Monoclonal antibodies to Coxiella burnetii that cross-react with strain Nine Mile.

Results are presented to show the binding properties of five monoclonal antibodies directed to Coxiella burnetii Priscilla with cross-reactions to the Nine Mile strain. The monoclonal antibodies preferentially recognize phase I epitopes by ELISA and recognize phase II epitopes by immunoblotting but do not allow differentiation between so-called chronic and acute strains of C. burnetii. The only difference in reactivity was in the staining pattern revealed after reactions with lipopolysaccharide I antigens.

Evaluation of selected parameters of a metabolic profile and levels of cadmium in reproductive organs of rabbits after an experimental administration.

Selected serum parameters of metabolic profile and distribution of cadmium in reproductive organs in rabbits were studied after an acute and subchronic administration. After an i.p. application of cadmium (1.5 mg.kg-1) a significant increase of glucose (5.90 mmol.l-1) and cholesterol (2.22 mmol.l-1) was observed. There were no significant differences in these parameters after the subchronic (5 month) p.o. application. The concentration of AST and ALT significantly decreased after a subchronic p.o. administration (1.0 mg.kg-1) but not after an acute i.p. application. There were no significant differences in the level of total proteins and total lipids. The highest accumulation in reproductive organs (testis, ovary, uterus) was observed after i.p. administration. The level in the testis was 40 times higher than in the control group (0.04 mg.kg-1; 1.93 mg.kg-1), 174 times higher in the ovary (0.03; 5.21) and 65 times higher in the uterus (0.04; 2.59). After a subchronic p.o. application, the concentration was only 2.5 times higher in the testis (0.04; 0.10), 16 times higher in the ovary and 6 times higher in the uterus than in the control group.

Immunogenicity of Coxiella burnetii whole cells and their outer membrane components.

The immunogenicity and protective efficacy of the phase I and phase II Coxiella burnetii whole cells (Cb I and Cb II) and their outer membrane components (OMC), i.e. phase I trichloroacetic acid extract (TCAE), phase I 29 K protein (PRO), phase I and II lipopolysaccharides (LPS I, LPS II), polysaccharides (PS I, PS II), and lipid A (LA I, LA II), were compared. The highest immune response was observed in BALB/c mice by Cb I in both humoral immunity and lymphocyte transformation assays, and in the protective effect as well. The immune response was also significant by Cb II, but their protective capacity was low. The OMC reacted variously. Only TCAE and PRO gave a high value of humoral immunity evaluated by the serological methods. All OMC reacted in the haemolytic plaque assay giving different responses. Lymphoproliferation of splenocytes was positive with all OMC using both Cb I and Cb II antigens with the exception of PS I and PS II in the case of Cb II antigen. The induction of protection against infectious Cb I was demonstrated after immunization with TCAE, PRO, and LPS I. Other OMC did not induce protection against this agent.

Comparison of various methods of lipopolysaccharide isolation from Coxiella burnetii strain Priscilla in the virulent phase I.

Four methods of isolation of a lipopolysaccharide (LPS) from Coxiella burnetii strain Priscilla in virulent phase I have been examined. Each isolation method afforded a specific LPS portion of the polydisperse LPS system present in the outer membrane of C. burnetii cell. The hot phenol/water method was found the most efficient with respect to yield and chemical composition of the smooth (S) LPS that was shown to prevail in the outer membrane LPS macromolecules of C. burnetii.

Analysis of the 3-deoxy-D-manno-2-octulosonic acid region in a lipopolysaccharide isolated from Coxiella burnetii strain Nine Mile in phase II.

Structural analysis of the 3-deoxy-D-manno-2-octulosonic acid (Kdo) region in a lipopolysaccharide (LPS) II isolated from Coxiella burnetii strain Nine Mile in avirulent phase II revealed the presence of three variously linked Kdo residues. The lipid A proximal Kdo is substituted at C-4 by a Kdo-(2-->4)-Kdo disaccharide and this structural arrangement of the Kdo residues is similar to that of enterobacterial LPSs.

The complex formation of influenza virus envelope glycoproteins with outer membrane proteins of Neisseria meningitidis or Borrelia burgdorferi.

The isolation of influenza virus envelope glycoproteins was achieved by one-step procedure consisting of treatment of purified virus with zwitterionic detergent and separation of viral constituents by sucrose density gradient centrifugation. Viral glycoproteins and proteins of outer membrane of N. meningitidis or B. burgdorferi formed complexes after removal of the detergent by dialysis. Complexing of viral glycoproteins and bacterial proteins was monitored by gel chromatography on Sepharose 6B, polyacrylamide gel electrophoresis and electron microscopy. It was demonstrated by immunoblot analysis, that virus-spirochete complexes elicited formation of antibodies in mice directed against osp A and osp B of spirochete, as well as against viral glycoproteins, respectively.

Improved procedure for the drying and storage of polyacrylamide slab gels.

