Chemical ligation of folded recombinant proteins: segmental isotopic labeling of domains for NMR studies.

A convenient in vitro chemical ligation strategy has been developed that allows folded recombinant proteins to be joined together. This strategy permits segmental, selective isotopic labeling of the product. The src homology type 3 and 2 domains (SH3 and SH2) of Abelson protein tyrosine kinase, which constitute the regulatory apparatus of the protein, were individually prepared in reactive forms that can be ligated together under normal protein-folding conditions to form a normal peptide bond at the ligation junction. This strategy was used to prepare NMR sample quantities of the Abelson protein tyrosine kinase-SH(32) domain pair, in which only one of the domains was labeled with 15N. Mass spectrometry and NMR analyses were used to confirm the structure of the ligated protein, which was also shown to have appropriate ligand-binding properties. The ability to prepare recombinant proteins with selectively labeled segments having a single-site mutation, by using a combination of expression of fusion proteins and chemical ligation in vitro, will increase the size limits for protein structural determination in solution with NMR methods. In vitro chemical ligation of expressed protein domains will also provide a combinatorial approach to the synthesis of linked protein domains.

The quantitative anatomy of the laminas of the spine.

STUDY DESIGN: This study evaluated the dimensions of the laminas from C2 to L5 using adult spine specimens. OBJECTIVES: To provide a set of quantitative data for the laminas from C2 to L5. SUMMARY OF BACKGROUND DATA: Anatomic evaluation of the pedicle and facet joint in the spine has been extensively reported. No detailed studies of the laminas from the cervical to the lumbar spines exist. METHODS: Thirty-seven spines from C2 to L5 were directly evaluated for this study. Anatomic evaluation of the laminas included the laminar height, width, thickness, and angulation. RESULTS: In general, the measurements were greater in male specimens than in female specimens, although significant differences (P < 0.05) between male and female specimens were noted in only three measurements. The greatest laminar height was at T11 (25.1 +/- 2.5 mm), and the least was at C4 (10.4 +/- 1.1 mm). The greatest laminar width was at L5 (15.7 +/- 2.0 mm), and the least was at T4 (5.8 +/- 0.8 mm). Laminar widths in the cervical region were slightly more than those in the thoracic region. The greatest laminar thickness was at T2 (5.0 +/- 0.2 mm), whereas the least was at C5 (1.9 +/- 0.6 mm). Laminar thickness tended to increase in the upper thoracic region and to decrease slightly in the lower thoracic region. The mean laminar thickness of the lower cervical region was least in the whole spine. The widest angle was at C3 (116.1 +/- 8.8 degrees) and T7 (112.3 +/- 8.0 degrees) and the narrowest was at C2 (99.1 +/- 8.0 degrees) and L3 (99.9 +/- 6.3 degrees). The slope angles of the laminas varied from 97.8 +/- 3.0 degrees at T9 to 129.0 +/- 7.5 degrees at L3. CONCLUSIONS: Surgical placement of sublaminar instruments may benefit from this quantitative study through the use of the provided anatomic parameters of the laminas.

Rate constants of acetylcholine receptor internalization and degradation in mouse muscles.

The rate constants for internalization and subsequent extrusion of acetylcholine receptors (AChRs) during degradation in adult innervated and denervated mouse diaphragm muscles were determined using proteinase K (PK) digestion. This procedure separated (125)I-alpha-bungarotoxin (Bgt)-labeled AChRs into PK-sensitive and PK-resistant compartments. The time course of the residual radioactivity in these two compartments suggested that they represented surface membrane and internalized compartments, respectively. The data were compatible with a mathematical model based on the assumption that during degradation of AChRs a surface compartment, A, fed an internal compartment, B, with an internalization rate constant (k(i)), and that B is drained from the cell with an extrusion rate constant (k(o)). Using the mathematical model, we were able to determine that k(i) and k(o) were, respectively, 0.068 (t(1/2) approximately 10.2 days) and 0.69-0.55 (t(1/2) approximately 1.0- 1.25 days) for innervated muscle and were, respectively, 0.69 (t(1/2) approximately 1.0 day) and 6.93 (t(1/2) approximately 0.1 day) for denervated muscle. Thus, the rate for internalization was about 8-10 times slower than that for extrusion from the cell for both the slowly degrading innervated (Rs) AChRs and for the rapidly degrading denervated (Rr) AChRs. This inequality between k(i) and k(o) therefore allows the combined quantity of A(t) + B(t), usually measured in AChR degradation studies, to approximate a single exponential.

Chiral resolution of atropine, homatropine and eight synthetic tropinyl and piperidinyl esters by capillary zone electrophoresis with cyclodextrin additives.

