CD1d restricted natural killer T cells are not required for allergic skin inflammation.

BACKGROUND: Invariant T-cell receptor-positive natural killer (iNKT) cells have been shown to be essential for the development of allergen-induced airway hyperreactivity (AHR). OBJECTIVE: We examined the role of iNKT cells in allergic skin inflammation using a murine model of atopic dermatitis (AD) elicited by epicutaneous sensitization with ovalbumin (OVA). METHODS: Wild-type (WT) and natural killer T-cell-deficient CD1d-/- mice were epicutaneously sensitized with OVA or normal saline and challenged with aerosolized OVA. iNKT cells in skin and bronchoalveolar lavage fluid were analyzed by fluorescence-activated cell sorting, and cytokine mRNA levels were measured by quantitative RT-PCR. AHR to methacholine was measured after OVA inhalation. RESULTS: Skin infiltration by eosinophils and CD4+ cells and expression of mRNA encoding IL-4 and IL-13 in OVA-sensitized skin were similar in WT and CD1d-/- mice. No significant increase in iNKT cells was detectable in epicutaneously sensitized skin. In contrast, iNKT cells were found in the bronchoalveolar lavage fluid from OVA-challenged epicutaneously sensitized WT mice, but not CD1d-/- mice. Epicutaneously sensitized CD1d-/- mice had an impaired expression of IL-4, IL-5, and IL-13 mRNA in the lung and failed to develop AHR in response to airway challenge with OVA. CONCLUSION: These results demonstrate that iNKT cells are not required for allergic skin inflammation in a murine model of AD, in contrast with airway inflammation, in which iNKT cells are essential. CLINICAL IMPLICATIONS: Understanding the potential role of iNKT cells in AD will allow us to have a more specific target for therapeutic use.

Morphometric sum optical density as a surrogate marker for ploidy status in prostate cancer: an analysis in 180 biopsies using logistic regression and binary recursive partitioning.

We sought to identify morphometric descriptors predictive of nondiploidy in prostatic adenocarcinoma on prostate needle biopsies using logistic regression (LR) and binary recursive partitioning (BRP) and compare the equivalence of both methods. A total of 180 prostate needle biopsies diagnosed as prostatic adenocarcinoma were selected. Deoxyribonucleic acid ploidy and morphometry were performed separately on Feulgen-stained sections from these biopsies using the CAS-200 system and Nuclear Morphometry Suite, respectively. Seven morphometric predictors were tested as predictor variables, including nuclear area, circularity, elongation, sum optical density (OD), configuration run length, coefficient of variation (CV), and angularity. Logistic regression (LR) identified a two-parameter model including sum OD and circularity that had a 93.9% overall correct prediction rate (area under curve=0.950; 95% CI: 0.913, 0.987). A reduced model including only sum OD was equally good without any significant loss of predictive accuracy (93.3% correct overall classification rate). BRP also selected sum OD as the most predictive parameter; a sum OD cut-off of 7.73 in this model identified 93.3% of the nondiploid cases correctly. Morphometric OD can be used as a surrogate marker of nondiploidy. LR and BRP models are both equivalent in identifying and correctly classifying nondiploid cases of prostate cancer using sum OD as the predictor variable.

The complement component C3 plays a critical role in both Th1 and Th2 responses to antigen.

BACKGROUND: Complement component C3 is synthesized by keratinocytes and is activated after skin injury. C3 is also synthesized by peritoneal macrophages, which are activated by the adjuvant alum. OBJECTIVE: We sought to investigate the role of C3 in inciting allergic skin Inflammation and systemic immune responses after epicutaneous sensitization or intraperitoneal sensitization with antigen. METHODS: C3-deficient (C3-/-) mice and wild-type (WT) control animals were subjected to epicutaneous sensitization with the antigen ovalbumin (OVA) on shaved and tape-stripped skin or intraperitoneal immunization with OVA in alum. RESULTS: Skin Infiltration by eosinophils and expression of mRNA encoding the TH2 cytokines IL-4 and IL-5 in OVA-sensitized skin sites was impaired in C3-/- mice. Splenocytes from epicutaneously sensitized C3-/- mice secreted less IL-4, IL-5, IL-13, and IFN-gamma in response to OVA stimulation than splenocytes from WT control animals. The defect in cytokine secretion by splenocytes was also observed after intraperitoneal immunization of C3-/- mice. C3-/- mice had impaired IgG1, IgG2a, and IgE antibody responses after both epicutaneous and intraperitoneal immunization. The defect in cytokine secretion of C3-/- mice was not due to defective proliferation to antigen, was not observed after anti-CD3 stimulation, and was corrected by the addition of purified C3 protein. CONCLUSION: These results suggest that C3 plays an important role in both the TH1 and TH2 response to antigen in vivo. CLINICAL IMPLICATIONS: The complement pathway might be a potential target in the therapy of allergic diseases.

