Characterization of antigen-presenting cells in human apical periodontitis lesions by flow cytometry and immunocytochemistry.

AIM: To analyse phenotypic characteristics of antigen-presenting cells (APC), isolated from human periapical lesions by flow cytometry and immunocytochemistry. METHODOLOGY: Sixteen periapical lesions were digested for 15 min with 0.05% collagenase. Mononuclear cells, separated from other inflammatory cells by density centrifugation, were processed for flow cytometry and/or immunocytochemistry. Single and double immunostainings were performed using monoclonal antibodies specific for human CD45, CD3, CD19, CD14, HLA-DR, CD1a, CD83 and CD123. RESULTS: Antigen-presenting cells (HLA-DR(+) cells) represented 32.9 +/- 17.8% of total mononuclear cells. Amongst them, B cells (HLA-DR(+) CD19(+)) were the predominant APC population, followed by activated macrophages (HLA-DR(+) CD14(+)), dendritic cells (DC) (HLA-DR(+) CD14(-) CD19(-) CD3(-)) and activated T cells (HLA-DR(+) CD3(+)). Based on the predominance of T cells (CD3(+)) or B cells and plasma cells (CD19(+) and CD19(lo), respectively) amongst mononuclear cell infiltrates, lesions were divided into T- and B-types. The percentage of DC in T-type lesions (27.1 +/- 6.8% of total HLA-DR(+) cells) was higher, compared with B-type lesions (10.3 +/- 5.2%) (P < 0.01). Within the DC population, the percentages of CD1a (Langerhans cell type) and CD123 (probably plasmacytoid DC type) did not differ significantly between the groups (P > 0.05). However, the percentage of mature DC (CD83(+)) was significantly higher in T-type periapical lesions (P < 0.05). CONCLUSIONS: Flow cytometry and immunocytochemistry are suitable methods for phenotypic analysis of APC after their isolation from human periapical lesions. APC, that were phenotypically heterogeneous, constituted a significant component of infiltrating cells. Lesions with the predominance of T cells were characterized by a higher proportion of mature DC (HLA-DR(+)CD83(+) cells) than lesions with predominance of B cells/plasma cells.

Ovulation induction and early pregnancy loss in a woman susceptible to autoimmune diseases: a possible interrelationship.

We report a 34-year-old woman with sequentially occurring autoimmune diseases that are possibly triggered by numerous ovulation inductions. At the ages of 26-32 years, she experienced 27 uncontrolled ovulation induction cycles using clomiphene citrate (CC) or CC plus human menopausal gonadotropin plus human chorionic gonadotropin. She became pregnant at the ages of 27, 30 and 31 with subsequent pregnancy loss in the 28th, 8th and 10th week of gestation, respectively. Insulin-dependent diabetes mellitus (IDDM) developed at the age of 28. During the second year of ovulation induction, at the age of 27, she developed arthralgia that worsened and became migratory from the age of 31. Thrombocytopenia appeared at the age of 33. The diagnosis of systemic lupus erythematosus (SLE) and antiphospholipid syndrome (APS) was established at the age of 34. To the best of our knowledge, this is the first case of concurrent IDDM, SLE and APS in a patient associated with ovulation inductions. Excessive levels of estradiol achieved during the ovulation inductions could play a role in the expression of multiple autoimmune diseases in the susceptible woman.

[Prevention of human rabies in Yugoslavia].

Over the last two decades human rabies prophylaxis in Yugoslavia was successful and not a single case of the disease was registered. However, every year there are registered cases of those injured by animals in which rabies was confirmed by laboratory evidence. In FR Yugoslavia every year 800-1000 people are vaccinated against rabies, which makes 10% of all the people injured by animals in Yugoslavia. During 1997 there were 10.266 patients injured by animals; 816 (7.95%) were vaccinated whereas 52 were injured by rabid animals or rabies-suspected animals; 662 were injured by unfamiliar or wild animals, while 102 patients were injured by familiar or preventively vaccinated animals. In other patients vaccination was avoided by a 10-day veterinarian surveillance of the dog. New literature estimated application of reduced so-called "Zagreb-2-1-1" vaccination protocol in severe injuries as unsuccessful in children because of active immune response suppression by simultaneously given immunoglobulin, and that is why this protocol must be abandoned.

[Rhombencephalitis as a manifestation of neuroborreliosis].

Rhombencephalitis is a rare neurological manifestation of stage II of Lyme disease. We presented two cases with no recollection of tick bite nor erythema migrans and with cranial nerve nuclei, pyramidal tract and cerebellar tracts involvement of sudden onset, inflammatory syndrome in cerebrospinal liquor, positive oligoclonal bands and elevated IgG index. A spontaneous remission occurred in both cases, but antibiotic was given intravenously for prevention of relapses and disease progression.

[New technology in maize breeding].

Results obtained by several approaches in the application of Biotechnology in maize breeding are reviewed. RFLP technology in the determination of genetic variation; gene transfer by the use of different methods of gene delivery and the determination of gene integration. Three technologies for foreign gene introduction have been applied; injection of plasmid pRT100 neo into archesporial tissue before micro and macro sporogenesis, slightly modified pollen-tube pathway technology and dry seed incubation in plasmid DNA solution. NPTII gene integration was followed by dot-blot and Southern blot analysis of plant DNA of both T1 and T2 plants. Gene expression was analysed by neomycin phosphotransferase activity. Transformed plants contained the selective NPTII gene sequence in an active form. Bacterial gene integration induced several heritable changes of plant phenotype. As an important change, alteration of the flowering time has been used as a criterion for selection and plant propagation to keep transformed progeny. Besides plant genome transformation, endogenous bacteria living in different maize tissue were found. As a perspective approach for biotechnology application in maize breeding biological vaccine construction has been selected. Therefore, antagonistic effect of gram positive bacterial strains to several pathogenic fungi was investigated. Results obtained after in vivo experiments are discussed.
