The effects of extracellular Ca2+ ions on purinergic responses were examined in swine tracheal smooth muscle cells (TSMCs) in primary culture. ATP (1 microM to 1 mM) and alpha, beta-methylene adenosine 5' triphosphate (alpha, beta-Me ATP) (100 microM and 1 mM) concentration-dependently increased [Ca2+]i in the presence and the absence of extracellular Ca2+. Responses to ATP (10 microM to 1 mM) in the presence of extracellular Ca2+ were significantly larger than those in its absence (n=8), whereas those to alpha, beta-Me ATP were not significantly different between the presence (n=7) and the absence (n=8) of extracellular Ca2+. Responses to ATP (1 mM) at extracellular Ca2+ concentration ([Ca2+]o) of 10 and 5 mM were significantly larger than that at extracellular EGTA concentration ([EGTA]o) of 1 mM (p< 0.01, n=5), whereas the responses to alpha, beta-Me ATP (1 mM) at 10 and 5 mM [Ca2+]o were significantly smaller than that at [EGTA]o of 1 mM (p<0.05, n=5). Increasing [Ca2+]o to 5 mM after the application of either 1 mM ATP (n=4) or 1 mM alpha, beta-Me ATP (n=4) in the absence of extracellular Ca2+ (1 mM [EGTA]o) further increased [Ca2+]i, though the increases in [Ca2+]i by agonists in 1 mM [EGTA]o had been already maximal. Incubating cells for 300 s in 5 mM [Ca2+]o before the application of ATP (1 mM) significantly increased the response to the drug than that obtained by incubating cells for 6 sec in 5 mM [Ca2+]o before the drug application (p< 0.01, n=4). However, alpha, beta-Me ATP (1 mM) induced similar responses by incubating cells for 30 or 300 s in 5 mM [Ca2+]o to that by incubating them for 6 s. These results suggest that the effect of alpha, beta-Me ATP in swine TSMCs in primary culture is mainly through Ca2+ release and that its effect on Ca2+ entry is smaller than other nucleotides.
Effects of intrathecal (i.t.) administration of monoamine antagonists on formalin-induced neuronal c-Fos expression in two sides of the lumbar dorsal horn were observed in rats with unilateral transection of the dorsolateral funiculus at T11-12 level. The results showed that: 1) pretreated with i.t. normal saline (control) and then an equal volume of formalin was injected into the two hindpaws, the number of Fos-like immunoreactive neurons were 44% lower on the side of lumbar dorsal horn with intact dorsolateral funiculus (57 +/- 3.1 vs. 103 +/- 3.8). 2) Pretreatment with i.t. phentolamine (a non-selective alpha-adrenoceptor antagonist) caused an increase of Fos-like immunoreactive neurons on the intact side so showing only a reduction rate of 23% to the lesioned side (p < .01); 3) pretreatment with i.t. cyproheptadine (a 5-HT-receptor antagonist) caused a similar reduction rate of 21% (p < .01) of Fos-like immunoreactive neurons on the intact side; and 4) combined i.t. pretreatment with phentolamine and cyproheptadine caused a reduction of Fos-like immunoreactive neurons of only 4% on the intact side, namely, the differences in the number of Fos-like immunoreactive neurons on two sides of the lumbar spinal cord owing to the unilateral dorsolateral funiculus lesion were nearly abolished by i.t. coinjection of phentolamine and cyproheptadine. The results indicate that 1) peripheral noxious inputs can provoke a spinally-descending inhibitory effect on the spinal nociceptive transmission via the dorsolateral funiculus and 2) the descending fibers in dorsolateral funiculus exert their action mainly through the release of either norepinephrine or 5-HT at the spinal level.
