Organically directed iron sulfate chains: structural diversity based on hydrogen bonding interactions.

Six organically directed 1-D iron sulfates hydrated and hydrolyzed to different extents have been prepared hydrothermally. [C2H10N2]1.5[Fe(SO4)(3)].2H2O (I), [C2H10N2][Fe(SO4)2(OH)].H2O (II), [C6H18N2]0.5[Fe(SO4)2(H2O)2] (III), and [C6H18N2]0.5[Fe2(SO4)(H2O)4(OH)].H2O (V) possess the linear topological structures observed in ferrinatrite, sideronatrite, kröhnkite, and copiapite minerals, respectively. [C4H12N2][Fe2(SO4)3(OH)2(H2O)2].H2O (IV) shows a novel linear structure that can be regarded as a hybrid of the tancoite and butlerite types. [C6N4H22]0.5[Fe(SO4)2(OH)].2H2O (VI) adopts a cis configuration, compared with II, to give a rare inorganic helical iron sulfate chain which is a new member of the organically directed transitional metal sulfates. The results reveal that the starting molar proportion of the reactants and the type of amines are critical for the structural motif. There is an obvious relationship between the constitution of chains and the type of amino groups, involving the amount of N-H...O hydrogen bonds.

EPS biofouling in membrane filtration: an analytic modeling study.

Biofouling is theoretically investigated by modeling solute transport within a biofilm, defined in this study as a swarm of solid biocolloids surrounded by liquid-like exopolymeric substances (EPS). A mathematical approach is employed to map the biofilm to an equivalent, simple spherical cell using a self-consistent method. It is found that the physical presence of EPS and their reaction with solute ions reduce the mass transfer coefficient, which significantly contributes to permeate flux decline in reverse osmosis and nanofiltration membrane processes.

Altered growth characteristics of skin fibroblasts from wild-derived mice, and genetic loci regulating fibroblast clone size.

Mouse fibroblast senescence in vitro is an important model for the study of aging at cellular level. However, common laboratory mouse strains may have lost some important allele variations related to aging processes. In this study, growth in vitro of tail skin fibroblasts (TSFs) derived from a wild-derived stock, Pohnpei (Pohn) mice, differed from growth of control C57BL/6 J (B6) TSFs. Pohn TSFs exhibited higher proliferative ability, fewer apoptotic cells, decreased expression of Cip1, smaller surface areas, fewer cells positive for senescence associated-beta-galactosidase (SA-beta-gal) and greater resistance to H(2)O(2)-induced SA-beta-gal staining and Cip1 expression. These data suggest that TSFs from Pohn mice resist cellular senescence-like changes. Using large clone ratio (LCR) as the phenotype, a quantitative trait locus (QTL) analysis in a Pohn/B6 backcross population found four QTLs for LCR: Fcs1 on Chr 3 at 55 CM; Fcs2 on Chr X at 50 CM; Fcs3 on Chr 4 at 51 CM and Fcs4 on Chr 10 at 25 CM. Together, these four QTLs explain 26.1% of the variations in LCRs in the N2 population. These are the first QTLs reported that regulate fibroblast growth. Glutathione S transferase mu (GST-mu) genes are overrepresented in the 95% confidence interval of Fcs1, and Pohn TSFs have higher H(2)O(2)-induced GST-mu 4, 5 and 7 mRNA levels than B6 TSFs. These enzymes may protect Pohn TSFs from oxidation.

[Magnetocardiography changes in coronary artery disease patients with normal or unspecialized resting electrocardiogram].

