Comparative responses of vaginal mucosa to chronic pyrimidinone-induced irritation.

Intravaginal studies in guinea pigs, Cebus monkeys (Cebus apella), and beagles were made with an anti-herpes drug, 2-amino-5-bromo-6-phenyl- 4(3H)-pyrimidinone (ABPP). Concentrations of 5 to 40 mg/ml, suspended in saline or polyethylene glycol, were given for periods of 2 1/2 to 90 days. Very little gross evidence of injury was observed. Histologically, the regimens brought about increasing degrees of mononuclear cell infiltrations (histiocytes, lymphocytes, and plasma cells) in the vaginal lamina propria of the respective species. In the Cebus monkey and beagle the nonkeratinized vaginal lining became progressively thinned. The integrity of focal denuded areas was preserved by a process of homogenization in the lamina propria of the Cebus monkey. In contrast, the most severely affected areas in the canine mucosa were covered by what appeared to be a coagulated exudative layer. The effect of 90 days of treatment in the beagle did not noticeably increase the severity of the findings beyond that observed after 30 days. Considerable reversibility of the lesions occurred in guinea pigs held for 1 week and in Cebus monkeys held for 3 weeks posttreatment. Local cell-mediated immunity was believed to be implicit with the mononuclear cell infiltration in the lamina propria of the vagina.

Cyclodextrin nephrosis in the rat.

The renal toxicity of the Schardinger dextrins, alpha and beta-cyclodextrin, is manifested as a series of alterations in the vacuolar organelles of the proximal convoluted tubule. These changes begin as an increase of apical vacuoles and the appearance of giant lysosomes. The giant lysosomes characteristic of cyclodextrin nephrosis are notable because of the prominent acicular microcrystals embedded in the lysosomal matrix. Giant vacuoles devoid of acid phosphatase reaction product are found in advanced lesions. The vacuolar apparatus shows advanced changes prior to manifestation of lesions in mitochondria and other organelles. These observations indicate a role of the vacuologenic apparatus in the nephrotic process. Intracellular concentration of toxin via the lysosomal pathway represents a perversion of the physiologic function of the proximal tubule which ultimately leads to cell death.

Urinary proteins in Sprague-Dawley rats with chronic progressive nephrosis.

By means of polyacrylamide concentration and electrophoresis on cellulose acetate strips, protein fractions have been determined in the urine of male Sprague-Dawley rats from 6 1/2-28 mo of age. An increasing percentage of albumin was expressed in the albumin/globulin ratios of 0.72, in a group of rats excreting a mean concentration of 1.91 mg/ml of protein in the urine, and 2.24 in a group of rats excreting, on the average, 17.62 mg/ml. A relative decrease in globulin, particularly of the alpha2 fraction, was shown. From a statistical model based on the regression parameters for 24-hr protein excretion and urine protein concentration, it was estimated that the doubling time for protein excretion was 3.2-3.69 mo. This interval is roughly one-half that reported for the Wistar rat.