Thirty-six patients of cerebral hemorrhage in basal ganglia were studied between medline structure shifting on CT scan and assessment of clinic neurological function, defect (AND). The differences were significant between the following two groups: including in parameter A (the distance from center of hematoma to normal midline), 26.01 +/- 6.06 mm in survivor group, 21.53 +/- 5.03 mm in death group; in parameter B (the distance from the farthest point in shifted midline to the normal midline), 3.84 +/- 2.66 mm in survivor group, 10.32 +/- 4.24 mm in death group; in parameter C (ratio of the partial length of shifted midline to all length of the normal midline), 0.347 +/- 0.136 in survivor group; 0.582 +/- 0.082 in death group. Having analized these parameters, the authors found that the mortality rates were 0 and 100% if B was < or = 4.0 mm and > 10.0 mm respectively and the mortality rates were 0 and 90% if C was < or = 0.30 and > 0.50. The authors also found that there was closed correlation between the parameters B or C described midline shifting and AND.

The effects of high dietary zinc on alloxan induced diabetic mice were studied. The results showed that high zinc could sharply decrease the body weight and increase food intake, water intake and urine of diabetic mice, and increase blood sugar levels in diabetic mice. After alloxan injection in diabetic model group, urine sugar levels reached [symbol: see text] at the 2nd day and [symbol: see text] at the 6th day. After alloxan injection in high-zinc-diabetic mice group, urine sugar levels reached [symbol: see text] and [symbol: see text] at the 2nd and the 4th day, respectively. These results suggested that the high level of dietary zinc may be damage to diabetic patients.
The dose-effect relationship of hypolipidemic effect of fish oil was studied. The rats were divided into four groups: 2.5, 5, 10 g/kg and high dietary fat control group. The experimental rat model of high serum lipids was developed. The results showed that there was a linear correlation between the levels of TG, HDL-c and the dosage of fish oil. Fish oil reduced TC level and elevated HDL-c level steadily. But no linear correlation was observed between TC, LDL-c and the dosage of fish oil. The level of TC and LDL-c decreased markedly at the dosage of 2.5 g/kg of fish oil.
Remitting hemiplegic spasticity in apoplexy (HSA) is an important problem in today's clinical study. Through teaching and clinical practice, the authors summed up the effective acupuncture methods for remitting HSA: puncture deeply the acupoints on the superior-spasm side (SSS) by filiform needles so as to obtain the intensive needling sensations in the deep tissues (ISDT) until the superior spasm is immediately remitted; tap the skin on the inferior-spasm side (ISS) by skin needles until the corresponding muscle contracts. The methods have showed a significant immediate and long-term therapeutic effect.
This experiment in rats was designed to investigate the effect and mechanism of nitric oxide (NO) in the induction of hypoxic pulmonary hypertension. The plasma concentrations of NO in normal controls and in the 1-, 2- and 3-week hyporemic ventilation groups were measured. The hemodynamic and pathological changes were observed in rats of the 2-week group after bolus injection of L-Arginine and NG-nitro-L-arginine. The results showed that NO concentrations of the 1-2- and 3-week groups were 5 +/- 2.67 mumol/L 2.1 +/- 0.41 mumol/L and 0.5 +/- 0.16 mumol/L respectively, which were significantly lower than the control group's 6.73 +/- 1.83 mumol/L (P < 0.05). Bolus injection of L-Arginine 100 mg.kg-1.d-1 could relieve chronic hypoxic pulmonary hypertension and decrease the thickening of pulmonary arteries, but L-NNA could antagonize the effect of L-Arginine. This experiment demonstrates that chronic hypoxemia may decrease the release of NO and result in pulmonary hypertension. L-Arginine may be used to relieve pulmonary hypertension, but L-NNA may antagonize the effect of L-Arginine.
To determine type II diabetes in association with alteration in beta-cell function, we studied 30 patients with newly diagnosed diabetes and 30 matched controls. The beta-cell 1st phase insulin response was measured by intravenous glucose tolerance test and the 2nd phase insulin response was measured by oral glucose tolerance test. The results revealed that in type II diabetes mellitus the pulse of 1st phase insulin secretion could not be found. The amount of 2nd phase insulin secretion of the patients was lower than that normal controls, and the patients' secretary pulse of insulin was not concomitant with glucose pulse. These findings again suggest it is the defect in beta-cell function, not the peripheral resistance an essential cause of overt type II diabetes.
