[Tissue levels of EGF and collagen in ovaries of women].

Tissue EGF concentration as well as collagen content were investigated in ovarian homogenates from both menstruating and menopausal women. EGF concentration was found to be higher, but not significant, in postmenopausal women, whereas total collagen as well as forms soluble in acetic acid and in this acid with pepsin were slightly higher in women with normal menstrual cycle. There were statistically significant negative correlations between above mentioned forms of collagen as well as ovarian EGF concentration in this group of patients. The potential role of epidermal growth factor in modulation of ovarian function and extracellular matrix remodelling within human ovary is discussed.

[Level of EGF in cord blood and biological maturity of the newborn].

The concentration of epidermal growth factor-EGF has been measured in umbilical vein serum among preterm and term newborns by means of specific RIA assay. The mean EGF concentration among term newborns was found to be higher than in preterm (0.236 +/- 0.094 vs 0.167 +/- 0.087 pg/ml; p < 0.05). We did not observed any correlation between EGF concentrations and the occurrence of respiratory distress syndrome in preterm babies. The role of EGF in human parturition has been discussed.

Epidermal Growth Factor↗

[Concentration of epidermal growth factor (EGF) in serum of women with endometrial cancer].

Serum Epidermal Growth Factor (EGF) concentrations were estimated in 40 women with endometrial carcinoma and in 10 women with proliferative endometrium. Medium EGF level in study group was 0.97 +/- 0.41 ng/ml whereas 0.78 +/- 0.19 ng/ml in control group. The highest values of EGF (1.39 +/- 0.44 ng/ml) were found in moderately differentiated adenocarcinomas (G2) whereas the low values (0.65 +/- 0.18 ng/ml) in poorly differentiated adenocarcinomas were noted. Statistically significant differences were observed among control group and moderately differentiated adenocarcinomas and between all groups of adenocarcinomas. In two patients with sarcoma uteri the lowest EGF (0.49 and 0.58 ng/ml) serum concentrations were found. Our results confirm that low EGF serum concentrations as well as EGF receptor on the cell surface probably may increase the risk of cancerogenesis in human endometrium.

[Finding antigens using immunoenzyme methods characterized by nonhistone proteins in hyperplastic and neoplastic endometrium in women].

Immune rabbit serum against NHCP extracted from woman endometrial adenocarcinoma were obtained to search for antigen in blood of patients with hyperplastic and neoplasmatic endometrium. The highest value of extinction were observed in carcinomas and hyperplastic endometrium values were similar. The lowest values were noted in control group with normal proliferative endometrium.

[Immunohistochemical localization of estrogen receptors in vesicovaginal fascia among female patients with stress incontinence--preliminary report].

Using monoclonal antibody assay technique 29 kD domain of estrogen receptor was found and partially quantitated by means of histological score in fibroblasts from vesico-vaginal fascia obtained during surgical procedure in incontinent women. This is an important proof that estrogen supplementation therapy is a reasonable approach in women affected by urinary stress incontinence.

Nonhistone chromosomal proteins (NHCP) in adenocarcinoma of human endometrium.

Concentration and electrophoretic pattern of nonhistone chromosomal proteins (NHCP) in cancer and normal endometrium were examined. In normal tissues the NHCP/DNA ratio ranged from 0.6 to 0.8 whereas in malignant tissues from 1.2 to 1.7. Mean nuclear concentration of NHCP was 14.16 pg/cell nucleus of normal endometrium and 33.35 pg/cell nucleus of cancer tissue. The two tissues differed in the electrophoretic pattern. Heavy fractions (> 60 kDa--36%) predominated in normal tissue whereas light fractions (< 30 kDa--48%) in malignant tissue samples. The percentage of intermediate fractions (30-60 kDa) was similar. Neoplastic endometrium shows, irrespective of the patient's age, a high NHCP content. It seems that NHCP, especially the fraction extracted with 0.35 M NaCl, because of its tissue and cell specificity are involved in neoplasm formation.

[Immunologic reaction differences of non-histone chromosome proteins (NHCP) in myometrium and leiomyoma of the uterus in women].

Immune sera against non-histone chromosomal proteins (NHCP) of myometrium and leiomyoma were raised in our laboratory in rabbits after injection with purified antigens. (NHCP) were extracted 0.35M NaCl from 6 myometrium and leiomyoma tissue samples. Sera contained precipitating and non-precipitating antibodies reacting in Ouchterlony and ELISA tests with homologous and heterologous antigens. Serum against leiomyoma NHCP reacted higher extinction values with myometrium than with homological antigens. NHCP tissue concentration (mg/g) was higher for myometrium in spite NHCP/DNA ratio bigger for leiomyoma. Above mentioned results indicate antigenicity alterations of leiomyoma tissue when compared to normal myometrium of woman uterus.

Epidermal growth factor receptor immunostaining and epidermal growth factor receptor-tyrosine kinase activity in proliferative and neoplastic human endometrium.

Epidermal growth factor receptor (EGFR) expression and EGFR-tyrosine kinase (EGFR-TK) activity were measured in proliferative (n = 12) and neoplastic (n = 31) human endometrium. Immunoreactivity of EGFR was related to clinicopathological features, estrogen receptor (ER) and progesterone receptor (PR) status, and patient outcome. All proliferative and 27 neoplastic specimens expressed the EGFR. Expression of the EGFR was higher in proliferative endometrium than in endometrial cancer (p < 0.0001). ER immunostaining was observed in 19 of the endometrial carcinomas, while PR expression was demonstrated in only 12 neoplastic specimens. EGFR expression was not related to the ER/PR immunostaining in endometrial carcinomas. Clinicopathological features (age, stage, histological type, grade or depth of invasion) and clinical outcome were unrelated to EGFR immunoreactivity. EGFR-TK activity was detected in 29 of 31 endometrial neoplasms with a 9 times higher mean activity in neoplastic than in proliferative endometrial specimens. There was no relationship between the EGFR-TK activity and EGFR immunostaining in human neoplastic endometrium (p = 0.77). A trend towards a poorer outcome of patients with the EGFR-TK activity above 40 pmol/min/mg was observed, but was not statistically significant. These results support the view that the EGFR expression is downregulated in endometrial carcinomas compared to proliferative endometrial specimens.

[Electrophoretic evaluation of blood serum proteins during corticosteroid treatment of women with chronic adnexitis].

In the period 1980-1986 in the II Department of Surgical Gynaecology, Medical Academy in Lublin 61 patients were treated with corticosteroids for chronic adnexitis. They were given prednisone (Encorton POLFA) 10 mg twice daily for 10 days. The clinical effect of regression of infiltrations was achieved in over half the cases. In the analysis of the clinical state, ESR value, leucocytosis and electrophoretic separation of serum proteins it was found that the effectiveness of the treatment was highest in the group of patients with initial ESR values greater than 10 mm after one hour. The mean values of white blood cell count before and after the treatment were without essential prognostic importance. The best results were found in patients without clinical evidence of acute inflammation but with alpha 2globulin proportion in electrophorogram typical for acute phase of inflammation.
