[Studies on the chemical constituents from Ampelopsis grossedentata].

OBJECTIVE: To study the chemical constituents from Ampelopsis grossedentata (hand-Mazz) W.T. Wang. METHODS: Chromatography and spectroscopic analysis were employed to isolate and elucidate the chemical constituents in the plant. RESULTS: Seven compounds were isolated and elucidated as ampelopsin (I), myricetin (II), myricitrin (III), gallic acid (IV), beta-sitosterol (V), stigmasterol (VI) and dihydroquercetin (VII). CONCLUSION: Among these compounds, IV, V, VI and VII were isolated from this plant for the first time, and compound VII was obtained from genus Ampelopsis for the first time.

[Determination of ampelopsin in the different parts of Ampelopsis grossedentata in different seasons by RP-HPLC].

OBJECTIVE: To determine ampelopsin in the root, stem and leaf of Ampelopsis grossedentata in different seasons by RP-HPLC. METHODS: The determination was carried out with ODS column (5 microns, 250 mm x 46 mm), using methanol-water-phosphoric acid(25:75:0.1) as the mobile phase at a flow rate of 1.0 ml/min and detected at the wavelength 289 nm. RESULTS: Ampelopsin content was in the order of leaf > stem > root. Ampelopsin content in Ampelopsis grossedentata collected in April and May was the highest.

[A study on the percutaneous penetration of artesunate from various reservoir vehicles].

OBJECTIVE: To determine the main factors which affect the percutaneous penetration of artesunate and provide efficient data for the artesunate transdermal delivery system. METHOD: Transdermal speed constant and accumulative amount of 12 hours were used for the estimations of various reservior vehicles, and the supplement orthodox design was used to study the effect of pH, various proportion of IPA/Water/IPM, and drug concentration. RESULT: Drug concentration and pH were the main factors which affected the percutaneous penetration of artesunate. CONCLUSION: The suitable reservior vehicle can prompt the percutaneous penetration of artesunate, and artesunate TTS will be made with further studies.

Cost-effectiveness of public sector substance abuse treatment: comparison of a managed care approach to a traditional public sector system.

Costs and cost-effectiveness of public sector substance abuse treatment in 2 California counties with similar substance abuse treatment system histories are compared; one county (MidState) has adopted managed care principles. As hypothesized, MidState's costs for the index treatment episode were significantly lower than SouthState's, although unexpectedly because of lower outpatient utilization. Treatment benefits in the 7 Addiction Severity Index functional areas were examined through cost-effectiveness analyses. MidState can claim greater cost-effectiveness for its treatment dollars for significant improvement in alcohol and medical functioning (compared to unsuccessful clients and those reporting no problems). When comparing both improved clients and those maintaining no problems to unsuccessful clients, MidState is more cost-effective for improving alcohol, medical, legal, and family/social functioning; and 3 outcomes important to community stakeholders and taxpayers (legal, medical, and psychiatric functioning) are more cost-effective than alcohol, drug, and employment improvement.

Evaluation of a preclinical model of bone metastasis for the study of adoptive immunotherapy.

The development of new therapeutic strategies for the treatment of bone metastases strongly depends on the availability of valid animal models. In this paper, we evaluate a preclinical model of bone metastases using a technique of tumor cell injection into the left heart ventricle of mice to study the efficacy of adoptive immunotherapy. Using flow cytometric analysis and histopathological and radiological examination, we investigated whether this experimental model of bony metastases using two murine cell lines of melanoma and breast cancer would be suitable for the study of adoptive immunotherapy for these diseases. We further report that anti-CD3-activated and IL-2-expanded tumor vaccine draining lymph node cells cause regression of tumor metastases, including bone metastases, following adoptive transfer to mice bearing 3-day metastases from the D5 melanoma cell line. These promising early results lead us to conclude the following: (1). this model of experimental bone metastases is suitable for the study of immunotherapy, and (2). further studies are warranted to extend these promising early findings of the therapeutic effects of adoptive immunotherapy in this animal model.

Gallium maltolate is a promising chemotherapeutic agent for the treatment of hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is a particularly lethal cancer with few treatment options. Since gallium is known to accumulate specifically in HCC tumors but not in non-tumor liver, we investigated two gallium compounds, gallium nitrate (GaN) and gallium maltolate (GaM), as potential new agents for treating HCC. MATERIALS AND METHODS: The anti-proliferative and apoptotic activities of GaN and GaM were assessed in vitro using four HCC cell lines. HCC gene expression data was analyzed to provide a mechanistic rationale for using gallium in the treatment of HCC. RESULTS: Both compounds showed dose-dependent antiproliferative activity in all four HCC cell lines after 6-day drug exposure (IC50 values range from 60-250 microM for gallium nitrate and 25-35 microM for gallium maltolate). Gallium maltolate at 30 microM additionally induced apoptosis after 6 days. HCC gene expression data showed significantly elevated expression of the M2 subunit of ribonucleotide reductase, which is a target for the antiproliferative activity of gallium. CONCLUSION: These data support clinical testing of gallium maltolate, an orally active compound, in the treatment of HCC.