To provide further understanding of uncinate process, 15 uncinate processes were examined under scanning electron microscope, and 32 high-resolution coronal CT scans were studied in patients with septal deformity or sinusitis. It was found that(1)pathological ultrastructural changes of uncinate process were noted in patients with chronic sinusitis; (2) medially deviated, laterally deviated, pneumatization and hypertrophy of uncinate process were demonstrated. The following conclutions were drawn(1)the anatomic variations in ostiomeatal complex is responsible for the formation of chronic sinusitis; (2)for patients with chronic sinusitis, uncinatectomy is the treatment of choice; (3)preoperative and intraoperative studies of CT scans are mandatory for avoidance of complications.
Five compounds were isolated from the alkaloid extract of Rhizoma Pinelliae Pedatisecta and their structures were determined as pedatisectine F(I), hypoxanthine (II), erythritol (III), uridine (IV) and pedatisectine G (V), of which I and V are new compounds, while II, III and IV were found in this plant for the first time.
Magnetite can markedly inhibit the rodent turn-around reaction induced by acetic acid, reduce the threshold dose of pentobarbital sodium and shorten rodent's incubation period of falling asleep. It has also the following effects; antagonizing metrazol which causes rodent convulsions, postponing the incubation period of being startled by Huisuling, cutting down the extent of rodent's foot swells caused by JCCJ, and diminishing bleeding time and congulating time.
The doses of linac convergent-beam irradiations to pineal region were measured in an anthropomorphic phantom. The feasibility of a linac accelerator as a radiosurgery tool to treat small sized intracranial masses was evaluated. The influences of some irradiation parameters on the dose distributions were discussed. Linac-based radiosurgery can result in dose distributions similar to those achieved by gamma knife, so it is another safe and effective treatment technique of neurosurgery. The smaller the aperture is and the greater the irradiation arc travel is, the larger the dose gradient will be and the better the dose distribution will be. The maximum target doses are dependent greatly upon the collimater aperture. The appropriate irradiation arc travel and margin isodose were also discussed.
A series of DNA primers specific for the specific fragments of nm23-H1 and nm23-H2 were designed and synthesized. The specific fragments of nm23-H1(185 bp) and nm23-H2(145 bp) were amplified from human blood DNA by using polymerase chain reaction (PCR). The recovered PCR products were treated with Klenow fragment, inserted into pGEM-3zf(+) vector with blunt-end ligation, and then transformed into competent cell JM109. The positive colonies were directly identified by colour screening on indicator plates. The recombinant plasmids were digested by Alu I and identified by PCR. The results showed that the authors had obtained the specific fragments of nm23-H1 and nm23-H2 respectively, and these specific fragments could be used for study the expression of nm23-H1 and nm23-H2 seperately.
Flavonol isomers, which differ only in the position of hydroxyl group substitute on their chemical structures, showed different chromatographic behaviors. These polar compounds, in general, can be analyzed by high performance liquid chromatography. In this paper, the separation of flavonol isomers by home-set up packed column supercritical fluid chromatograph with carbon dioxide modified by ethanol containing 0.5% (V/V) phosphoric acid as mobile phase is described. The effects of temperature, pressure, composition of mobile phase and packed column type on the separation were studid. The addition of phosphoric acid to the mobile phase, which can not result in corrosion on the chromatographic system, enabled flavonol to elute from the column, and showed that the composition of the mobile phase was the most pronounced factor influencing the separation. Three columns were evaluated for the separation. It showed that the phenyl-bonded silica column was the best and the ODS column was the worst for the isomers separation. Increasing pressure shortened the retention time of each compound with good resolution, and higher temperature led to longer retention time.

The first clinical trial of a DNA vaccine designed to protect against malaria has just commenced. This vaccine has been designed to induce protective CD8+ T cell responses against Plasmodium falciparum infected hepatocytes. Herein, we review the rationale behind the development of vaccines that induce protective CD8+ T cells, the strategy for the development of a DNA vaccine designed to protect against falciparum malaria, and the experimental data in rodent models and nonhuman primates which has provided the foundation for trials of DNA vaccines against P. falciparum malaria in humans.
