Isolation, expression and characterization of a human apolipoprotein B 100-specific cDNA clone.

The isolation and characterization of a human apolipoprotein B 100-specific cDNA clone (lambda gt-B1) containing a 1321 base pairs (bp) spanning insert is described. It encodes the 3'-nontranslated 281 bp long region up to the polyadenylation site and 1040 bp of the C-terminal coding region of 345 amino-acid residues of human apo B 100 and the stop codon. The lambda gt-B1 cDNA clone has been isolated from a human hepatoma cDNA expression library by immunoscreening using affinity-purified polyclonal anti apo B 100 antibodies. The nucleotide sequence of the apo B 100 insert has been determined. A part of the polypeptide sequence derived from this nucleotide sequence was identical with the amino-acid sequence obtained by protein sequencing of a purified cyanogen bromide fragment of apo B 100. The fusion protein consisting of beta-galactosidase and the 345 amino-acid residue long C-terminus of apo B 100 had an apparent molecular mass of 148 kDa in NaDodSO4 polyacrylamide gel electrophoresis. In Northern blot hybridization analysis the insert of the apo B 100-cDNA clone hybridized to a 20 to 22 kb mRNA from adult human liver.

[Psychodynamic aspects of severalsheumatic diseases as revealed in tests].

Three groups of patients with rheumafactor-negative diseases (Palindromic Rheumatism, Reiter's disease, Psoriatic Arthritis) took psychodiagnostic investigations. Tests administrated: MMPI, Rorschach-test, Szondi-test and TAT. The findings of the qualitative psychoanalytic investigations and interpretations of the three groups were compared with the psychological topics of patients with chronic Rheumatoid Arthritis as well as with the findings of the psychoanalytic explorations by Zander. Finally some considerations to the link as well as to the reduction of movement responses in the Rorschach-test are made.

Mineralization (calcification) of coronary arteries.

Mineralogical investigations of calcifications located in coronary vessels were performed on the material obtained from the endarterectomized arteries of 18 patients (15 M, 3 F, aged 36-65) during surgical revascularization procedures consisting in coronary artery bypass grafting. The samples were tested using scanning microscopy, X-ray diffractometry, infrared spectroscopy, atomic absorption spectroscopy, electron microprobe and neutron activation spectroscopy. The results of analyses were calculated with the use of computer programmes. Two types of mineralization were determined: 1. secret mineralization identified as higher than normal content of elements in biological tissues, not demonstrating any mineral grains, and 2. apparent mineralization, appearing micro- and macroscopically as grains composed mainly of hydroxyapatite containing admixture of carbonate groups, i.e. a mineral identical with apatite present in bones, or as calcification of other tissues (heart valves, lungs etc.). The authors suggest that the phenomenon of mineralization should be taken into consideration in the preventive treatment of coronary atheriosclerosis.

[Hemolysis after bioprosthesis implantation in the mitral valve].

Rare case of hemolysis following an implantation of the biological mitral valve is presented. In 1985, biological mitral valve of Ionescu-Shiley type was implanted to a 45-year female patient for post-rheumatic mitral defect. Patient was in the III NYHA class before surgery. Her health status improved after operation. Several echocardiographic examinations have shown normal functioning of the implanted valve (normal gradient across the valve, no retrograde wave). Upper respiratory infection with subsequent progressive weakness, anemia, yellow skin, and heart failure occurred in 1991. Echocardiography has shown significant increase (twice) in the gradient across the valve (max 26, mean--14 mm Hg) and retrograde wave of the II degree as well as mitral insufficiency of the II degree. Laboratory tests have shown marked anemia with reticulosis, increase in blood bilirubin, increase in LDH activity (by about ten times), and a decrease in haptoglobins. Mitral valve dysfunction with hemolysis has been diagnosed. Patient was reoperated. Intraoperatively, incompletely healed valvular ring, and lesions to the leaves such as: fibrosis, thickening and partial deficits. Biological mitral valve was replaced with Corin-Carbotest artificial valve. Postoperative course was uncomplicated.

[Psychodynamics of psoriasis vulgaris in the Rorschach test].

The intention of this report has been to proof the pattern of psychosomatic aspects in Psoriasis vulgaris, as it was developed by Vogel. The method used was the psychoanalytic interpretation of the Rorschach-Test. First this method is described. Important test-results are reported and discussed in comparison with the psychosomatic aspects found by Vogel. There is some coincidence, particularly in sexualisation of intentional and oral desires, and also in the somatisation of the conflict between libidinous and aggressive impulses. Finally an example of test-setting is reported to demonstrate the method of interpretating.
