The bovine major histocompatibility complex (BoLa): close linkage of the genes controlling serologically defined antigens and mixed lymphocyte reactivity.

Detection of linkage between genetic loci in cattle has been hampered by the lack of large full -sib families. A unique source of full-sib families is now available from embryo transplantation. Lymphocytes from six full-sib families, ranging in size from three to seven siblings, were tested for serologically defined BoLA antigens (BoLA-A). In addition, mixed lymphocyte reactivity (MLR) was tested between all paired combinations of cells within each family to distinguish BoLA-D specificities. Serologically identical siblings within each family were reciprocally nonreactive in MLR, and vice versa; thus, no recombinants were detected between the BoLA-A and the BoLA-D loci. Classical genetic linkage analysis revealed that these loci are significantly closer than 11.9 centimorgans.

Hematology and biochemistry reference values for female Holstein cattle.

Reference intervals are presented for 14 hematology and 32 biochemistry variables from four age groups of female Holstein cattle (n = 172) selected randomly from six well managed farms. Each animal was examined by a clinician and with the history available considered to be clinically normal at the time of blood collection. The variable observations were examined for outliers and Gaussian distribution prior to parametric or where necessary, nonparametric analysis. Many differences were noted between age groups but few between farms.

Hematology and biochemistry reference values for the light horse.

Reference hematology and biochemistry intervals are presented for a number of variables of clinical interest determined for blood samples obtained from 60 thoroughbred mares, 12 thoroughbred foals and 50 standardbred horses in training. The observations for each variable were examined for outliers and Gaussian distribution. Parametric analysis was used where the observations were Gaussian initially or after any of four transformations, otherwise nonparametric analysis was required for estimation of the 2.5 and 97.5 percentiles. Description of the sample collection procedures, laboratory methods and statistical analysis are available allowing comparison and judicious application of these reference intervals by interested researchers and clinicians.

Microtechnique for most-probable-number analysis.

A microtechnique based on the most-probable-number (MPN) method has been developed for the enumeration of the ammonium-oxidizing population in soil samples. An MPN table for a research design ([8 by 12] i.e., 12 dilutions, 8 replicates per dilution) is presented. A correlation of 0.68 was found between MPNs determined by the microtechnique and the standard tube technique. Higher MPNs were obtained with the microtechnique with increased accuracy in endpoint determinations being a possible cause. Considerable savings of time, space, equipment, and reagents are observed using this method. The microtechnique described may be adapted to other microbial populations using various types of media and endpoint determinations.

Cyclic AMP metabolism in mouse parotid glands. Properties of adenylate cyclase, protein kinase and phosphodiesterase.

Catecholamines induce unique growth and secretory responses in salivary glands. An analysis of three enzyme activities involved in cyclic AMP metabolism was carried out to identify the specificity of these responses for salivary glands. Although parotid adenylate cyclase has an unusually high specific activity, its kinetic properties and responses to NaF, guanine nucleotides, and isoproterenol are similar to other tissues not stimulated to grow after isoproterenol stimulation. Solubilized adenylate cyclase was separated from other membrane proteins by isoelectric focusing on polyacrylamide gels. There was a single broad peak of activity witha pI of 5.9. Parotid protein kinase has a subcellular distribution and substrate preference similar to hepatic protein kinase. Activation by cyclic AMP is also similar to that reported for other tissues, with a Ka of 1.2 - 10(-7) M. Parotid cyclic AMP and cyclic GMP phosphodiesterases are a heterogeneous group of enzymes with relatively low specific activity as compared with mouse pancreas, liver and brain. Isoelectric focusing of supernatant phosphodiesterases revealed at least sixpeaks of enzyme activity in the pI range of 4-6. Previous reports of a large increase in parotid cyclic AMP levels after in vivo administration of catecholamines and specific growth and secretion could be the result of a relatively high specific activity adenylate cyclase associated with low specific activity cyclic AMP phosphodiesterases.

Observation and documentation of bedside blood transfusion.

Correct administration of blood and the management of the transfused patient are a fundamental element of transfusion safety. Recently published guidelines from the British Committee for Standards in Haematology highlight that most serious transfusion complications occur within the first 15 minutes. They recommend baseline observations before and 15 minutes after commencement of each unit of blood. Using a Royal College of Physicians (RCP) proforma, 100 clinical case notes were examined retrospectively for standards of transfusion documentation and bedside observation. The audit identified that bedside practice was variable and that only 16% of cases met this standard. Most wards continued with hourly observations in uncomplicated cases. Trust guidelines and a staff education programme have been implemented to address the audit results.

Impact of a worksite behavioral skills intervention.

Sixty-four male and female sedentary employees were randomly assigned to an intervention group or control group to determine the effects of behavioral skill training on adoption and maintenance of exercise. Both received a 9-month membership at a local fitness facility. The control group received a 12-week semistructured course, which included a facility orientation and three meetings with a personal trainer. The intervention group received a 12-week behavioral skills course and were encouraged to participate in a 12-week semistructured exercise course followed by a 3-month problem-solving support intervention. Both groups improved their daily energy expenditure, the amount of moderate and vigorous activity they performed, and their strength and flexibility. The study sample was too small to show substantial differences between the intervention and control group. Changes in mediator variables were mixed.

Reasons for pre-hospital delay in reporting chest pain.

This paper looks at why there are still delays between onset of chest pain in myocardial infarction and administration of thrombolytic therapy. It looks at the research into these delays and sets out a number of reasons for them. It also recommends courses of action and further studies to address the problem.
