Diflunisal. A new oral analgesic with an unusually long duration of action.

The analgesic efficacy of single 500- and 1,000-mg doses of diflunisal (Dolobid), a new nonsteroidal anti-inflammatory analgesic, was compared in a double-blind study with that of acetaminophen, 600 mg, the combination of acetaminophen, 600 mg, with codeine phosphate, 60 mg, and placebo in 159 oral surgery outpatients. Using a self-rating record, patients rated their pain and its relief hourly for 12 hours after medication. Both doses of diflunisal were significantly more effective than acetaminophen alone and produced peak analgesia comparable to that of the acetaminophen-codeine combination. Diflunisal proved to have an unusually long duration of analgesic action. Acetaminophen and the combination were significantly superior to placebo through hours 2 and 5, respectively; both doses of diflunisal were significantly superior through the end of the 12-hour observation period. None of the active treatments produced more side effects than the placebo.

Influence of androgens on inositol secretion and sperm transport in the epididymis of rats.

The effects of unilateral efferent duct ligation, bilateral castration or testosterone replacement on the spermatocrit and inositol concentrations in luminal fluid removed by micropuncture from the rat epididymis were investigated. By 5 days after efferent duct ligation, there were substantial falls in the spermatocrit in the caput and corpus, but no change in the cauda. After 7 days there was an increase in the spermatocrit in the cauda. After castration there were more marked reductions in the spermatocrit in the caput and corpus but there was no change in the cauda. The spermatocrit in the corpus, but not in the caput, remained normal when castrated animals were treated with testosterone. The inositol concentrations in luminal fluid from all three sites were not changed after ligation of the efferent ducts but were greatly reduced after castration; this fall was prevented by testosterone treatment. The results support the suggestion that inositol is secreted in the rat epididymis and also indicate that the secretory process is controlled by androgens.

An evaluation of the analgesic efficacy of proquazone and aspirin in postoperative dental pain.

The analgesic efficacy of 75, 150, and 300 mg proquazone, a new nonsteroidal antiinflammatory agents, was compared to that of 650 mg aspirin and a placebo in outpatients who had moderate or severe pain following the surgical removal of impacted third molars. Estimates of the relative potency of proquazone to aspirin ranged from 4.9 to 6.2 for total analgesic effect and from 7.7 to 8.4 for peak analgesic effect. Each dosage level of proquazone and aspirin provided significant analgesia compared to placebo and was well tolerated. Adverse effects were transitory and did not appear to be dose related for proquazone.

Concentrations of myo-inositol in the luminal fluid of the mammalian testis and epididymis.

The concentration of free myo-inositol was measured in the luminal fluid removed by micropuncture from the testis and epididymis of rat, hamster, rabbit, rhesus monkey, baboon, ram and boar. The highest concentration (mM) was found in the epididymal luminal fluid from the cauda epididymidis of the hamster (90) and the rat (50), and the lowest was seen in the rabbit (< 0.5), and boar (1.0-1.2), whilst the rhesus monkey (17), the baboon (1.3) and the ram (0.9-5.9) were intermediate. The species also differed with respect to the changes in luminal inositol coicentration along the epididymis.

Control of the yeast Torulopsis glabrata in the stomachs of glucose fed lambs by oral dosing with nystatin and amphotericin "B".

In the absence of specific dietary lipids, large amounts of ethanol (ethyl alcohol) are produced from glucose in the stomachs of ruminant neonates by the resident yeast Torulopsis glabrata. Using new born lambs, oral dosing with the antifungal antibiotics Nystatin and Amphotericin "B" was shown in samples of stomach contents, to control the ethanol production entirely, and to suppress the yeast numbers partially but not permanently. Of 11 lambs receiving heavy and continous doses orally of the pure antibiotic powders, 7 died, of these 5 had developed adhesive peritonitis.

The hydrogenation of unsaturated fatty acids by five bacterial isolates from the sheep rumen, including a new species.

Five strictly anaerobic bacteria able to hydrogenate unsaturated fatty acids were isolated from sheep rumen. One was characterized as Ruminococcus albus, two as Eubacterium spp. and two as Fusocillus spp., one of which is named as a new species. The Fusocillus organisms were able to hydrogenate oleic acid and linoleic acid to stearic acid, and linolenic acid to cis-octadec-15-enoic acid. The R. albus and the two Eubacteria did not hydrogenate oleic acid but converted linoleic and linolenic acids to a mixture of octadecenoic acids; trans-octadec-II-enoic acid predominated but several isomeric cis and trans octadecenoic acids were produced together with isomers of non-conjugated octadecadienoic acids. The intermediate and final products of hydrogenation by each organism were compatible with the results from mixed rumen bacteria.
