PURPOSE: The effects of novel nitric oxide (NO) donors administered intracavernosally and transurethrally on erectile function in the anesthetized cat were evaluated. MATERIALS AND METHODS: In pentobarbital-anesthetized cats, increases in intracavernosal pressure, penile length, and duration of erectile response were determined after intracavernosal and transurethral injections of novel NO donors (MAHMA/NO, PAPA/NO, DEA/NO, PIPERAZI/NO and PROLI/NO). All parameters were measured after administration of NO donors intracavernosally via a 30-gauge needle and urethrally via a Jelco i.v. catheter in a volume of 200 microliters. Systemic arterial pressure was also assessed in these experiments. All NO donors were compared with a triple-drug control combination comprised of papaverine (1.65 mg.), prostaglandin E1 (0.5 microgram.), and phentolamine (25 micrograms.). RESULTS: MAHMA/NO, PAPA/NO, DEA/NO, PIPERAZI/NO and PROLI/NO induced dose dependent increases in intracavernosal pressure and penile length (p < 0.05) when administered intracavernosally. The increases in cavernosal pressure and penile length were comparable to those observed with the triple-drug control combination. The maximum increase in cavernosal pressure in response to PROLI/NO and PAPA/NO was associated with no significant change in systemic arterial pressure. Transurethral administration of PROLI/NO and PIPERAZI/NO induced dose-dependent increases in cavernosal pressure and penile length (p < 0.05). The response was similar to that of the triple-drug control combination, except that transurethral PROLI/NO and PIPERAZI/NO had no significant effect on systemic blood pressure. CONCLUSIONS: NO donors caused dose-dependent increases in cavernosal pressure when administered intracavernosally and transurethrally. These data suggest further exploration of the use of NO donors for the treatment of erectile dysfunction.
OBJECTIVE: Studying gene diagnosis of trisomy 21 by means of quantitative polymerase chain reaction(PCR). METHODS: Polymorphic short tandem repeat(STR) at D21S11 served as the gene marker, and a pair of primers was synthesized. The samples from 11 normal subjects and 28 cases of trisomy 21 were analyzed by quantitative PCR. RESULTS: 10 normal subjects showed two bands with a ratio of 1:1, and a subject showed one band. Of 28 cases of trisomy 21, 24 cases showed two bands with a ratio of 2:1, 3 cases showed three bands with a ratio of 1:1:1, and 1 case showed one band. CONCLUSION: Polymorphic STR at D21S11 is a valuable gene marker for diagnosis of trisomy 21. Trisomy 21 can be diagnosed rapidly and accurately within 24 hours by quantitative PCR.
BACKGROUND: Islet isolation exposes the islet to a variety of cellular stresses and disrupts the cell-matrix relationship--events known to be associated with apoptosis. The purpose of this study was to determine whether islet isolation leads invariably to islet cell death and to specify the mechanisms involved. METHODS: Canine islets were isolated using Liberase CI and purified using a centrifuge. Islets were sampled for up to 5 days in culture and analyzed by routine histology, electron microscopy, immunocytochemistry, and reticulin staining for basement membrane. Apoptosis was assessed by cell death enzyme-linked immunosorbent assay and terminal deoxynucleotidyl transferase-mediated decoxyuridine triphosphate nick and labeling (TUNEL) assay. Activation of the prosurvival ERK1/2 and proapoptotic p38 and JNK were determined by immunoblotting. RESULTS: Immediately after isolation, the peri-insular basement membrane was absent, and integrin-alpha 5 expression diminished. DNA fragmentation rose from 2.5 +/- 1.8 (arbitrary units) on the day of isolation to 42.4 +/- 6.7 48 hours later (P < .05), coinciding with the appearance of pyknotic nuclei and apoptotic bodies. The apoptotic index determined by TUNEL assay increased from 5% +/- 1% on the day of isolation to 60% +/- 2% on day 5 (P < .01), and most of the affected cells were beta-cells. Finally, the p38 and JNK activity were elevated relative to ERK1/2. CONCLUSIONS: During isolation, islet cells undergo profound changes in structure and function, resulting in beta-cell apoptosis. These findings suggest that strategies directed to the manipulation of the cell-matrix relationship and the modulation of mitogen-activated protein kinase signal transduction may offer a valuable new approach to improving islet transplant outcome.
