A comparison of methods used to enumerate Escherichia coli in conventionally treated sewage sludge.

AIMS: This study examined the suitability of three analytical methods for isolating and enumerating Escherichia coli from conventionally treated sewage sludge. METHODS AND RESULTS: Crude sewage, mesophilic anaerobic digested (MAD) sludge, and final product sludge samples were taken from six sewage treatment works for analysis. Two of the three methods tested were membrane filtration techniques, utilizing chromogenic E. coli/coliform (CEC) media and membrane-lactose glucuronide agar (MLGA); the third method was a most probable number (MPN) technique utilizing Colilert in Quantitray 2000 (Idexx). The methods were evaluated for variation, consistency, false-positive and false-negative results, as well as method correlation. The methods gave good and consistent recovery of E. coli for a range of conventionally treated sewage matrices. All of the methods had a false-positive rate of <3%, although MLGA had a high false-negative rate (35.5%) compared with Colilert (3.81%) and the CEC method (6.75%). This resulted in slightly lower presumptive counts but comparable numbers of confirmed counts. CONCLUSIONS: The three detection methods tested, chromogenic, MLGA and Colilert gave comparable recoveries, and did not vary by greater than one order of magnitude (1 log). SIGNIFICANCE AND IMPACT OF THE STUDY: Forthcoming revisions to the Use of Sludge in Agriculture Regulations (1989) will categorize sewage sludge as untreated, conventionally treated or enhanced treated in accordance to microbiological standards. The standard will be based upon numbers of E. coli removed through the sludge treatment process and the numbers remaining in the final product. It is recommended that the Colilert 2000 (Idexx, Westbrook, Maine) and CEC methods would be equally suitable to assess the reduction of indigenous E. coli in conventionally treated sludges, and that MLGA be used with follow-up confirmatory testing.

BSI workshop on cytokines in reproduction.

The British Society for Immunology Spring Meeting, held at the University of Bristol on April 17-19th, 1996, to mark the bicentenary of Edward Jenner's vaccination against smallpox, provided the setting for the recent meeting of the UK Materno-Fetal Immunobiology Group (MFIG). The them for the MFIG workshop was 'Cytokines in Reproduction' and it was attended by over 60 delegates.

Inactivation of photosensitizing merocyanine dyes by plasma, serum and serum components.

Merocyanine dyes with an oxygen in the electron donor heterocycle were rapidly degraded by plasma, serum and serum components. Replacement of the oxygen by a sulfur or selenium atom rendered the dyes refractory to degradation. The degradation of labile merocyanine dyes was temperature dependent and oxygen independent. The plasma component that was responsible for the degradation of merocyanine dyes was sensitive to heat and detergent, suggesting an enzymatic process. The identification of the structural requirements for sensitivity/resistance to degradation provides the experimenter with a simple means to manipulate the stability of merocyanines in high serum or plasma environments and may expand the clinical utility of merocyanine photosensitizers beyond their traditional role in the extracorporeal purging of bone marrow grafts.

Binding constant determination of WIN 22169, a novel polymeric ligand.

WIN 22169 is a co-polymer containing approximately 11 repeating units of polyoxyethylene and diethylenetriamine pentaacetic acid (DTPA). WIN 66368, a magnetic resonance imaging (MRI) contrast agent, is the gadolinium III complex of WIN 22169. WIN 22169 has been characterized with respect to its equivalent weight, acidity constants and excess acid or base, as well as its metal ion binding constants. The logs of the equilibrium binding constants of the ligand to Gd3+, Ca2+, Zn2+ and Cu2+ were found to be 16.6, 7.47, 12.2 and 14.0. The Gd selectivity constant, a measure of the preferential binding of the ligand toward Gd3+ versus the three in vivo ions: Ca2+, Zn2+ and Cu2+, of WIN 66368 was calculated to be 7.9. This value compares favourably to that for Gd DTPA which has a Gd selectivity constant of 7.04.

Range of mental illness among the elderly in the community. Prevalence in Liverpool using the GMS-AGECAT package.

A sample of 1070 elderly persons aged over 65 living in the Liverpool community was interviewed using the community version of the Geriatric Mental State (GMS) and the findings processed to provide a computerised diagnosis by AGECAT. Levels of organic disorder, probably dementia, reached 5.2%, intermediate between those of London and New York derived from previous studies. Levels for depressive illness overall were below those of other studies at 11.3% while levels for neurotic disorder were much the same at 2.4%. The rise in the prevalence of dementia with age was further confirmed. The GMS AGECAT Package provides a method for standardising both the collection of data and the diagnostic process for comparative epidemiological studies and other research.
