Dopamine-glutamate interaction in rat striatal slices: changes of CCDPK II, PKA, and LDH activity by receptor-mediated mechanisms.

AIM: To study the effects of dopamine (DA) and glutamate (Glu) and their receptor agonists/antagonists on Ca2+/calmodulin-dependent protein kinase II (CCDPK II), cyclic AMP-dependent protein kinase A (PKA) activities and the LDH release in rat striatal slices, and to examine the interaction between DA and Glu transmitter systems in striatum. METHODS: The activities of CCDPK II, PKA, and the release of LDH were determined with the 32P-incorporation and colorimetry respectively in rat striatal slices. RESULTS: (1) Exogenous DA, D1 receptor agonist SKF 38393 and D2 receptor agonist LY 171555 reduced CCDPK II activity in striatal slices; Glu also inhibited CCPPK II activity in a concentration-dependent manner. NMDA receptor antagonist MK-801 could antagonize the inhibitory effect of SKF 38393 and LY 171555 on the CCDPK II activity. D1 and D2 receptor antagonists SCH 23390 and spiperone could also antagonize the decrease of CCDPK II activity induced by Glu; (2) DA and SKF 38393 markedly increased PKA activity in striatal slices, which was reduced by MK-801; (3) DA and Glu increased the release of LDH from the striatal neurons in a concentration-dependent manner. MK-801 antagonized the increase of LDH induced by DA. Spiperone, rather than SCH 23390, could reduce the release of LDH from striatal neuron in the presence of Glu. CONCLUSION: The interaction between DA and Glu transmitter systems is found in the regulation of the CCDPK II and PKA activities and cell function in the striatum.

Gene transfer of vascular endothelial growth factor plasmid/liposome complexes in glioma cells in vitro: the implication for the treatment of cerebral ischemic diseases.

Vascular endothelial growth factor (VEGF) is most promising in therapeutic angiogenesis for ischemic vascular disease. This paper aimed to study VEGF gene therapy for the treatment of cerebral ischemia. The glial cell was chosen as the target cell for gene transfer, and the expression of VEGF was studied in vitro. VEGF plasmid/liposome complexes were constructed by mixing VEGF plasmid with liposome, and then cultured C6 glioma cells were transfected with these complexes by lipofectamine method. As control, the same kind of cells were exposed to liposome only. Immunohistochemistry was performed to both groups at 24, 48 and 72 hours after transfection. The transfected cells expressed VEGF significantly higher than the control. The present result demonstrated the feasibility of choosing the glial cell as the target cell for VEGF gene transfer, and found the rationale for the cerebral VEGF gene therapy.

[Effect of rice variety resistance on population dynamics of Nilaparvata lugens and Sogatella furcifera].

The effect of rice varieties with different insect-resistance on dynamics of Nilaparvata lugens and Sogatella furcifera population was examined, based on the investigation of their inhabiting ratio, egg amount, survival rate of egg and nymph, nymph development, and the amount of their offspring population. Four rice varieties, TN1, N22, AsD7 and Ptb33 were investigated. Ptb33 had a significantly lower inhabiting ratio, egg amount, and survival ratio of eggs and nymphs of both parasitic populations, a prolonged development time from nymphs to adults, and an inhibited offspring population, indicating an evident resistance of this host variety. N22 and AsD7 inhibited the development of offspring S. furcifera and N. lugens, respectively. The parasitic populations of S. furcifera and N. lugens developed more rapidly on TN1, compared with N. lugens population on N22 and S. furcifera on AsD7. The relationship between the quantitative dynamics of both population and rice variety replacement was discussed, and the possibility of using resistant rice varieties to control these two insect populations was also analyzed.

[Effect of grazing on reproduction in Leymus chinensis population].

The experiment on the effect of grazing on the asexual and sexual reproduction and biomass allocation in Leymus chinensis population was conducted in Songnen grassland. The results show that animal grazing reduced both asexual and sexual reproductive channels. With the increase of grazing intensity, the densities of asexual shoots and rhizome tillers, seed biomass, numbers of seeds per inflorescence and sexual shoot ratio decreased significantly, especially after heavy grazing. The rates of biomass allocation to rhizome increased significantly with increasing grazing intensity, while the rates of biomass allocation to assimilation, unassimilation organs and seed production decreased remarkably. In the haviest grazing stage, there were almost no sexual shoots.