OBJECTIVE: To clone genes associated with the genesis of human esophageal cancer. METHODS: Identifying missing or low expressing cDNAs in human esophageal cancer tissues by mRNA differential display and examining its mRNA expression in 4 human cancer cell lines, 9 fetal tissues and other matched esophageal cancer tissues by Northern blot, dot blot and RT-PCR. RESULTS: One cDNA fragment named C6-2A, was cloned and sequenced. There was no identical sequence with C6-2A in BLASTN database; but in querying Genbank EST, the authors found that C6-2A was identical with ne27b03.s1NCI-CGAP-C03 humans sapiens cDNA clone IMAGE:898541 3' and zv30g07.rl Soares ovary tumor NbHOT homo sapiens cDNA clone 755196. 6/6 esophageal cancer tissues in Northern blot and 7/8 in dot blot did not or slightly express C6-2A. RT-PCR analysis showed that C6-2A was expressed much lower in 17/20 esophageal cancer tissues than adjacent microscopically normal mucosa, highly expressed in fetal esophageal mucosa, skin, cerebrum, placenta; moderately expressed in fetal stomach and liver, but not detected in fetal heart, small intestine and kidney. CONCLUSION: The high frequency of deletion of decreased expression of C6-2A in esophageal cell lines and human esophageal cancer tissues suggested that C6-2A might be involved in the carcinogenesis of esophagus.

AIM: To study the action of free radical in the spin-labeled podophyllotoxin derivative, podophyllic acid piperindyl hydrazone nitroxide radical (GP-1) on its antitumor activity and toxicity, by comparison with those of its free radical reduced product, podophyllic acid piperindyl hydrazone (GP-1-H). METHODS: After treatment with GP-1 and GP-1-H, the inhibitory effects on the growth of mouse transplantable tumors were determined; MTT formazan formation, [3H]deoxythymidine ([3H]TdR) incorporation, cell cycle progression, and mitotic index of SGC-7901 or L1210 cells were measured; the acute toxicity and immune function of mice were assayed. RESULTS: At doses of 1/6 and 1/12 LD50, the inhibitory rates against Lewis lung carcinoma were 60.3% and 42.1% (GP-1), 38.9% and 10.3% (GP-1-H), respectively; more effective antitumor activity of GP-1 against P388, HePS, and S-180 than that of GP-1-H were found. In vitro, GP-1 exhibited more powerful inhibitory effects on the proliferation and DNA synthesis of SGC-7901 and L1210 cells than GP-1-H. GP-1 and GP-1-H arrested the L1210 cells at G2/M phase with a corresponding decrease of the cells in G1 phase, and increased the mitotic index of the cells; but the effects of GP-1-H were weaker than those of GP-1. After treatment with doses of 1/4 and 1/8 LD50 for 5 d, no significant difference on immune function of mice between GP-1 and GP-1-H was found. CONCLUSION: GP-1 had more powerful antitumor activities than GP-1-H. The free radical in the spin-labeled podophyllotoxin derivative, GP-1, played an important role in its antitumor activity.
OBJECTIVE: To investigate the change and clinical significance of tissue factor pathway inhibitor (TFPI) and antithrombin-III (AT-III) in the plasma of patients with hepatic inflammatory disease (HID). METHODS: The plasma levels and activation of TFPI and AT-III in patients with HID (Group I, n = 82) were compared to that in healthy people (Group II as control, n = 30). RESULTS: The plasma level of TFPI:Ag in Group I was higher (P < 0.05) and its activation was also increased dramatically (P < 0.01), the level of AT-III: A was lower in patients with chronic hepatitis B (P < 0.05) and no difference was found in other indices between the two groups. CONCLUSIONS: The elevation of TFPI:Ag and TFPI:A in hepatic inflammatory diseases is an indication of inflammation, and hepatic tissue injury in broad, the lower level of AT-III liver secretions.
AIM: To elucidate the protection of l-stepholidine (SPD) on neuronal morphology and function against the striatal ischemic injury in rat. METHODS: The forebrain ishemia to Sprague Dawley rats was induced with four-vessel occlusion. Histological examination was performed on the dorsolateral striatum with cresylviolet stain. In striatal slices of rat as an in vitro ischemic model, the activity of calcium/calmodulin-dependent protein kinase II (CCDPK) and lactate dehydrogenase (LDH) was examined by the method of 32P-incorporation and colorimetry, respectively. RESULTS: In the SPD-treated groups, most of the neurons in the striatum kept the normal morphological appearance after 30-min ischemia followed by 6-h or 12-h reperfusion. The number of neurons was much more in SPD groups than that in vehicle group. The sparse and abnormal neurons were observed in the vehicle group. SPD attenuated the ischemic effect on the CCDPK activity in striatal slices. In addition, SPD inhibited the leakage of LDH from neurons induced by ischemia in incubated striatal slices. CONCLUSION: SPD protected striatal neurons against ischemic injury and antagonized the inhibitory action on CCDPK activity induced by ischemia. SPD reduced the leakage of LDH from striatal neurons induced by ischemia.

