Cultural estimation of yeasts on seaweeds.

Five per cent suspensions of freshly harvested seaweeds were used as an inoculum to develop a selective medium for epiphytic yeasts. Conditions for satisfactory yeast growth and visualization as red colonies on membrane filters were obtained by supplementing a basal glucose-Trypticase-yeast extract-agar at pH 7.0 with 100 mg each of chloramphenicol and 2,3,5-triphenyl tetrazolium chloride per liter. Maximal counts were obtained by triturating the algae in prechilled (4 C) seawater with a blender for 2 to 5 min. Inhibitory phenolic materials released from phaeophytes during this process were removed with a modified Cholodny filtration. A preliminary survey indicated that yeasts were epiphytic on all nine species of seaweeds and that maximal populations occurred on the chlorophytes and rhodophytes especially during the periods of warmer water.

SCE, X-radiation sensitivity and mutation rate in multiple sclerosis.

In order to detect any underlying DNA abnormality that may be present in multiple sclerosis (MS), the incidence of SCE, X-radiation sensitivity and the frequency of 6-thioguanine-resistant cells were determined in the lymphocytes of 34 MS patients. As a group, MS patients showed an increase in the SCE rate compared to control. However, there was no increase in X-radiation sensitivity. The frequency of 6-thioguanine-resistant mutant cells was also normal.

A mucosal apposition technique for ureteroneocystostomy after renal transplantation in cats.

Renal transplantation is a successful treatment for terminal renal failure in cats. However, in the initial clinical study, there was a frequent occurrence of obstruction of the implanted ureter at the bladder wall or stoma. This resulted in the use of a modified "drop-in" technique that had proved effective in the prevention of obstruction in five normal cats. When applied to renal transplant recipients, ureteral obstruction was reduced, but continued to occur. The modified "drop-in" technique was abandoned and replaced with a technique that apposed the cut edge of the ureteral mucosa to the torn edge of the bladder mucosa. This technique is recommended as it has prevented ureteral obstruction in six successive cases. Prevention of ureteral obstruction, and the required corrective surgery, markedly reduces patient morbidity and mortality, length of hospitalization, and expense to the client.

Vocal cord paresis in acute ischemic stroke.

We prospectively determined the frequency of vocal cord paresis (VCP) among first-ever acute ischemic stroke patients. Vocal cords were examined endoscopically within 48 h of stroke onset, at 1 week and 1 month. Of the 54 study patients, 64.8% had lacunar (group 1), 22.2% cortical/large subcortical (group 2), 9.3% lateral medulla (group 3) and 3.7% other brainstem (group 4) infarcts. VCP was found in 11 (20.4%): 11.4% of group 1, 16.4% of group 2, 100% of group 3 and 0% of group 4. VCP was contralateral to the brain lesion in groups 1 and 2, and ipsilateral in 80% of group 3. VCP was strongly correlated with dysphonia (p < 0.0001) and resolved in 2/11 patients after 1 week and in 5/11 after 1 month. Our finding of VCP among acute ischemic stroke patients questions the belief that the nucleus ambiguous is invariably bilaterally innervated by supranuclear centers.

Radiation sensitivity of human malignant lymphocytes.

A simple and rapid in vitro technique to assess the sensitivity of human malignant lymphocytes to roentgen irradiation is described. A variety of established malignant lymphocyte cell lines were cloned in microwells and clone survival was used as the end-point. The survival of the clonogenic malignant lymphocyte down to a fraction of approximately 0.001 could be measured accurately. Except for a T-cell line, the radiation sensitivities of the cell lines were similar to that of normal T-lymphocytes.

Prediction of biological behaviour of human breast cancer using multiple parameters. A preliminary study.

Prediction of early metastases in individual patients has been attempted by combined evaluation of a battery of recognised parameters such as blood vessel invasion (BVI) of tumor cells, Barr body frequency (BBF), plasminogen activator (PA), collagenase, estradiol receptors (ER), progesterone receptors (PgR), and peroxidase activity (PRA) in 18 malignant and 3 benign (control) breast tumors. Since breast tumor cells spread through the blood vessels, the cases were divided into BVI (+) and BVI (-) groups. Results show that in the former group, all the cases had poor prognosis and two even had distant metastases within one year. In BVI (-) group, 9 out of 12 appeared to have good prognosis.

Expression of tumor cell properties in synovial cells in culture.

The tumorigenic properties of human rheumatoid arthritis synovial cells in culture were investigated. The synovial cells developed good colonies and secreted plasminogen activator (PA) and collagenase in the cell cultures, as do Hela cells. Since PA and progesterone receptor (PgR) are considered to be end products of estradiol action in breast cancer cells, the estrogen receptor (ER) and PgR content in these cells was also assayed. Large amounts of ER and PgR were detected in the synovial cells in culture, even though these cells are not targets for sex steroids. Study of the cytomorphologic changes in the synovial cells in culture revealed many characteristics generally observed in neoplastic cells. Whether any or all of these observations have any implication in prognosis or therapy in this disease remains to be studied.

Steroid receptors in breast cancer in Indian women.

The distribution of receptors for estradiol (ER), progesterone (PgR) and androgens (DHTR) in 104 primary and 49 metastatic breast tumours from Indian women is herein being reported for the first time. When compared to reports in the literature, a lower percentage (47%) of ER (+) tumours and a higher percentage (49%) of DHTR (+) tumours were observed. There were more ER (-) tumours in premenopausal as compared to postmenopausal women, and more ER (-) tumours had lymph node metastases at the time of surgery. Both these correlations were statistically significant.
