[Microbiologic studies of in situ removal of iron and manganese].

While undoubtedly a good understanding of soil microbiology in terms of pedology exists, little is presently known about unsaturated subsoils, and aquifers. Yet, in the North German Basin, such underground environments contain to a 90 m depth up to 10(4) g-1 dry wt of heterotrophic bacteria, oligocarbophilic bacteria, denitrifying bacteria, nitrate reducing bacteria, iron precipitating and reducing bacteria, manganese oxidizing and reducing bacteria and other physiological groups of microorganisms. These bacteria are facultative anaerobes. When estimating microscopically, their counts reach up to 10(7) g-1 dry wt. In an oxygen free aquifer which received an oxygenated water for 10 years to remove iron and manganese, the counts of heterotrophic bacteria increased 1-2 orders of magnitude. The counts of nitrifying bacteria also increased whereas those of desulfurizers remained non-affected. In general, the oxidizing bacteria benefited more from the treatment than the reducing bacteria. The soil Eh value was increased, and BOD 5, 10 or 20 values were diminished in comparison to the oxygen free aquifer.

Manganese oxidation by an intracellular protein of a Pseudomonas species.

Cultures of a Pseudomonas sp. strain MnB 1 produce an intracellular, manganese oxidizing protein (abbrev. as Mn ox. protein) during the stationary phase of growth. This protein is heat labile, can be inactivated by protease and has a pH-optimum for manganese oxidation at pH 7.0. Mn2+ is oxidized only at concentrations below 3-10(-5) M. The occurrence of the protein is not dependent on the presence of Mn2+, but is clearly related to the cessation of growth after the end of the exponential growth phase. Oxygen, coenzymes, and low molecular weight components of the cell extract seem not to be involved in the reaction as electron acceptors for the oxidation of Mn2+. Continued manganese oxidation by Mn ox. protein results in a progressive decrease in activity which corresponds to the amount of formed manganese oxide.

Morphology and ultrastructure of Crenothrix polyspora Cohn.

Naturally grown cell material of Crenothrix polyspora from the well of a waterworks was studied by means of phase-contrast and Nomarski interference microscopy as well as by transmission electron microscopy. The material consisted of clusters of sheathed filaments up to 2 cm long. Propagation forms observed were nonmotile, spherical cells that arose by simple ("macrogonidia") or multiple ("microgonidia") septation of the filamental tips. Ultrastructural analysis revealed Crenothrix to be procaryotic and gram negative, with several layers of sheath material surrounding the filaments. On thin sections, individual cells had elaborate membrane systems in the form of lamellar stacks. They resembled thylakoids of photosynthetic bacteria. Spectrophotometric analysis gave no indication of photosynthetic pigments. The cells also contained large hexagonal bodies, rod-shaped fibrillar elements, and polyphosphate granules.

[Determination of the toxicity of industrial waste by means of a bacterial test (author's transl)].

The decision on whether industrial waste should be deposited on general or special dumps depends upon the toxicity of the waste and thus upon the risk of ground-water pollution and the killing off of micro-organisms responsible for breaking down waste products. Since chemical analyses that are to provide information on toxicity are frequently very costly, an attempt was made to determine the toxicity of solid or semi-solid waste with the aid of a bacterial test. As in the perforated-plate test, here, too, the formation and size of inhibition zones were used to determine the toxicity after diffusion of substances contained in industrial waste. About 30 different types of industrial waste were examined with this method. For almost all substances a positive correlation of the (known) constituents of waste to the formation of inhibition zones was found to exist. The influence exercised by solubility and diffusibility and other problems are dealt with.