OBJECTIVE: To study the therapeutic effects of Chinese herbs Drynaria, Cynomorium, Epimedium and Chain Fern on osteoporotic ovariectomized (OVX) rat models. METHODS: 54 Wistar female rats at the age of 3 months were randomly divided into 2 groups: control groups and experimental group. The control group included: normal groups (N), shamed oprations group(S) and ovariectomized group (OVX) (10 in each group); The experimental group: group A (Drynaria), group B (Cynomorium), group C (Epimedium) and group D (Chain Fern) (6 in each group). Each of the experimental groups was fed separately with the 4 Chinese herbs (0.25 g/100 g B.W/day) respectively by mouth every day for 2 months. Double tetracycline labels were carried out on these animals before the sacrifice. Their left tibia was reserved for the preparation of bone specimen and was studied by bone histomorphometry. RESULTS: The percentage of trabecular volume in total bone volume (TV/TBV) decreased significantly in OVX rats as compared with shame group (P < 0.01). However, after treatment with Drynaria or Epimedium, TV/TBV increased significantly as compared with that before treatment (P < 0.05). The surface area of tetracycline label in Drynaria group lowered and osteoid percentage increased in Epimedium group. CONCLUSIONS: The 4 Chinese herbs have some therapeutic effects on osteoporotic rat models. Drynaria and Epimedium appeared to work better.
OBJECTIVE: To observe the effect of repairing stricture of the cervical esophagus with platysma myocutaneous flaps. METHODS: Fifteen patients with stricture of the cervical esophagus were treated by repair with platysma myocutaneous flaps. The causes of stricture included anastomotic stenosis of the cervical esophagus after colon graft transplantation due to esophageal caustic injury (12 patients), anastomotic stenosis after stomach transposition due to resection of the upper esophageal cancer (2), and stricture caused by limited erosive injury of the cervical esophagus (1). The lengths of stricture were about 3.5-4.5 cm. The stricture was incised longitudinally. The platysma myocutaneous flaps about 4 cm-5 cm x 6 cm-7 cm were resected and were transposed to repair the stricture. RESULTS: All flaps survived and no death or serious complications occurred after operation. All patients gained normal functions in swallowing food. CONCLUSION: Repairing stricture of the cervical esophagus with platysma myocutaneous flaps is simple, safe and effective.
OBJECTIVE: To investigate the influence of roxithromycin (RXM) on the production of glycocalyx (GLX) and the synergic effect of RXM and fleroxacin (FLRX) on P. Aeruginosa biofilms. METHODS: Bacterial biofilm (BF) was established and was influenced by RXM and FLRX with different concentrations. The samples were detected with scanning electron microscope and by a rapid method after BF was stained with AgNO3 solution. The synergism of antibacterial activities of RXM and FLRX to P. Aeruginosa was studied by computer image analysis and MTT method. RESULTS: GLX production was reduced significantly by both 1/16 MIC and 1/4 MIC RXM (P < 0.01). 1/16 MIC and 1/4 MIC RXM showed no bactericidal activities to P. Aeruginosa in biofilms, while viable counts in biofilms almost had no difference. However, RXM could enhance the bactericidal activity of FLRX to P. Aeruginosa in biofilms. When FLRX of different concentrations was combined with 1/16 MIC and 1/4 MIC RXM respectively, the difference of the stained BF was counted up by computer image analysis system with significance of P < 0.05 and the viable counts were reduced significantly (P < 0.05). The results was in accordance with SEM pictures. CONCLUSION: RXM has no direct bactericidal activities to P. Aeruginosa in biofilms, but it could inhibit GLX production. It can be considered that RXM could enhance antibacterial activities of FLRX by enhancing the permeability of FLRX into biofilms.
OBJECTIVE: To evaluate the clinical effects, pharmacokinetics, post-antibiotic effect (PAE) and toxicity of netilmicin as a single daily dose in the treatment of lower respiratory tract infection. METHODS: 48 cases were divided into 3 groups: In the first group, Netilmicin(6 mg.kg-1.d-1) was administered in a single daily dose; in the second group, netilmicin (200 mg/d) was combined with cefazolin (3 g, Q12 h); and in the third control group, the combination of cefazolin and Amikacin was used. Pharmacokinetics were studied in 7 patients using the TDX system, and PAE induced by Netilmicin was determined by the Avantage microbiologic system. Clinical symptoms, laboratory studies, chest X-rays, and side effects were observed. RESULTS: The overall clinical effects of the first group were better than those of the third group. The mean serum concentration of netilmicin was 27.23 mg/L, the valley serum concentration was 0.23 mg/L, T1/2 beta was 5.059 h, AUC was 70 micrograms.h-1.ml-1.netilmicin at concentrations 0.5, 1.0 and 4 times the MIC showed different degrees of PAE against 4 strains of bacterium. Nephrotoxicity and ototoxicity were not found in the treatment group. CONCLUSIONS: Netilmicin in a single daily dose resulted in a high peak serum concentration and big AUC. As a concentration-dependent bactericidal agent, netilmicin showed a longer PAE and better therapeutic effects.