Chiral resolution of atropine, homatropine and eight synthetic tropinyl and piperidinyl esters were studied by capillary zone electrophoresis with cyclodextrin additives. Atropine and eight synthetic derivatives were successfully resolved by heptakis-(2,3,6-tri-O-methyl)-beta-cyclodextrin (TM-beta-CD) at concentrations ranging from 10 to 40 mM. Homatropine was baseline resolved by 10 mM beta-cyclodextrin and hydroxypropyl-beta-cyclodextrin (HP-beta-CD), respectively. The developed method was employed for the determination of atropine enantiomers in human serum.

Increased expression of a plant actin gene during a biotrophic interaction between round-leaved mallow, Malva pusilla, and Colletotrichum gloeosporioides f. sp. malvae.

Two actin genes, actA from the hemibiotrophic anthracnose fungus, Colletotrichum gloeosporioides (Penz.) Penz. & Sacc. f. sp. malvae, and act1 from its host, Malva pusilla (Sm.) were cloned from a cDNA library developed from infected host tissue. The actin gene, actA, of C. gloeosporioides f. sp. malvae, which is similar to that of other euascomycetes, appears to be expressed constitutively. The actin gene of M. pusilla is most similar to one of the actin genes of Arabidopsis thaliana that is unique in being responsive to environmental stimuli such as wounding. Expression of actA was used to follow the growth of the fungus in the plant tissue. Low actA expression occurred until 72-96 h after inoculation and then increased rapidly, corresponding with the timing of the shift from slower biotrophic fungal growth to much more rapid necrotrophic growth. In contrast, expression of act1 approximately doubled during the biotrophic phase and then rapidly declined during the necrotrophic phase. Increased host actin expression could be due to host cytoskeleton rearrangement in response to biotrophic infection, and the subsequent decrease in host actin expression could be due to host cell disruption resulting from tissue maceration during necrosis. This is the first report of a host actin gene that can increase in expression during a compatible plant-pathogen interaction.

Interleukin-1beta-induced nitric oxide production in rat aortic endothelial cells: inhibition by estradiol in normal and high glucose cultures.

Expression of inducible nitric oxide synthase (iNOS) and the resultant increased nitric oxide (NO) production are associated with septic shock, atherosclerosis, and cytokine-induced vascular injury. Estrogen is known to impact vascular injury and vascular tone, in part through regulation of NO production. In the current study, we examined the effect of physiological concentrations of estradiol on interleukin-1beta (IL-1beta)-induced NO production in rat aortic endothelial cells (RAECs). 17Beta-estradiol significantly decreased IL-1beta-induced iNOS protein levels and reduced NO production in RAECs. High glucose (25 mM) elevated the increase in IL-1beta-induced iNOS protein and NO production. Nevertheless, estradiol still inhibited IL-1beta-induced iNOS and NO production even in the presence of high glucose. These data suggest that estradiol may exert its beneficial effects in part by inhibiting induction of endothelial iNOS, a possible mechanism for the protective effect of estradiol against diabetes-associated cardiovascular complications.

The projection of the cervical disc and uncinate process on the posterior aspect of the cervical spine.

BACKGROUND: Surgical techniques of foraminotomy for decompression of the cervical nerve have been well described in the literature. Excessive resection of the facet joint and laminae may decrease segmental stability and increase scar formation. How much bony resection is adequate to remove a soft or hard disc herniation is not known. No studies regarding this subject are available. METHODS: Thirty-nine adult dry bone spines from C3 to C7 were used and four measurements on each vertebra were taken in this study. The first three measurements included the vertical distances between the superior borders of the lamina and the vertebral body measured at the midline of the laminae, the middle of the lamina, and the lamina-facet junction, respectively. The fourth was the horizontal distance between the medial most border of the superior facet and the tip of the uncinate process. RESULTS: No significant differences between male and female specimens were found in any measurements in this study. The mean vertical distances from the superior border of the lamina inferior to the superior border of the vertebral body measured at the three points for all levels were approximately 1-3 mm, although the standard deviations for those were relatively high. The tip of the uncinate process was located from 2 mm at C3 to 1 mm at C6 medial to the medial most border of the superior facet, and then changed to be located 1 mm lateral to the medial most border of the superior facet. CONCLUSIONS: This study suggests that a semicircular laminotomy placed on the inferior aspect of the lamina above may be adequate for a lateral soft disc herniation because the inferior border of the disc is higher than the superior border of the inferior lamina, whereas a traditional foraminotomy is needed for a hard disc pathology.

The quantitative study of the lateral region to the lumbar pedicle.

