NMR studies of oligosaccharides derived from hyaluronate: complete assignment of 1H and 13C NMR spectra of aqueous di- and tetra-saccharides, and comparison of chemical shifts for oligosaccharides of increasing degree of polymerisation.

A series of oligosaccharides was prepared from hyaluronate by depolymerisation with bovine testicular hyaluronidase. Complete assignment of the 1H and 13C NMR spectra was obtained for the disaccharide, the tetrasaccharide, and the NaBH4-treated tetrasaccharide, by using various 1D and 2D NMR methods. The 1H assignments for the tetrasaccharide differ from the incomplete data reported recently (ref. 11). The 13C NMR spectra of the aqueous di-, tetra-, hexa-, and octa-saccharides of this series show that all resonances, apart from those subject to obvious end effects, have chemical shifts comparable to those of the corresponding resonances of hyaluronate in D2O. The observed 13C chemical shifts suggests that cooperative intramolecular hydrogen bonds probably play a minor role in determining the conformation of hyaluronate in water.

1H NMR spectroscopic studies of lipid extracts from human fatty liver.

Lipid extracts of biopsy samples from normal and non-alcohol-induced fatty human liver were studied by 1H-NMR at 200 MHz. Spectra of the lipid extracts from 10 mg samples were obtained in 6 min with routine acquisition parameters and allowed the calculation of the phosphatidylcholine to total fatty acyl chain ratio, the cholesterol to total fatty acyl chain ratio, the average fatty acyl chain length, the unsaturation ratio and the acylated glycerol to total fatty acyl chain ratio. The data suggest that lipids with a higher ratio of de novo synthesized fatty acyl chains are stored in non-alcohol-induced fatty liver. NMR lipid analysis appears to be a reliable method for the rapid assessment of hepatic lipid composition on bioptic specimens.

Structural determination of the capsular polysaccharide produced by Klebsiella pneumoniae serotype K40. NMR studies of the oligosaccharide obtained upon depolymerisation of the polysaccharide with a bacteriophage-associated endoglycanase.

The Klebsiella pneumoniae K40 capsular polysaccharide has been isolated and investigated by use of methylation analysis, specific degradations and NMR spectroscopy. The polysaccharide was depolymerised by a bacteriophage-associated endogalactosidase, and the resulting oligosaccharide was characterised by one-dimensional and two-dimensional NMR spectroscopy and direct chemical ionisation MS. The repeating unit of the K40 capsular polysaccharide was shown to be a linear hexasaccharide with the composition-->3)- alpha-L-Rhap-(1-->2)-alpha-L-Rhap-(1-->4)-alpha-D-GlcpA++ +-(1-->2-)- alpha-D-Manp-(1-->2)-alpha-D-Manp-(1-->3)-alpha-D-Galp-(1--> (Rha, rhamnose).

Analysis of lipids in crude extracts by 13C nuclear magnetic resonance.

Phospho- and glycolipids with mono- or polyunsaturated fatty acid chains, free and acylated steroids, carotenes and carotenoids, chlorophylls, and related pheophytins were detected in crude extracts by NMR spectroscopy without any need for prior separation. A broad range of molecules belonging to different lipid classes could be identified in one-dimensional 13C NMR spectra obtained by applying a polarization transfer pulse sequence (DEPT 135 degrees). Directly detected or 1H-detected two-dimensional heterocorrelated NMR experiments were performed to facilitate the assignment of peaks arising from carotenoids, unsaturated fatty acid chains, and chlorophylls. 13C NMR data of crude lipid extracts from the macroalagae Ulva rigida and Fucus virsoides are shown to yield an informative overview of their lipid content. NMR is thus proposed as a simple, nonselective, and nondestructive technique for the first screening of the main lipid classes in complex lipid mixtures.

1H- and 13C-NMR studies of solutions of hyaluronic acid esters and salts in methyl sulfoxide: comparison of hydrogen-bond patterns and conformational behaviour.

The 1H- and 13C-NMR spectra of the ethyl and benzyl esters and the tetrabutylammonium and tetraethylammonium salts of hyaluronic acid [[symbol: see text]2)-beta-D-GcpA+-1----3)-beta-D-GlcpNAc-(1[symbol: see text]n] in Me2SO-d6 have been assigned using 1D and 2D techniques. The chemical shifts of the resonance of GlcNAc C-3 suggest that the relative orientations of the monosaccharides at the (1----3) linkage in the esters and salts are different. Small differences in the chemical shifts of the resonance GlcA C-4 suggest only a slight conformational variation around the (1----4) linkage. The 13C-NMR data also suggest similarities in conformation between the esters in Me2SO-d6 and the salts in water. The chemical shifts of the 1H resonances for NH and OH groups and their temperature dependence for the esters and salts in Me2SO reveal markedly stronger inter-residue hydrogen bonds between the carboxyl and NH groups and between HO-4 of GlcA and O-5 of GlcNAc for the salts. The 3J2,NH values indicate a slightly different orientation for the acetamido group. For solutions in Me2SO, the higher segmental flexibility of the esters is supported by the line widths, whereas the reduced viscosity for the tetrabutylammonium salt showed a sigmoidal concentration dependence and suggests association of chains which could contribute to the segmental rigidity. The linear concentration dependence for the benzyl ester suggests a higher overall flexibility without chain association.

Short-TE projection reconstruction NMR microscopy of trabecular bone.

The aim of this study was to assess the potential of projection reconstruction (PR) NMR microscopy in the quantitative evaluation of trabecular bone architecture. Short-TE PR spin-echo microimages were acquired at 7.05 T on normal bone explants. The main structural parameters such as bone volume fraction (BVF), trabecular thickness (Tb.Th.) and trabecular separation (Tb.Sp.) were obtained from the 3D microimages using the method of directed secants. Quantitative structural data were then compared with those derived from conventional spin-echo microimages. Our study indicates that projection reconstruction NMR microscopy promises to be more accurate than the conventional FTI method in the analysis of trabecular bone.

[Breast feeding in the province of Padua from 1978 to 1993].

INTRODUCTION AND AIMS: The authors report the results of an epidemiological study on breastfeeding from 1978 to 1993, followed by the same operators using the same method in the same territory, a semi-rural area of the province of Padua. In particular, they report the results for 1993 and compare them with those of earlier years and with those in Selvazzano, a town lying on the outskirts of the city of Padua. METHODS: A total of 963 of the 1115 women who had given birth during the study period were interviewed (85.6%). RESULTS: The frequency of breastfeeding rose from 61% in 1978 to 81% in 1993 and its mean duration from 4 to 6.3 months. The frequency of breastfeeding in Selvazzano was slightly lower (72%) as was the mean duration (5.8 months). The factor which most strongly influenced breastfeeding was the mother's work: its frequency was 66.1% for factory workers, 89.9% for housewives, 79.2% for office workers and teachers, 85.7% for shopkeepers-craftworkers and 75% for managerial staff. From the overall data it was observed that the frequency of breastfeeding increased in parallel to the length of time off work after birth: it rose from 69% among working women taking 3 months off work to 86% in housewives and those taking > or = 13 months off work. The level of education had less of an effect on the decision to breastfeed: the frequency of breastfeeding in mothers with high-school diplomas or university degrees was 85% compared to 79% in those with a lower level of education. According to this study the age of the mother did not influence the frequency of breastfeeding. CONCLUSIONS: The authors emphasise the need to carry out concrete new initiatives in favour of working mothers.
