Tissue distribution of rat macroglobulins in tumour-bearing rats.

Rat macroglobulins were determined in blood sera and extracts of tissues of intact rats and rats with Walker carcinoma by rocket immunoelectrophoresis. The serum levels of alpha1-macroglobulin (alpha1MG) alpha2-macroglobulin (alpha2MG) and pregnancy-associated alpha1-glycoprotein (alpha1PAG) were 1.86 +/- 0.07 mg/ml, 0.12 +/- 0. 02 mg/ml and 18.32 +/- 4.07 AU/ml respectively in control rats. Maximum concentrations of alpha1MG were found in heart, lung and spleen and lesser quantities were in liver and thymus, while alpha2MG and alpha1PAG were not found at all in tissue extracts from control rats. Serum alpha2MG and alpha1PAG concentrations increased more than 30-fold in tumour-bearing rats compared to control animals, while alpha1MG serum concentration was little changed. Increases in all three macroglobulins occurred in the tissues of tumour-bearing rats, particularly alpha1PAG. The tissue concentrations of alpha1MG and alpha2MG were similar and the tissue distribution was also similar with highest concentrations in heart and lung. Considerable quantities of the proteins were found in the tumour and part of peritoneum which made contact with the tumour. Changes in the protein concentrations in serum and tissue extracts of tumour-bearing rats suggest that all members of rat macroglobulin family are disturbed during the development of the Walker carcinoma, though only alpha2MG and alpha1PAG were substantially elevated.

[Immunoenzyme method of lactoferrin analysis].

The authors propose a highly effective method for isolation of lactoferrin from female colostrum. Lactoferrin is measured by the sandwich enzyme immunoassay. Its concentrations in the serum, urine, amniotic and cerebrospinal fluid in health are measured. The advantages of the method for lactoferrin purification and the practical significance of this protein are discussed.

[Pregnancy-associated alpha2-glycoprotein and trophoblastic beta1-glycoprotein in the diagnosis of pyoinflammatory diseases after Cesarean section].

Blood serum pregnancy-associated alpha 2-glycoprotein and trophoblastic beta 1-glycoprotein were measured in puerperants with the normal postpartum period and with that complicated by pyo-inflammatory diseases after abdominal delivery. The levels of both proteins were found reduced in the patients with pyo-inflammatory diseases developing after cesarean section as early as on day 1 postpartum. that of alpha 2-glycoprotein by 1.2 times, that of trophoblastic beta 1-glycoprotein twofold.

[The effect of steroid sex hormones on the biosynthesis of alpha 2-macroglobulin and pregnancy-associated alpha 2-glycoprotein in cultures of peripheral blood mononuclear cells].

The peripheral blood mononuclears are capable of intense biosynthesis of alpha 2-macroglobulin and of weak biosynthesis of pregnancy-associated alpha 2-glycoprotein. Sex hormones of men and non-pregnant women exert no influence on the protein biosynthesis. During pregnancy alpha 2-macroglobulin biosynthesis is shortly activated, although it does not depend on the influence of sex hormones. All the steroid sex hormones provide a short-term biosynthesis of this protein during the II trimester of pregnancy, while testosteron inhibits it during the III trimester. Possible mechanisms of control of biosynthesis of these proteins are discussed.

[Complex evaluation of the feto-placental system in gestosis of different degree of severity].

Comprehensive studies of the fetoplacental system in pregnant women with gestoses of varying severity (n = 260) and in those with normal pregnancy (n = 200), carried out in pregnancy terms of 29 to 41 weeks, have revealed a lowering of the levels of trophoblastic beta-globulin and pregnancy-associated alpha 2-glycoprotein, elevated concentration of pregnancy-associated protein A, a higher incidence of placental types II and III (as evidenced by ultrasonic examination), reduced rate of growth of the fetal biometric parameters and a reduction of fetal systolic output, augmenting with the gestosis severity. A comprehensive assessment of the fetoplacental system permits obtain a more complete picture of the gestosis severity.

[An immunoenzyme method of determining pregnancy-associated alpha 2-glycoprotein].

Methods for purification of pregnancy-associated alpha 2-glycoprotein, obtained antibodies to it, and enzyme immunoassay of this protein are described. Enzyme immunoassay permits measurement of this protein in blood serum and mononuclear cell cultures in a wide range of concentrations. The method sensitivity may be improved by removing the antibodies cross-reacting with animal proteins.

[Pregnancy-associated alpha 2-glycoproteins in the diagnosis of postpartum suppurative and septic diseases].

The levels of pregnancy-associated alpha 2-glycoprotein (alpha 2-GP) were measured in the blood sera of puerperants with physiologic postpartum and postoperative (after cesarean section) periods, and with those complicated with pyoseptic diseases. alpha 2-GP levels were changed in the blood sera of puerperants with postpartum infectious diseases. The authors discuss the possibility of predicting the postpartum complications with the use of alpha 2-GP measurements.

[Use of a peroxidase marker to detect the diffusion of protein in gel].

The method for the assessment of the protein gel diffusion rate implies the employment of protein conjugates with horseradish peroxidase; the diffusion of these conjugates can be easily visualized by covering the gel with a cellulose acetate film wetted with a solution of the substrate specific for peroxidase. The necessary concentration of the protein can be selected with this method, as well as the time of the diffusion of the proteins tested as chemoattractants in studies on cellular chemotaxis under an agar layer.
