Unprecedented SnCl2-mediated cyclization of nitro arenes via N-N bond formation.

[reaction: see text] A mild, efficient, one-pot protocol for the cyclization of nitro-aryl substrates using SnCl(2) has been described. The mechanistic course of the reaction suggests the involvement of a hydroxylamine intermediate leading to an intramolecular cyclization via N-N bond formation. The versatility of the methodology has been demonstrated by using two nitro-aryl substrates derived from dihydroisoquinolines and dihydro-beta-carbolines. The intramolecular cyclization led to the formation of indazoles in high yields and purities.

Comparative structural analysis of 5,6,7,9-tetra-O-acetyl-4,8-anhydro-1,3-dideoxy-D-glycero-L-gluco-nonulose and its 1-O-acetylated analog, 1,2,3,4,6-penta-O-acetyl-beta-D-galactopyranose using X-ray crystallography.

Comparative X-ray diffraction analysis of 5,6,7,9-tetra-O-acetyl-4,8-anhydro-1,3-dideoxy-D-glycero-L-gluco-nonulose (1) and a structurally related analog, 1,2,3,4,6-penta-O-acetyl-beta-D-galactopyranose (2), are reported. Both crystals have one molecule in the unit cell and the pyranose rings in both exist in the 4C1 conformation.

Diastereoselective annulation of 4-hydroxypyran-2H-ones with enantiopure 2,3-dideoxy-alpha,beta-unsaturated sugar aldehydes derived from respective glycals.

One-pot condensations of 4-hydroxypyran-2H-ones 1 and 2, respectively, with various enantiopure 2,3-dideoxy-alpha,beta-unsaturated carbohydrate enals in the presence of l-proline in EtOAc at room temperature generated pyrano-pyrones. It was observed that, while benzyl-protected carbohydrate enals on condensation with 1 or 2 under the above conditions produced an inseparable diastereomeric mixture in a ratio of 1:1, the acyl-protected carbohydrate enals on treatment with 1 or 2 under identical conditions yielded products with moderate to very high diastereoselectivity. A remarkable asymmetric induction was noticed from the C-4 stereogenic center of the acyl-protected carbohydrate enals. An almost complete diastereoselectivity was observed in those reactions that involved condensation of 1 with acetyl-protected enals 5 and 7. The reaction of 2 with 5 also proceeded diastereoselectively to furnish the corresponding annulated product. The reaction presumably took place by C-1,2-addition of the pyrone onto the iminium salt of the alpha,beta-unsaturated carbohydrate enal generated in situ, followed by beta-elimination and cyclization of the 1-oxatriene involving a 6pi-electron electrocyclic process to yield a 2H,5H-pyrano[3,2-c]pyran-5-one derivative.

Comparison of ocular hemodynamics measured by a new retinal blood flowmeter and color Doppler imaging.

BACKGROUND AND OBJECTIVE: To compare measurements of a new retinal blood flow device with central retinal artery blood velocity. MATERIALS AND METHODS: One randomly selected eye from each of 13 subjects was examined. Blood flow was measured by laser blood flowmeter and velocity by color Doppler imaging. The correlation between measurements was analyzed using regression analysis where a P value of less than .05 and coefficient of regression values of greater than .5 were considered significant. RESULTS: The laser blood flowmeter produced vessel diameter measurements of 90.1 +/- 18.7 microm (mean +/- standard deviation), velocity of 19.7 +/- 8.06 mm/sec, and flow measurements of 4.24 +/- 2.41 microL/min. Central retinal artery peak systolic velocity, end diastolic velocity, and mean velocity correlated significantly with laser blood flowmeter velocity (P = .01, r = .66; P < .01, r = .77; and P = .003, r = .76, respectively) and flow (P = .01, r = .71; P = .03, r = .6; and P = .01, r = .69, respectively). CONCLUSIONS: The laser blood flowmeter produces retinal artery flow and velocity measurements that correlate with central retinal artery peak systolic velocity end diastolic velocity, and mean velocity measurements. Further validation of the laser blood flowmeter's accuracy in measuring real flow warranted and likely requires more invasive in vivo studies (in animal models). However, this study supports the ability of the laser blood flowmeter to measure retinal blood flow.