OBJECTIVE: To investigate the magnetocardiography (MCG) changes in coronary artery disease (CAD) patients with normal or unspecific changes in resting electrocardiogram (ECG). METHODS: MCG mapping was performed by MCG-7 (MaGIC, Magiscan GmbH) installed in an unshielded room. All patients underwent ECG and coronary angiogram examinations and patients with normal or unspecified ECG changes and coronary artery narrowing > or = 70% in at least 1-vessel were defined as CAD group (n = 120). Patients with normal coronary angiogram served as control (n = 82). Four parameters: ACTM (average classification of total maps), RAM (ratio of abnormal maps), CVEI (complex ventricular excitation index) and R-max/T-max ratio, were analyzed in CAD and control groups. RESULTS: RAM (62% vs. 35%) and ACTM (2.62 +/- 0.98 vs. 2.29 +/- 0.90, P < 0.05) were significantly higher in CAD group than in control group. CVEI was found in abnormal zone (-100 - 0) in CAD group while in normal zone (0 - 100) in control group. The ratio of Rmax/Tmax in CAD group was also significantly higher in CAD group than in control group (6.41 +/- 3.29 vs. 4.10 +/- 2.00, P < 0.01). ROC curve analysis indicates that RAM, CVEI and Rmax/Tmax ratio were helpful parameters for CAD diagnosis and the diagnostic sensitivity was 67.1%, 65.9% and 64.3%; the specificity was 65.1%, 68.3% and 68.3% respectively. CONCLUSION: MCG was a useful tool for diagnosing chronic myocardium ischemia in CAD patients with normal or unspecific changes resting ECG.

Hydrodynamics of an ideal aggregate with quadratically increasing permeability.

In this study, we consider the ideal aggregate with quadratically increasing permeability kappa = k2r2 and derive the analytical expression of the stream function within the porous aggregate by incorporating the Brinkman and continuity equations. The hydrodynamic properties of the aggregate are investigated by taking account of the hydrodynamic radius, settling velocity, and fluid collection efficiency, which are found to be solely dependent on the permeability prefactor k2. The fractal dimension Df and prefactor k2 of the ideal aggregate are found to be 5/3 (=1.67) and 0.20, respectively, and well describe the hydrodynamics of aggregates formed in the diffusion-limited-cluster-aggregation (DLCA) regime. More important, hydrodynamic similarity between the ideal aggregate and impermeable solid sphere is discovered in terms of variations of the hydrodynamic radius, settling velocity, and fluid collection efficiency with respect to the aggregate radius.

Genetic regulation of hematopoietic stem cell exhaustion during development and growth.

OBJECTIVE: During aging, hematopoietic stem cell (HSC) exhaustion is more severe in BALB/cByJ (BALB) mice than in C57BL/6J (B6) mice. Our objective is to determine whether HSC exhaustion during development from fetus to adult also is more severe for BALB than for B6 mice. MATERIALS AND METHODS: Hematopoietic stem cells from fetal liver cells (FLCs) and from young adult bone marrow cells (BMCs) were compared using the competitive repopulation assay to measure long-term repopulating ability (LTRA) and HSC expansion after serial transplantation. LTRAs were measured in repopulating units (RU), as the ability to produce donor-type erythrocytes and lymphocytes in lethally irradiated recipients relative to the congenic fresh marrow competitor. To test expansion, FLCs or BMCs were serially transplanted into lethally irradiated carriers whose marrow cells were compared using fluorescence-activated cell staining (FACS), and subsequently tested for LTRA. RESULTS: BALB and B6 FLCs, respectively, repopulated 2.6 and 13.5 times as well as BMCs. LTRAs correlated with HSC expansion for BALB, but not B6. Per million donor cells, CD34(-) HSC-enriched fractions (HEFs) and total RU values were 6.8 and 4.6 times higher for FLCs than for BMCs in BALB carriers, while these ratios were only 1.2 and 0.97 higher in B6 carriers. CONCLUSION: In B6 HSC development, LTRA is dissociated from expansion. Although 1 x 10(6) BMCs have much lower LTRA, they expand HSCs as well as 1 x 10(6) FLCs. HSC expansion is partly exhausted in BALB, but not B6, during development.

Hammerhead ribozyme targeting connective tissue growth factor mRNA blocks transforming growth factor-beta mediated cell proliferation.