An improved procedure for the drying and storage of polyacrylamide (PAA) slab gels of various sizes and acrylamide contents is presented. A PAA slab gel is preferentially fixed in methanol-acetic acid-glycerol-water (1.0:1.0:0.1:2.0) and then dried under low pressure in an adapted photoprint dryer between Cellophane and polyethylene foils at 75-80 degrees C for 50-90 min. Strong, transparent and flexible sheets are obtained that can be stored for extended periods. The potential of the procedure is exemplified following electrophoresis of both lipopolysaccharides and protein isolated from Coxiella burnetii bacterium.

Evidence for the structural heterogeneity of the polysaccharide component of Coxiella burnetii strain Nine Mile lipopolysaccharide.

Highly purified lipopolysaccharide (LPS) preparation obtained from Coxiella burnetii strain Nine Mile in phase I was used to determine the structure and monosaccharide composition of the polysaccharide component. The procedure included sodium dodecyl sulphate-polyacrylamide gel electrophoresis followed by silver staining and gel chromatographic fractionation of acetic acid-hydrolyzed LPS. Five fractions (A-E) were analysed by GLC-mass spectrometry. D-Mannose and D-glycero-D-mannoheptose were present in an appreciable amount in all polysaccharide fractions (A-D), whereas the virenose and dihydrohydroxystreptose contents varied. The highest content of both rhamnose and ribose was found in the low-molecular weight polysaccharide fraction D. The former sugar is being reported for the first time to be a LPS constituent. D-Xylose and D-glucose content varied considerably in the individual fractions and was the highest in fraction A. Glucosamine and galactosaminuronic acid were present in all polysaccharide fractions and, surprisingly, L-glycero-D-mannoheptose was also found, but its presence was limited within the certain degree of polymerisation of the polysaccharide chains. Mild acid hydrolysis of LPS resulted in a partial release of dihydrohydroxystreptose and virenose residues, which were collected and identified in fraction E. The data presented indicate a strong microheterogenity within the individual polysaccharide chains with respect to their sugar composition, size, and shape. Thus, the chemical structure of Coxiella LPS appears to represent a significant departure from the structures described for enteric LPSs.

[Campylobacter pylori and treatment of duodenal ulcer].

The authors subjected to endoscopic examination 90 patients with duodenal ulcers. From each patient they collected a bioptic specimen of the gastric antrum and duodenal bulbus for microbiological and histological examination. They found that for detection of C. pylori the microscopic examination was most useful. The presence of C. pylori in the gastric antrum was detected in 87% of the subjects and in the duodenal bulb only in 48%. The difference is significant at the 0.01 level. To 43 patients of the group Ranitidine was administered. The ulcer was cured within 4 weeks in 35 subjects. Ranitidine did not affect the presence of C. pylori and antral gastritis. After cure of the ulcer the authors administered Tarivid to 10 patients with persisting C. pylori positivity. The presence of C. pylori and antral gastritis were not affected by this treatment.

[A prospective prognostic study of liver cirrhosis].

The authors conducted a prospective, longitudinal, multicentre study in patients with liver cirrhosis; a total of 192 patients were on follow-up, of this number 125 died during the study. Mean age, 56 years, was markedly more advanced in the dead; men prevailed among the patients. Duration of complaints and the interval since the diagnosis had been established was shorter in the dead than in those alive; a positive HBs antigen was likewise more often present in the former. Clinical and laboratory parameters in this group were assessed with a view to the differences between the dead and those alive and with a view to the time course of changes in these differences. Multifactorial discriminative analysis of these data for various intervals before death was also performed. The most pronounced differences were noted in clinical manifestations and laboratory signs of hepatic insufficiency, especially in the presence and degree of ascites, jaundice, encephalopathy, and in the values of bilirubin, Quick' test and blood albumin levels; thus the degree of liver insufficiency appears to be the most important prognostic marker in patients with liver cirrhosis. Moreover, a significant increase in urea and leucocyte levels was found in the terminal stages of the disease. Ascites, encephalopathy and a progressive increase in serum bilirubin levels seem to be the best markers for discriminative analysis of prognosis for the last six months of life. Results of this study, while confirming some findings by other authors, offer also new insights into the whole issue.

Soluble chromogenic substrates for the assay of endo-1,4-beta-xylanases and endo-1,4-beta-glucanases.

New soluble chromogenic substrates were prepared for specific and rapid assays of endo-1,4-beta-xylanases and endo-1,4-beta-glucanases. A soluble beechwood 4-O-methyl-D-glucurono-D-xylan was dyed with Remazol brilliant blue R, and hydroxyethylcellulose was coupled to Ostazin brilliant red H-3B. The assays are based on photometric measurements of the enzyme-released dyed fragments soluble in the presence of organic solvents which precipitate the original substrates and their high-molecular-weight fractions. The assays are advantageous for rapid analyses of large amount of samples and also permit evaluation of the activities of both enzymes in the presence of exo-beta-glycanases and beta-glycosidases, at a high level of reducing compounds and viable cells, on the cell surface and on cell membranes and organelles.
