and in 100% of relapses. In 68.3% of cases at onset and in 90% of relapses the protein is present in more than 20% of the blasts. Relapses always show higher percentages of positive expression than those seen at onset. Our results demonstrate no statistical correlations between the expression of the oncoprotein Bcl-2 and FAB subtypes, sex, age, or white cell peripheral blood count. CONCLUSIONS: The majority of blasts from AML patients express the oncoprotein Bcl-2, which is able to protect leukemic cells from apoptosis. Since numerous chemotherapies are cytotoxic in that they induce apoptosis, we feel that in vitro studies of cells from AML patients are necessary in order to broaden our knowledge about the effects of the most common therapeutic drugs and of those substances which, alone or in association, can modulate Bcl-2 expression.

[Diagnostic value of renal scintigraphy in the neonatal period: presentation of some clinical cases].

The valuation of renal function using radiotracers represents an attractive methodology to value most renal and upper urinary tract diseases in newborns. Restrincting damage, lower irradiation and lack of dangerous side-effect are particularly desiderable in newborns. The radiotracers of choice in our studies were Tc-99m-labeled radiopharmaceutical (DTPA and DMSA), because of their favourable dosimetry. The given dose has been calculated according to the table set forth by the EAMN-Pediatric Task Group. We report some particular cases in which renal scintigraphy represented a good alternative in comparison with more injurious methods in diagnostic classification of newborns.

Plural origins of molecular homochirality in our biota Part II. The relative stabilities of homochiral and mixed oligoribotides and peptides.

By computer simulations--molecular mechanics and molecular dynamics with the amber force field (Weiner et al, (1986), J. Comp. Chem. 7, 230-252)--we have determined the stabilities of oligoribotide strands built with D- and L-riboses, and of peptide chains with D- and L-amino acid residues. In particular, complementary double-chains of oligoribotides were studied, since they are an important feature of the growing mechanism of modern nucleic acids. Peptide chains on the other hand, grow without need of a template. We found that mixed oligoribotides are less stable than homochiral ones, and that this chiral effect is less noticeable in peptide chains. The results support the interpretation that L-riboses act as terminators to the template-assisted growth of oligo-r-GD (enantiometric cross-inhibition; Joyce et al., (1987), Proc. Natl. Acad. Sci. USA 84, 4398-4402). Based on this effect, a chemical pathway is proposed which could, under assumed prebiotic conditions, bypass the hindrance of homochiral growth.