[Environment capacity of eco-tourism resort].

The results of quantitative analysis on the amount of tourist, service-environment capacity, eco-environment capacity, and their relations in Five-finger Mountain eco-tourism resort indicate that the amount of tourist in common situation and in its extreme was 1918 and 2301 visitor-hour per day, and the service-environment capacity and eco-environment capacity were 6000 and 2400 visitor-hour per day, respectively. The eco-environment capacity was smaller than its service-environment capacity, and would become the first limiting factor to the increase of tourist amount, which was mainly due to the ecological fragility of resort, the lower resistance of biological communities to the disturbance, and the slower speed of ecosystem restoration after its being destroyed.

[Energy allocation to growth and reproduction in Leymus chinensis population].

Studies on energy allocation to growth and reproduction in Leymus chinensis population show that the average calorific value of modulus was decreased in the order of ear > leaf sheath of vegetative shoot > leaf sheath of sexual shoot > leaf of vegetative shoot > stem of vegetative shoot > stem of sexual shoot > leaf of sexual shoot > litter, and the energy allocation rate of vegetative and reproductive growth was in the order of vegetative shoot > rhizome > litter > sexual shoot. The dynamics of energy allocation rate of modulus was different with seasons. Among all reproductive stem modulus, the rate of energy allocation to stem and leaf sheath of sexual shoot was higher than that to ear and leaves.

An experimental study of endovascular brachytherapy using liquid 32P filled balloon catheter.

OBJECTIVE: To observe the effect of endovascular brachytherapy on the prevention of restenosis after interveneional therapy and to investigate its possible mechanisms. METHODS: In the balloon-injuried rabbit model, pathological sections of iliac arteries were observed and the changes of vascular histomorphology were estimated by computer analysis of photomicrograms. Using immunohistochemical techniques, proliferating cell nuclear antigen (PCNA) was quantified to assess the proliferation of vascular smooth muscle cells. RESULTS: After rabbit iliac arteries were injured by balloon overstretch angioplasty, neointima were shown to be less proliferative in the irradiated group than in the control group, from PCNA scores. The formation of neointima was suppressed with the external elastic laminar area increasing and without the luminal area decreasing in the irradiation group. CONCLUSION: Endovascular brachytherapy using a liquid 32P filled balloon catheter system could prevent restenosis possibly by inhibiting the formation of neointima and improving positive vascular remodeling.

[Aberrant expression of MUC2 and MUC3 genes in gastric carcinoma and its significance].

OBJECTIVES: To explore the clinicopathological significance of the expression of MUC2 (mucin 2) and MUC3 (mucin 3) genes in tissues of the normal gastric mucosa, intestinal metaplasia (IM) and gastric carcinoma. METHODS: MUC2 and MUC3 apomucins were detected by immunohistochemistry; MUC2 and MUC3 mRNAs were detected by in situ hybridization. RESULTS: In the normal stomach, antibody detecting MUC2 apomucin and oligonucleotide probe detecting MUC2 mRNA were not reactive with mucus-producing cells in the superficial epithelium and neck, which were weakly reactive with antibody and probe detecting MUC3. In the duodenum, MUC2 apomucin and mRNA were found at the peri- and supranuclear area of goblet cells, but MUC3 apomucin and mRNA were at the cytoplasm of goblet cells and columnar cells. MUC2 and MUC3 apomucins and its mRNAs were found in 85.2%, 88.9% and 31.6%; 57.1% of specimens of IM, 67.4%, 66.7% and 57.9%, 43.6% of specimens of gastric carcinoma. There were no associations between expressions of MUC2 and MUC3 genes and different types of IM. Patients with moderate/well differentiation had higher proportion of MUC2 apomucin expression than those with poor differentiation (P < 0.05), and patients with positive staining of MUC3 apomucin in the cancerous tissues had higher proportions of metastasis of lymph nodes (P < 0.01), serosal invasion (P < 0.05) and clinical stages III-IV (P < 0.05). CONCLUSIONS: MUC2 and MUC3 genes are mainly expressed in the mucosa of the duodenum, and marked expression is seen during the neoplastic transformation of stomach mucosa. MUC3 apomucin might have a poor prognostic significance.

