Prostaglandin release by aliphatic alcohols from the rat isolated lung.

1. A comparative study has been made of the abilities of members of a homologous series of aliphatic alcohols to release prostaglandins from the rat isolated perfused lung. 2. Infusions of low concentrations (0.002 mmol--0.2 mmol) of methyl, ethyl, n-propyl, i-propyl, n-butyl and t-butyl alcohol for periods of up to 5 min was accompanied by the continuous output of prostaglandins into the venous effluent, as measured by biological assay. Maximal release occurred with methyl and ethyl alcohols, the releasing abilities of the rest being in the order of n-propyl> i-propyl> n-butyl> >t-butyl. Radiomimmunoassay revealed that both PGE2 and PGF2 alpha were released by ethyl alcohol with the former being in up to 10 times greater quantities than the latter. However, the amounts of these prostaglandins failed to account for the total release of active substances as measured by bioassay. 3. Examination of concentration-response relationship showed that as the concentration of methyl, ethyl and n-propyl alcohol was increased prostaglandin output also increased, but that after reaching a maximum further increase in alcohol concentration resulted in diminished release. In contrast, the lowest concentration of i-propyl, n-butyl and t-butyl alcohol utilized (0.002 mmol) caused highest prostaglandin output and release declined as the concentration was increased. Release by ethyl alcohol (0.02 mmol) was inhibited by t-butyl alcohol (0.2 mmol). 4. Thus aliphatic alcohols can not only release prostaglandins but also depress release. Both properties appear to be possessed by each member of the series, their relative prominence varying in each compound. Release is the predominant action of the low molecular weight, short chain compounds whereas depression of release is the most prominent property of those of higher molecular weight, particularly the secondary and tertiary alcohols. 5. It is suggested that release occurs in response to low alcohol concentrations altering the nature of the water-cell membrane interface, thus producing changes in membrane protein configuration and activation of phospholipase A. Inhibition of release is probably a consequence of non-specific depression of cell function.

Ovarian and peripheral venous blood prostaglandin F2alpha levels in pregnant and nonpregnant women.

Prostaglandin F2alpha (PGF2alpha) was measured by radioimmunoassay in human blood plasma collected from both ovarian veins and from a peripheral vein in eight pregnant and seven nonpregnant women. In the pregnant women, significantly higher levels of PGF2alpha were found in venous blood plasma taken from the ovary which contained the corpus luteum than were found in either the ovarian vein blood plasma from the opposite ovary or in the blood plasma from a peripheral vein. Thus, PGF2alpha levels in the venous blood plasma from the ovary without a corpus luteum were not significantly different from those found in the peripheral venous blood plasma. In seven nonpregnant women, significantly higher levels of PGF2alpha were found in the venous blood plasma taken from the ovary which contained a corpus luteum than were found in the blood plasma taken from a peripheral vein. The former levels did not differ significantly from those found in the venous blood plasma taken from the ovary without a corpus luteum in these nonpregnant women. Thus, in nonpregnant women a significant difference was found between PGF2alpha levels in the venous blood plasma from the ovary without a corpus luteum and those in peripheral venous blood plasma.

Sperm wash in three culture media: maximization of motile sperm recovery during swim-up incubation.

Three different culture media commonly used during in vitro gamete manipulations were studied for their efficacy in sperm wash procedure. Highest numbers of motile sperm were recovered at 6 hours following incubation in WT-6 and Ham's F-10 media. However, WT-6 yielded higher motile sperm numbers than Ham's F-10. Swim-up sperm number reached a peak at 3 hours following incubation in BWW. A period of 2 to 6 hours of incubation of sperm pellets overlayed with sperm wash media resulted in highly enriched motile sperm fractions free of dead spermatozoa and seminal debri.

Prostaglandin F2 alpha and E2 release by peritoneum with and without endometriosis.

Peritoneal endometriotic implants and adjacent normal peritoneum from five patients were analyzed for prostaglandin (PG) release. Each tissue biopsy was incubated using medium 199 in triplicate at 37 degrees C for six hours, and PGF2 alpha and PGE2 concentrations were measured in the incubation medium every two hours. This study demonstrates that peritoneum involved with endometriosis releases significantly more PGF2 alpha and PGE2 (P less than .05) than adjacent normal peritoneum, and suggests that peritoneal endometriotic implants may be a source of the elevated peritoneal fluid PG levels previously reported in patients with endometriosis.