Dynamic diffuse double-layer model for the electrochemistry of nanometer-sized electrodes.

A dynamic diffuse double-layer model is developed for describing the electrode/electrolyte interface bearing a redox reaction. It overcomes the dilemma of the traditional voltammetric theories based on the depletion layer and Frumkin's model for double-layer effects in predicating the voltammetric behavior of nanometer-sized electrodes. Starting from the Nernst-Planck equation, a dynamic interfacial concentration distribution is derived, which has a similar form to the Boltzmann distribution equation but contains the influence of current density. Incorporation of the dynamic concentration distribution into the Poisson and Butler-Volmer equations, respectively, produces a dynamic potential distribution equation containing the influence of current and a voltammetric equation containing the double-layer effects. Computation based on these two equations gives both the interfacial structure (potential and concentration profiles) and voltammetric behavior. The results show that the electrochemical interface at electrodes of nanometer scales is more like an electric-double-layer, whereas the interface at electrodes larger than 100 nm can be treated as a concentration depletion layer. The double-layer nature of the electrode/electrolyte interface of nanometer scale causes the voltammetric responses to vary with electrode size, reactant charge, the value of formal redox potential, and the dielectric properties of the compact double-layer. These voltammetric features are novel in comparison to the traditional voltammetric theory based on the transport of redox molecules in the depletion layer.

Delayed changes in T1-weighted signal intensity in a rat model of 15-minute transient focal ischemia studied by magnetic resonance imaging/spectroscopy and synchrotron radiation X-ray fluorescence.

Previous studies have found that rats subjected to 15-min transient middle cerebral artery occlusion (MCAO) show neurodegeneration in the dorsolateral striatum only, and the resulting striatal lesion is associated with increased T1-weighted (T1W) signal intensity (SI) and decreased T2-weighted (T2W) SI at 2-8 weeks after the initial ischemia. It has been shown that the delayed increase in T1W SI in the ischemic region is associated with deposition of paramagnetic manganese ions. However, it has been suggested that other mechanisms, such as tissue calcification and lipid accumulation, also contribute to the relaxation time changes. To clarify this issue, we measured changes in relaxation times, lipid accumulation, and elemental distributions in the brain of rats subjected to 15-min MCAO using MRI, in vivo 1H MR spectroscopy (MRS), and synchrotron radiation X-ray fluorescence (SRXRF). The results show that a delayed (2 weeks after ischemia) increase in T1W SI in the ischemic striatum is associated with significant increases in manganese, calcium, and iron, but without evident accumulation of MRS-visible lipids or hydroxyapatite precipitation. It was also found that 15-min MCAO results in acutely reduced N-acetylaspartate (NAA)/creatine (Cr) ratio in the ipsilateral striatum, which recovers to the control level at 2 weeks after ischemia.

Bone marrow stromal cells transduced with human hemangiopoietin gene support hematopoiesis in vitro.

BACKGROUND AND OBJECTIVES: The aim of this study was to construct a eukaryotic expression vector containing human hemangiopoietin (hHAPO) gene and express it in mouse bone marrow stromal cell line HESS-5, then support hematopoiesis in vitro with gene-modified HESS-5 (hHAPO-HESS-5). DESIGN AND METHODS: The polymerase chain reaction (PCR) products of HAPO were digested with BamHI and BgII. Then the HAPO gene segment obtained was again cloned into pIRES2-EGFP to construct recombinant eukaryotic expression vector HAPO-pIRES2-EGFP. The recombinant vector was identified by enzyme digestion analysis, PCR, and sequencing. HESS-5 cells were transformed by recombinant vector and positive clones were selected with G418. The expression of HAPO gene in the transformed cells was detected by studying EGFP expression, reverse transcription (RT)-PCR, and Western-blotting analysis. Support of human hematopoiesis by hHAPO-HESS-5 cells was evaluated in co-culture experiments with human CD34+ cells. RESULTS: Enzyme digestion analysis and sequencing showed that the target gene had been cloned into the recombinant vector. The expression of HAPO gene in the transformed stromal cells was demonstrated by fluoro-microscopy and RT-PCR analysis. HAPO protein was also detected in the supernatant of hHAPO-HESS-5 by Western blot analysis. As expected, stably transfected hHAPO-HESS-5 cells significantly increased in both relative and absolute numbers of CD34+ cells after 14 days of culture. The PKH26 study demonstrated that cell division was faster in CD34+ cells co-cultured with hHAPO-HESS-5 cells than in cells cocultured with vector-HESS-5 cells. The hHAPO-HESS-5 cells also supported human hematopoiesis in vitro more efficiently than did control vector-HESS-5 cells. INTERPRETATION AND CONCLUSIONS: A recombinant eukaryotic expression vector has been constructed and expressed successfully in transformed cells. The hHAPO-HESS-5 cells support rapid generation of primitive progenitor cells and maintain reconstituting ability of hematopoietic stem cells in vitro. Therefore, it would be possible to use stromal cells expressing HAPO gene as seed cells in the bone marrow transplantation.