Clinical application of radioimmunoimaging with 99mTc-BDI-1 in the diagnosis of bladder cancer.

OBJECTIVE: To evaluate the clinical application of radioimmunoimaging (RII) with 99mTc-BDI-1 in the diagnosis of bladder cancer. METHODS: 32 patients with bladder cancer and 5 with normal bladder were studied. RII was performed 1 hour after intravesical administration of 111-222 MBq 99mTc-BDI-1 followed by washing out and perfusing bladder with 50 ml PBS. The radioactivity ratio of target over background (CT/CB) was calculated. RESULTS: The radiochemical purity of 99mTc-BDI-1 was greater than 95%. RII showed radioactive accumulation area for most of bladder cancers but no radio-active concentration for normal bladder. The sensitivity and specificity were 91.7% and 81.8% respectively with the assignment of the positive criterion of CT/CB > or = 1.40. There was no statistical difference in sensitivity between tumors with diameters > or = 1 cm (1.0-4.2 cm) and < 1 cm (0.2-0.8 cm). CT/CB was related to the pathological grade (G1-G3) of the tumor. CONCLUSIONS: RII by intravesical administration provides a new noninvasive method for the diagnosis of bladder cancer in morphology and in nature of tumor with high sensitivity and specificity. It may be used for the early diagnosis of bladder cancer, follow-up after surgery, and diagnosis of in situ tumor.

[Measurement of gastric emptying with Xianshizi in patients with resected esophageal cancer].

OBJECTIVE: To determine gastric emptying radiological in patients with esophageal cancer and to obtain a similar effect produced by radionuclide study. METHODS: A new product-Xianshizi (XSZ) made of the mixture of barium and silica gel was used quantitatively to evaluate gastric emptying. 17 patients with resected esophageal cancer before and after operation, and 7 healthy volunteers were studied. RESULTS: Preoperative patients had obviously delayed gastric emptying compared with the control group (P < 0.01). Postoperative patients had more delayed gastric emptying compared with themselves (P < 0.05). CONCLUSIONS: Postoperative gastroparesis is related to delayed gastric emptying. The results of this study were identical with those obtained by scintigraphy. Compared with isotope studies, X-ray exam after taking meal composed of XSZ may eventually contribute to detecting delayed gastric emptying quantitatively.

[Evaluation of the results of therapeutic lamellar keratoplasty and penetrating keratoplasty for fungal corneal ulcer].

OBJECTIVE: To evaluate the effects of therapeutic lamellar and penetrating keratoplasty for fungal corneal ulcer. METHODS: Donor corneas preserved in pure glycerine or water-free calcium chloride were used in corneal transplantation for fungal corneal ulcers that were poorly responsive to the anti-fungal medical treatment, 5% iodine was used in lamellar keratoplasty to kill the fungi in the recipient bed, and diluted or original solution of fluconazole was used in penetrating keratoplasty to wash the anterior chamber. RESULTS: In the lamellar keratoplasty group, there were 23 cases; 18 cases were cured with one surgery and 3 cases were cured after second surgery, the success rate being 91.3%. There were 2 cases with recurrence of corneal inflammation, and the lesion was controlled by penetrating keratoplasty. The penetrating keratoplasty group included 31 cases, of them, 27 cases were cured with one surgery and 4 eyes were enucleated due to recurrent corneal inflammation or refractive glaucoma. In the total 54 cases, the inflammation was controlled in 50 cases after therapeutic corneal transplantation, success rate being 92.6%. CONCLUSIONS: For those fungal corneal ulcers that are poorly responsive to anti-fungus eye drops, it is recommended to perform lamellar keratoplasty if the ulcer is located in the anterior 1/2 stroma, the key points of surgery are to remove the affected tissue completely and cauterize the recipient bed with 5% iodine. For deep ulcers or nearly perforative or perforated ulcers, diluted or original solution of diflucan is used to wash the anterior chamber during penetrating keratoplasty, that can help to improve the success rate.

