Effect of Escherichia coli lipopolysaccharide on the glucagon and insulin binding to isolated rat hepatocytes.

Number and affinity constant of low affinity binding sites of insulin and glucagon to isolated hepatocytes decreased when the cells were incubated with Escherichia coli 0111:B4 lipopolysaccharide. This effect agrees with a non-specific binding of lipopolysaccharide to hepatocytes, similar to the well-recognized non-specific binding of albumin. Also, binding of different lectins to their glycoprotein receptors did not affect the [14C]lipopolysaccharide interaction with the cell membrane surface. Endotoxin depresses gluconeogenesis from lactate when the precursor was incubated with the cells for short time intervals. The longer the preincubation interval with lipopolysaccharide, the higher the inhibition of gluconeogenesis in the absence and in the presence of glucagon. The effect of endotoxin was also studied on the glucagon-induced synthesis of cyclic AMP and the glucagon binding. Levels of cyclic AMP and hormone binding decreased with increasing both endotoxin concentrations and preincubation intervals at which cells were in contact with endotoxin.

[Effects of starvation, a hyperprotein diet and treatment with nicotinamide on L-threonine aldolase and allothreonine aldolase in the rat liver].

In order to evaluate if the aminoacid L-threonine and its isomer allothreonine are metabolized by a single protein or by two specific enzymatic proteins, in the rat liver, we have studied the behaviour of L-threonine aldolase activity and allothreonine aldolase activity in rats which are injected with nicotinamide (1 g/ Kg) or have received a special diet containing 91% of casein or that have been starved for several days. The L-threonine aldolase enhances its activity in all three cases, while the allothreonine aldolase does not show significative changes of activity. These different behaviours of the two enzymatic activities, support the hypothesis that in the rat liver two proteins are necessary for the metabolism of the L-threonine and its isomer allothreonine.

Specificity of the reaction of rat liver L-serine dehydratase with aminothiols.

The degree, the mechanism, and the specificity of the inhibition of purified rat-liver L-serine dehydratase (EC 4.2.1.13) by L-cysteine and aminothiols have been investigated. All the aminothiols tested react with free pyridoxal-5'-phosphate (PLP); only L-cysteine reacts with PLP bound to the enzyme giving an absorption spectrum similar to that of the thiazolidinic ring formed in the chemical reaction. The reaction between L-cysteine and PLP bound to the enzyme has a particular behaviour, different from all the other aminothiols. A stereospecific interaction between L-cysteine and PLP bound to the enzyme, forming one of the two possible diastereoisomers, is postulated.

Accumulation and metabolism of uneven fatty acids present in single cell protein.

Liquipron and Toprina, obtained by growing yeasts (Candida maltosa and Candida lipolytica) on n-hydrocarbons, were investigated to ascertain the biological significance and possible toxicological implications of their high content of uneven fatty acids (UFA). It was confirmed that the extent to which UFA accumulate in adipose tissue of rats fed the 2 products reflects only partially their UFA contents. The presence of UFA in rat tissues does not appear to alter intermediate metabolism. The capacity of liver mitochondria ot oxidize palmitic acid was similar in control and in Liquipron-treated rats. Palmitic acid and heptadecanoic acid did not compete for oxidation when mixed at concentrations which reflect their presence in the tissues of animals fed high levels of Liquipron.