BACKGROUND: An anatomic study of the lumbar nerve lateral to the pedicle was performed on 12 cadavers. Three courses of the lumbar nerve were noted around the pedicle. The direct measurements, including the angle of the lumbar rami with the spinal cord, the axis of the pedicle with the cord in the coronal plane, and the distance between the lateral border of the pedicle to the rami, were made bilaterally. RESULTS: The results showed that both angles increased as the lumbar spine descended. The spaces between the lumbar rami and the pedicle were found to be less than 5 mm. CONCLUSION: The region latero-superior to the pedicle had a similar relationship with the region medial inferior to the pedicle. Nerve entrapment was observed around the lumbar pedicle.

PCR-based quantitation of Cryptosporidium parvum in municipal water samples.

A PCR method for the quantitation of Cryptosporidium parvum oocysts in municipal drinking water samples was investigated. Quantitative PCR uses an internal standard (IS) template with unknown target numbers to compare to standards of known concentrations in a standard curve. The IS template was amplified using the same primers used to amplify a portion of a 358 bp gene fragment that encodes a repetitive oocyst wall protein in C. parvum. Municipal water samples spiked with known numbers of C. parvum oocysts were tested by quantitative PCR using the IS and the Digene SHARP Signal System Assay for PCR product detection. The absorbance readings for target DNA and IS templates versus the number of molecules of the target DNA were plotted to generate standard curves for estimating oocyst numbers. The method allowed the quantitation of oocysts from log 3 to log 5 spiked into municipal water samples.

M3/M1-Selective antimuscarinic tropinyl and piperidinyl esters.

The binding affinities of some tropinyl and piperidinyl esters for the submandibulary glands (M3/M1) and heart ventricle (M2) were determined from displacement experiments using 3H-labelled N-methylscopolamine as radioligand. The antimuscarinic activities of these esters were also evaluated on guinea pig bronchi. The esters inhibited the M3-mediated carbachol-induced contraction of the bronchial smooth muscle and a reasonable correlation was obtained between the binding affinities of the esters for the submandibulary glands (pKM3,M1) and their inhibitory activities (pIC50) on guinea pig bronchi. A promising compound, N-methylpiperidinyl cyclohexylphenylpropionate (NCPP) which combined good antimuscarinic activity (pA2=9.34) with a 20-fold selectivity at the M3/M1 receptors, was identified. Quantitative structure-activity relationships (QSAR) showed that the size of the ester was the main structural feature determining both binding affinity for the M3/M1 receptors and inhibitory activity on guinea pig bronchi. Esters with substituted acyl side chains (fewer hyperconjugable H atoms at the alpha-carbon) are generally associated with better activity and affinity.

The effect of a levonorgestrel-releasing intrauterine device on human endometrial oestrogen and progesterone receptors after one year of use.

Thirty-four women bearing a levonorgestrel-releasing intrauterine device, 20 micrograms/day (LNG-IUD-20), for 12-15 months were recruited. Endometrial biopsies were collected during the late proliferative phase of the cycle (on cycle days 10-12) before (control) and after the use of the IUD for 12 months, and assayed for oestrogen receptors (ER) and progesterone receptors (PR). An immunohistochemical technique with the peroxidase-antiperoxidase detection system (PAP method) was employed. D75 and JZB39 were the primary antibodies for ER and PR respectively. The immunostaining semiquantitative analysis was performed with a computerized microscope image processor, and expressed as 'grey value'. Both endometrial ER and PR populations were significantly lower after insertion of the IUD (P < 0.01) than in control biopsies. The intensity of nuclear staining and the percentage of positively stained cells for ER and PR in women with LNG-IUD were each about 50% of those in control biopsies. The results suggested that LNG released locally from the IUD has a depressive action on the ER and PR, which may contribute to the contraceptive effectiveness of this type of IUD and also to the possible causes of LNG-IUD-induced irregular bleeding and amenorrhoea.

Detection and characterisation of transforming growth factor-beta in porcine colostrum.

Porcine colostrum and milk collected at different stages of lactation were assessed for transforming growth factor-beta (TGF-beta) activity using an epithelial cell bio-assay. A high level of TGF-beta activity was recorded in all colostrum samples after transient acidification treatment, ranging between 126 and 260 ng/ml for samples collected at the time of parturition and 73 ng/ml for the sample collected 12 h after parturition. Without transient acidification treatment TGF-beta activity was detected only in 2 samples collected at the time of parturition (12 ng/ml) and 12 h after parturition (9 ng/ml), respectively. TGF-beta activity was undetectable in milk collected 5 days after parturition. These results suggest that TGF-beta exists mainly in latent form in porcine colostrum and the concentration declines rapidly as lactation proceeds. After enrichment with cation exchange chromatography a low level of TGF-beta activity was detected in porcine milk. Further separation by size exclusion chromatography revealed two molecular mass forms of TGF-beta in both colostrum and milk samples, a major peak of about 80 kD and a minor peak of about 25 kD representing latent and active forms of TGF-beta, respectively. A further experiment showed that the latent form of TGF-beta in colostrum can be activated at pH 3.5 or less. It is speculated that TGF-beta found in the colostrum may play a physiological role in regulating postnatal adaptation of the gastro-intestinal tract in newborns.