OBJECTIVE: To evaluate the results of combination chemotherapy with cisplatin (DDP) plus 5-fluorouracil(5-FU) in the treatment of advanced esophageal cancer. METHODS: From 1988 through 1997, 156 patients with advanced esophageal cancer were enrolled in this study. The median age of the patients was 60. Squamous-cell cancer was the most common type of malignancy. All but 16 patients received no previous chemotherapy. RESULTS: Complete response was observed in 26 patients and partial response in 71 patients, with an overall response rate of 62.2%. The response appeared to correlate with dose intensity, as 65.7% response rate was achieved with a dose of DDP 75-100 mg/m2 versus 43.7% response rate with DDP 50-70 mg/m2. The median response duration was 4.5 months and the median survival period was 8.2 months. Nausea/vomiting, neutropenia and phlebitis were the main adverse reactions in all treated cases. CONCLUSION: A high response rate was obtained in advanced esophageal cancer treated by DDP plus 5-FU with tolerable toxicity.
OBJECTIVE: To evaluate antibacterial activities in vivo and in vitro of fleroxacin and lomefloxacin. METHODS: Minimal inhibitory concentration (MIC) and serum bactericidal activity (SBA) domestic fleroxacin and lomefloxacin were determined by agar disk dilution method and microplate method respectively. A clinical observation of fleroxacin and lomefloxacin for treating acute bacillary dysentery was carried out. RESULTS: MIC(90) of fleroxacin against E. coli, K. pneumoniae, S. dysenteriae, S. aureus was 0.25-2 mg/L, being the same as that of ofloxacin. However, MIC(90) of lomefloxacin against the above mentioned species was twice or fourfold as that of ofloxacin and fleroxacin. The percent age of peak SBA > or = 1:8 with ofloxacin, fleroxacin and lomefloxacin against E. coli, K. pneumoniae, S. dysenteriae was 80%-100%, 90%-100% and 50%-80% respectively (P < 0.01). The clinical cure rates of acute bacillary dysentery were 93.3%, 100% and 100% and the bacterial clearance rates were 93.3%, 100% and 100% in the lomefloxacin, fleroxacin and ofloxacin group respectively. CONCLUSION: The antibacterial activity in vivo and in vitro of fleroxacin against E. coli, K. pneumoniae, S. dysenteriae was similar to that of ofloxacin and stronger than that of lomefloxacin.
OBJECTIVE: To study the feature of pancreatic microcirculatory impairment in caerulein-induced acute pancreatitis. METHODS: The AP model of subcutaneous injection of caerulein was studied by intravital fluorescence microscopy of erythrocytes labeled by FITC (FITC-RBC) and by scanning electron microscopy of vascular corrosion casts and light microscopy of Chinese ink-injected/cleared tissues. RESULTS: Animals treated with caerulein showed hyper-amylases. Contractions of intralobular arteriolar sphincter, presence of vacuoles in all the layers of sphincter, gross irregularity in capillary network of acini were observed in caerulein-induced groups. The decrease of pancreatic capillary blood flow (P < 0.01), reduction of functional capillary density, and irregular intermittent perfusion (P < 0.05) were found. CONCLUSIONS: Impairment and contraction of pancreatic intralobular arteriolar sphincter are the initial microcirculatory lesions in the early phase of acute pancreatitis induced by Caerulein, and play a key role in the pancreatic ischemia and pancreatic microvascular failure in acute pancreatitis.
OBJECTIVE: Effect of zinc on proliferation and differentiation of osteoblast in rats was studied in vitro. METHODS: Osteoblasts were isolated from the rat calvaria and subcultured in DMEM media. Rats were divided into four groups in the study, with administration of 0 micromol/L (as control), 10 micromol/L, 25 micromol/L and 50 micromol/L of Zn(2+), respectively. DNA synthesis at varied time points was determined with incorporation of (3)H-thymidine, and cell cycle was analyzed by flow cytometry. Synthesis of collagen was measured with incorporation of (3)H-proline. Activity of alkaline phosphatase (AKP) and osteocalcin content were measured by enzyme dynamics method and radioimmunoassay, respectively. RESULTS: Incorporation of (3)H-thymidine in the three groups with different zinc-supplement was significantly higher than that in the control group at each time point, and supplement with 25 micromol/L and 50 micromol/L of Zn(2+) could promote the transition of cell cycle from phases G(0)/G(1) to S phase. Syntheses of collagen and osteocalcin and activity of AKP in the groups with zinc-supplement were greater, as compared with those in the control group. CONCLUSION: Zinc could promote proliferation and differentiation of the rat osteoblast in vitro.