[Protective effects of total flavonoids of Broussonetia papyrifera on oxidative injury of ultraviolet A to human keratinocytes].

OBJECTIVE: To investigate the oxidative damage of ultraviolet A (UVA) to human immortalized keratinocytes line HaCaT and the protective effects of total flavonoids of Broussonetia papyrifera (TFBP) gotten from the leaves of broussonetia papyifera. METHODS: Based on the culture of the human keratinocytes, the experiment group added with different dosages of TFBP before exposure to the radiation, received the UVA radiation together with the treatment group. The levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were determined in cultured HaCaT cells as well as the cell activity with MTT reduction assay. Human immortalized keratinocytes HaCaT cells received ultraviolet A with the different dosages between 0.46 and 2.76 J/cm(2) respectively. The protective effects of TFBP at different concentrations were also evaluated. RESULTS: The cell activity decreased gradually from 96.3% to 37.5% with the increase of UVA dosage from 0.46 J/cm(2) to 2.76 J/cm(2). After 10 mg/L up to 200 mg/L of TFBP were added the cell activity increased, the levels of MDA decreased from (5.14 +/- 0.58) nmol/mg pro to (2.98 +/- 0.14) nmol/mg pro, the levels of SOD increased from (23.09 +/- 3.91) U/mg pro to (34.50 +/- 1.59) U/mg pro and the activity of GSH-Px increased somewhat. CONCLUSION: Ultraviolet A causes significant oxidative injury to HaCaT cells under the conditions of this study. TFBP gotten from the leaves of broussonetia papyrifera has certain protective effect on HaCaT epithelial cells.

Inhibition of K562 leukemia angiogenesis and growth by expression of antisense vascular endothelial growth factor (VEGF) sequence.

Vascular endothelial growth factor (VEGF), a major angiogenic factor, plays a key role in the growth of solid tumor. Recently, expression of VEGF and its receptors has been found on leukemic cells as well as on endothelial cells. VEGF may fulfill a fundamental role in promoting tumor angiogenesis and proliferation by stimulating both endothelial cells and leukemic cells. To investigate the role of VEGF in the angiogenesis and growth of leukemic cell, we used an antisense strategy to downregulate VEGF expression in K562 cells, a human erythroleukemia cell line. Expression of antisense-VEGF in K562 cells reduced the secretion of VEGF protein and inhibited cell survival. The proliferation and migration of human umbilical vein endothelial cells were decreased in response to the conditioned medium (CM) from K562 cells expressed antisense-VEGF, compared to CM from K562 cells transfected with vector control. Moreover, subcutaneous injection of nude mice with antisense-VEGF K562 cells inhibited tumor growth with a reduction of the density of microvessels and an increased apoptosis in those tumors, compared to vector control K562 cells. These results suggest that the efficient downregulation of the VEGF production in leukemic cells using antisense-VEGF may constitute a novel strategy of treatment in leukemia.

Comparison of NIDEK CATz wavefront-guided LASIK to traditional LASIK with the NIDEK CXII excimer laser in myopia.

PURPOSE: To compare the visual and refractive outcomes in myopic LASIK between wavefront-guided and traditional treatment. METHODS: The study included 162 eyes of 81 myopic patients undergoing LASIK by one surgeon. The right eye of each patient underwent customized LASIK with the NIDEK NAVEX laser system (NIDEK, Gamagori, Japan) (study group); the left eye of each patient underwent traditional LASIK with the NIDEK CXII excimer laser system (control group). RESULTS: No significant differences were noted in refractive error, uncorrected visual acuity, and best spectacle-corrected visual acuity after surgery between the two groups (t test, P>.05). Postoperatively, 3.7% of patients in the study group reported glare, which was significantly lower than 12.4% in the control group (chi-square test, .01<P<.05). Higher order aberrations increased after surgery in both groups, but less increase was noted in patients in the study group. A statistically significant reduction was noted in the postoperative increase of Zernike coefficients C7, C8, and C12 in the study group compared to the control group (t test, P<.05). CONCLUSIONS: Customized LASIK significantly reduces the rate of postoperative glare and induction of higher order aberrations compared to traditional LASIK.