PURPOSE: Excessive scarring following trauma or surgery of cornea, conjunctiva or retina can greatly impair visual outcome. At present, no agents are clinically available that selectively reduce activity of genes that regulate fibrosis. Connective tissue growth factor (CTGF) has been linked to fibrosis in several tissues, including cornea and conjunctiva. In this study, hammerhead ribozymes targeting CTGF mRNA were synthesized, kinetic parameters were measured, and the effect on TGF-beta-mediated cell proliferation was measured in cultured human fibroblasts. METHODS: The mRNA sequence of human CTGF was scanned for potential hammerhead ribozyme cleavage sites, and predicted secondary folding structures around the sites were calculated. Synthetic 12mer ribozymes and 33mer oligonucleotide mRNA targets corresponding to two sites were synthesized, and kinetic constants calculated from Hanes-Wolff plots of in vitro cleavage reactions. The ribozyme with higher percentage cleavage and kinetic rate was cloned into an expression plasmid (pTR-UF21) and stably transfected into cultured human fibroblasts. An inactive ribozyme plasmid served as a negative control. The effects of the ribozyme on expression of TGF-beta-induced CTGF mRNA and protein levels were measured using ELISA and real-time TaqMan quantitative RT-PCR. Finally, the effect of the CTGF ribozyme on TGF-beta-mediated proliferation of fibroblasts was measured using a non-radioactive cell proliferation microtiter assay. RESULTS: Of the eight potential hammerhead ribozyme cleavage sites in human CTGF mRNA, two sites (CHR 745, and CHR 859) were identified with optimal secondary folding. CHR 859 cleaved 94% of the target mRNA, compared to 46% cleavage for CHR 745 after 16 hr of reaction. CHR 859 had a K(m) of 1.56 microM and a K(cat) of 2.97 min(-1), while CHR 745 had a K(m) of 7.80 microM and a K(cat) of 5.7 min(-1). The turnover numbers (K(cat)/K(m)) of CHR 859 and CHR 745 were 1.9 x 10(6) M min(-1) and 7.4 x 10(5) M min(-1), respectively, indicating CHR 859 is 2.6 times more efficient than CHR 745 in destroying CTGF mRNA. Stable transfection of CHR 859 into human fibroblasts reduced CTGF mRNA levels 55% and protein levels 72% compared to the inactive ribozyme control. Furthermore, TGF-beta-induced cell proliferation was reduced 90% in fibroblasts stably transfected with CHR 859 compared to control cell groups. CONCLUSIONS: The CHR 859 hammerhead ribozyme cleaved human CTGF mRNA with high kinetic efficiency in vitro, effectively reduced levels of CTGF mRNA and protein in cultured human fibroblasts, and blocked TGF-beta-induced cell proliferation without nonspecific toxicity. These data support the concept that CTGF mediates TGF-beta-induced cell proliferation, and imply that regulating CTGF expression with ribozymes may be effective in reducing ocular scarring.

Molecular mechanisms of irradiation-induced apoptosis.

The following review focuses on our current knowledge as to how the cell death regulatory machinery is activated to mediate irradiation-induced cell death. In particular, we will address recent developments related to the following questions: 1.) Which cell death regulatory genes mediate irradiation-induced cell death? 2.) What is the mechanism of irradiation-induced activation or suppression of cell death regulatory genes (proteins)? 3.) How does the condition of the cell death regulatory machinery affect the cell's sensitivity or resistance to irradiation? Now more than ever, it seems clear that irradiation -induced apoptosis is a complex process involving all three major cell death regulatory pathways: the mitochondria pathway (Bcl-2/Apaf-1), the Iap pathway, and the death receptor pathway. Depending on the cellular context, one or multiple pathways may be activated to mediate irradiation-induced cell death. Therefore, a comprehensive understanding of these processes demands systematic strategies in contrast to traditional approaches that focused on one gene/protein. For this reason, we will also examine recent studies applying genomic (proteomic) methods in this area.

[IR absorption spectral characteristics of chicken bile].

It has been proved by medicinal practice that the extraction from chicken bile has excellent antiphlogistic and anti-tumour effects. Chicken bile and the biles of various animals were dried and after IR spectral analyses, different IR spectra can be obtained with the resulting powders. In comparison with IR spectra, the IR spectrum from chicken bile is significantly different with that of other animals; whereas there are very good consistency among the chicken biles from different origin samples. The analyses of chicken bile show the chemical composition of chicken bile is different from that of other animals and the biles of different chicken have essentially same chemical composition.