The influence of acupuncture on the impedance measured by four electrodes on meridians.

The impedance on the pericardium merdian near Quze (P3) and control points in 12 cases was measured by a four electrodes impedance instrument. The amplitude of impedance was recorded before, during and after needling the Neiguan. The result showed that the mean impedance on the meridian and control points before the needling were 52.8 +/- 11.0 (omega) and 61.7 +/- 10.3 (omega) respectively which had significant difference (P<0.05). During the needling, impedance decreased significantly on the meridian by 9.2 +/- 5.6 (omega) (P<0.001) while impedance decreased by only 0.12 +/- 2.4 (omega) on control points without significance (P>0.05). The impedance changed back to 51.1 +/- 11.3 (omega) and 59.9 +/- 11.0 (omega) on the meridian and control points respectively during the 5-10 minutes after withdrawing the needling. In some cases, impedance changed intermittently during the needling. The experiment implies that interstitial fluid increases during the needling by axon reflection and blood capillary expanding which may be one of the mechanisms of acupuncture regulation.

Magnetic resonance imaging evaluation of thecal sac diameters following laminectomy in the lumbar spine.

Sagittal magnetic resonance images (MRIs) from 20 patients who had postoperative persistent or recurrent low back pain were reviewed to study the thecal sac diameter at laminectomy sites. The thecal sac anteroposterior diameter was measured in three sites: the maximum AP diameter at the laminectomy site, and the proximal and distal levels to the laminectomy site. A comparison between the thecal sac AP diameter at laminectomy sites and the average diameter of both the proximal and distal levels was then made. Based on the measurements, the diameters of the thecal sac were divided into three groups. Patients in group A (5 cases) had a constriction (the thecal sac AP diameter at the laminectomy site was less than the average diameter of both the proximal and distal levels) of the thecal sac at the laminectomy site. Patients in group B (12 cases) had an expansion (the thecal sac AP diameter at the laminectomy site was more than the average diameter of both the proximal and distal levels) of the thecal sac. Patients in group C (3 cases) demonstrated no change. This study suggests that there was no correlation between the thecal sac AP diameter at laminectomy site and postoperative low back pain.

Anatomic considerations for anterior instrumentation of the lumbar spine.

One hundred seventy lumbar vertebrae from L1-L4 were used to quantitatively evaluate the lumbar vertebral body and examine the relationship of the maximum posterior angles of screw placement to the spinal canal. Anatomic evaluation included dimensions of the vertebral body. Three entrance points on the lateral aspect of the vertebral body for screw insertion and an additional point 3 mm from the posterolateral corner of the spinal canal were defined and marked. The maximum posterior screw angles were determined as the angles between the line connecting the entrance point with the additional point and the coronal plane. Results showed that vertebral body dimensions increased from L1-L4. The average vertebral body depth, width, and height were approximately 26 mm, 36 mm, and 22 mm at L1, and 30 mm, 44 mm, and 23 mm at L4, respectively. The spinal canal may be penetrated if the screws are directed posteriorly 2 degrees-5 degrees at L1 - L2 and 9 degrees - 14 degrees at L3-L4 starting at the junction between the pedicle and vertebral body, 22 degrees - 32 degrees at L1-L4 from the level of 10 mm anterior to the junction, and 43 degrees -50 degrees from the level of 20 mm anterior to the junction. Therefore, mid-body screws should be directed perpendicular to the lateral plane of the vertebral body. For a more anteriorly placed screw, slightly posterior angulation is recommended.

Computed tomographic evaluation of the internal structure of the lateral sacral mass in the upper sacra.

Because dimensions of the upper sacral cortexes vary greatly among individuals, preoperative computed tomographic (CT) evaluation of individual sacrum may help surgeons choose sacral screw insertion techniques. Axial CT scans were performed on 40 dry sacrum specimens to quantitatively evaluate the internal structure of the lateral sacral mass in the first and second segments. The results showed that the greatest cortical thickness in the S1 vertebra was found in the anterior cortex (3.4+/-0.9 mm), followed by the anterolateral (3.2+/-1.2 mm), and anteromedial (2.9+/-1 mm). The greatest cortical thickness in the S2 region was noted in the anteromedial cortex (2.4+/-0.5 mm), followed by the anterior and anterolateral (2.2+/-0.9 mm). The mean percentage of the anterior cortex thickness versus the lateral sacral mass depth was 12.8+/-3.7 for S1 and 11.1+/-2.8 for S2. Bicortical screw placement is recommended to achieve stronger fixation, but care should be taken not to violate the vital structures anterior to the sacrum.
