Rat corticosteroid-binding globulin (CBG) biosynthesis by fetal hepatocytes in culture.

The ability of fetal liver to synthesize and secrete corticosteroid-binding globulin (CBG) was investigated using primary cultures of hepatocytes transplanted from 15- and 18-day-old rat fetuses. Analysis of culture medium, after labelling with [14C]leucine, by crossed immunoelectrophoresis, clearly demonstrated CBG, albumin and alpha-fetoprotein (AFP) secretion. The relative rate of CBG secretion was more important at the younger stage. Indirect immunofluorescence permitted localization of CBG in fetal hepatocytes. The results suggest that the high level of CBG found in fetal serum is mainly produced by the liver of the fetus itself.

In Belgium, viral hepatitis A is predominantly childhood disease.

Data from medical histories and seroepidemiological surveys of blood donors offer good grounds for regarding viral hepatitis A in Belgium primarily as a childhood disease. The epidemiology of anti-HA has thoroughly altered over the last few decades, which correlates with radical socioeconomic changes and improved hygienic living conditions. The rate of HLA-antigens shows in general no different patterns in groups with or without a recognized history of HAV infections.

Immunochemical characterization and quantitation of human sex steroid binding plasma protein.

The interest in the measurement of human sex steroid binding plasma protein (h-SBP) is now increasing since it allows the estimation of the free fraction of circulating hormones in plasma. Up to this date, this protein could only be determined by measuring the total binding capacity of serum for dihydrotestosterone (DHT). The purpose of the present work was to purify the protein, to prepare a rabbit monospecific antiserum and to develop an immunoelectrophoretic assay of h-SBP. The immunological assay is specific, accurate and sensitive. A good correlation with the radioligand assay was found. The h-SBP levels obtained by immunoelectrophoretic assay of different serum samples were 5.3 +/- 1.4 (SEM) mg/L in normal men and 13.4 +/- 2.6 (SEM) mg/L in normal women.

Quantitative determination of the HBsAg.

HBsAg concentrations down to 0.05 mg/ml can be measured by a quantitative immunoelectrophoresis. By a quantitative radioimmunoassay, HBsAg concentrations from 0.001 mg to 1 mg/ml can be measured.

The prevalence of ay and ad subtypes of the hepatitis B surface antigen in Belgium.

Subtyping in various groups of persons positive for hepatitis B surface antigen gave the following results: Among asymptomatic blood donors (healthy carriers) subtype ad was found in 20, subtype ay in 24. Among symptomatic patients with acute hepatitis B ad was found in 25, ay in 29. Among patients of an institution for the mentally retarded ad was found in 9, ay in 23. Among patients of a Among patients of a geriatric institution ay was found in 9; among medical personnel of the same geriatric institution ay was found in 3 persons. Among symphilis patients ad was found in 6, ay in 29; and in patients of a dialysis centre ay was found in 6. These data show that neither subtype is really dominant in Belgium.

Inhibition of lipoprotein lipase activity by a monoclonal immunoglobulin in autoimmune hyperlipidemia.

Autoimmune hyperlipidemia (AIH) may be induced a variety of antibodies which inhibit different stages of the lipolytic process by which the lipid load is removed from the circulating lipoproteins. In a patient having a monoclonal gammopathy and a nephrotic syndrome with a glomerulonephritis and a marked hypertriglyceridemia, it was found previously that the monoclonal IgG gamma Lac. reacted with human VLDL as well as with human serum albumin. Here it is demonstrated that the purified IgG gamma inhibits the lipolysis of triglyceride substrates by reacting with a substance (Lac. S) necessary for lipoprotein lipase activity. The interaction of IgG lambda Lac. with serum or HDL-activated triglyceride substrates inhibits the lipolytic activity of human and rat plasma post heparin and also adipose tissue lipases. It slightly inhibits the activity of swine pancreatic lipases. The Lac S. which reacts with IgG Lac. is associated to whole and delipidated VLDL and HDL and not to LDL or purified APo-A. It may be an Apo-C or a non-peptidic co-factor of the lipases which remains bound to the apoprotein core after delipidation. Its lack of species specificity and its presence as traces in HSA preparations favors the latter hypothesis. The Lac. substances is different from the Pg and As substances which were found to react with IgA anti-Pg and IgG anti-As antibodies in previously reported antilipoprotein AIH.

[Antiheparin immunoglobulin in malignant lymphoma with hypertriglyceridemia, in the hamster].

A massive hypertriglyceridemia associated with low post heparin lipolytic activity, was demonstrated during the growth of a transplanted lymphoma in Hamsters. An immunoglobulin, with high anti-heparin activity, was extracted from the tumor. It could inhibit both the anti-thrombin and prolipase activity of heparin. The role of this anti-heparin immunoglobulin in triglyceride metabolism allows us to consider the hypertriglyceridemia of lymphoma-bearing Hamsters as one of the two previously described types of auto-immune hyperlipidemia.

[Analysis of the hemocyte proteins from Astacus leptodactylus].

Three soluble proteins exist in the haemocytes of Astacus leptodactylus in an appreciable amount. The first one is "fibrinogen" apparently in its monomer state, which is at the origin of the clottable plasmatic protein. The second is assumed to be a slow hemocyanin and the third to be a molt fraction.

[Immunologic analysis of the variations of hemolymph proteins of Astacus leptodactylus during the premolt].

The variations in the hemolymph proteins of crayfish were studied by quantitative protein determination, elctrophoresis on polyacrylamide gel gradient and tandem crossed immuno-electrophoresis, from stage C 4 to the end of stage D. The total protein concentration varies during the premolt, with two peaks in D 0 and D I'''-D2, but the plasmatic "fibrinogen" rate is about the same during this period. But three fractions exist which show important changes during the molt cycle.

Coagulation in the crayfish, Astacus leptodactylus: attempts to identify a fibrinogen-like factor in the hemolymph.

1. A series of tests were conducted to determine whether or not the hemolymph of the crayfish Astacus leptodactylus contains a plasma coagulation factor. 2. The total protein amount is higher in the plasma than in the serum. 3. Serum and plasma do not exhibit similar electrophoretic banding patterns. Plasma contains one band or a series of supplementary fractions with a high molecular weight. 4. Electrophoregrams of plasmatic clottable extract, obtained by the classical methods employed in crustacean serology, show a main fraction or a series of polymers with the same electrophoretic behavior as the additional fractions seen on the plasma pattern. 5. This solution clots when treated with CaCl2 and a cellular extract. 6. Immunoelectrophoresis demonstrates the presence of a clottable protein precipitate line in plasma, but this protein also gives a very faint similar line in serum.

[The effect of freezing on the electrophoretic separation of hemolymph proteins of several arthropods].

Electrophoresis in 6% or gradient polyacrylamide gel were realised with fresh and stored hemolymphs. The experiments state that there are variations in relation to storage duration, in the separation pattern of hemocyanin fractions in Astacus leptodactylus, Palinurus vulgaris and Carcinus mediterraneus. Moreover, important changements in the supplementary factor of crayfish plasma are revealed.