[A cohort study on the relationship between vegetable intake and risks of lung cancer in the Tin Corporation (YTC) miners in Yunnan].

OBJECTIVE: To understand the relationship between vegetable intake and risk of lung cancer in the Tin Corporation (YTC) miners in Yunnan. METHODS: The reproducibility and validity of frequency of vegetable intake in Food Frequency Questionnaire (FFQ) were studied and the correlations between different vegetable intake and frequency and lung cancer relative risk among high risk population of lung cancer by Chi-square, single- and multi-Cox regression were analyzed. RESULTS: Results showed that the reproducibility and validity of wax gourd, cucumber, spinach, wild celery, fresh bush pepper, fresh soybean, sweet potatoes, asparagus, fresh bamboo shoot were good (r = 0.20 - 0.33, P < 0.05); There were significantly different incidences of lung cancer in different frequencies of wax gourd, cucumber, wild celery, fresh bush pepper out of 11 vegetables intake, and also in different total vegetable intakes (chi(2) = 8.83 - 30.64, P < 0.05). 11 vegetables and total vegetable intake were significantly negatively correlated with risks of lung cancer (RR < 1, P < 0.05). Only 2 vegetable frequencies were negatively correlated with risk of lung cancer by adjusting confounders such as age, occupational history, and smoking (P < 0.05). CONCLUSION: This suggested that vegetables intake may have a role in decreasing the incidence of lung cancer.

[The effect of Shenmai injection on the expression of TNFalpha mRNA of the macrophages in scalded mice].

OBJECTIVE: To explore the effect of Shenmai injection on the expression of TNFalpha mRNA of peritoneal macrophages in scalded mice. METHODS: BALB/c mice inflicted with 11% TBSA III degree scalding injury were employed as the model. Shenmai injection was composed of 100 mg of red ginseng and 100 mg of lily turf root in 1 ml injection solution and was administered to mice via peritoneal injection in a dose of 2.5 ml per kilogram of mice body weight per day for 5 days. The expression of TNFalpha mRNA was determined by semi-quantitative RT-PCR. RESULTS: The expression of macrophage TNFalpha mRNA was significantly increased in the scalded mice, while the expression was obviously lowered after the administration of Shenmai injection (P < 0.01) to scalded mice and the survival rate of the scalded mice was thus markedly increased (P < 0.05). CONCLUSION: The beneficial effect of Shenmai injection on the scalded mice might be related to its inhibitory effect on the hyperexpression of macrophage TNFalpha mRNA. Shenmai injection could lower down the expression of TNFalpha in scalded mice and raise the survival rate of scalded mice.

Data acquisition, data storage, and data presentation in a modern genetics laboratory.

Modern genetic analysis can be divided into three main areas of investigation. The first is data acquisition, in the form of genomic sequence and the cataloguing of polymorphism data of the single nucleotide polymorphism variety (so called SNPs). Once identified, such genetic information can be adapted into high throughput tests to examine genetic information in large populations, making the analysis of sufficiently large numbers both cost and time effective so that relatively low-penetrant genetic effects can be accurately detected. The third step is correlating variation with phenotype (e.g. disease susceptibility or resistance) for a variety of disorders is paramount in our motivation and indeed is a common goal of modern human genetic analysis. While the technology to acquire vast amounts of genetic data is now well established and continues to expand, the ability to deal with such data, from the process of acquisition, storage, and analysis depends fundamentally on a solid informatics infrastructure as an essential component. Indeed, most of the major gains in productivity in this field are to be realized on the informatics front, and involve automating data acquisition, defining and sorting data in databases for quality control and analysis and facilitating access to data for the large variety of data analyses. Informatics-related issues including those relating to data acquisition, database structure, and analysis tools are summarized here in an effort to define some of the issues relevant to establishing informatics infrastructure in a small genetics laboratory focused on resequencing human immune response genes. From inherited diseases to drug efficacy to the specific genetic changes occurring during tumor development, this new field of medical genetics promises a profound impact on the state of human health. Ultimately, any and all advances in this field will continue to depend on major investments in informatics.
