A genetic mouse model carrying the nonfunctional xeroderma pigmentosum group G gene.

A genetic mouse model with a disrupted XPG allele was generated by insertion of neo cassette sequences into exon 3 of the XPG gene by using embryonic stem (ES) cell techniques. The xpg-deficient mice showed distinct developmental characteristics. Their body was marked smaller than that in wild-type littermates since the postnatal day 6, and this postnatal growth failure became more severe with developmental proceeding. Their life span was very short, all of the mutants died by postnatal day 23 after showing great weakness and emaciation. In addition, the mutant homozygous mice also showed some progressive neurological signs, like the lower level of activity and a progressive ataxia. Further examination indicated there was developmental retardation of the brain in the mutant mice. Their brain weight, and thickness of cerebral cortex and cerebellar cortex were significant different from the controls. These characteristics, like small size brain, brain developmental retardation and progressive neurological dysfunctions in the homozygotes were similar to the typical clinical phenotype of the XPG patients and Cockayne syndrome, we believe that the xpgdeficient mice will be an animal model for studying the function of the XP-G protein in nucleotide-excision repair and mechanisms related to the clinic symptoms of XP-G and Cockayne syndrome in humans.

[Development of the human/rat chimera model with neonatal rats].

The purpose of this study was to transplant neonatal rat with human cord blood Lin(-) cells to test the possibility of this xenograft model. The Lin(-) cells were purified from human cord blood (CB) using negative selection strategy based on different lineage-specific antigens. The Lin(-) cells were injected into the liver of neonatal rats using a microinjector at an average of 5 x 10(5) cells for each. Peripheral blood (PB) and spleen were collected at 2,4 and 8 weeks after injection. Flow cytometry was performed to detect human cells in the rat PB, PCR was used to detect human cells in PB as well as spleen. The results showed that a definite proportion of human cells existed in peripheral blood of chimeric rat and the human specific beta2 microglobulin gene fragments were detected in spleen genomic DNA of chimeric rat. It is concluded that human/rat chimera model can be developed with neonatal rats. Human/rat xenograft model may provide a useful and convenient method for human hematopoietic stem cell assay in vivo.

[Investigation of the status of ophthalmology in China].

OBJECTIVE: To identify the quantity, constitution, distribution, human resources, medical service, equipment utilization conditions of our ophthalmic organizations. METHODS: Adopt the ophthalmology status survey forms developed by the Department of Health Administration of Ministry of Health and Office of National Committee for Prevention of Blindness to investigate the registered medical treatment organizations in the whole mainland of China in 1996. The collected data were analyzed using SAS software. RESULTS: There were 4,151 ophthalmological organizations, 43,204 beds for ophthalmic patients, 22,577 eye doctors, and 16,448 ophthalmic nurses in China in 1996. 41.55% of eye doctors worked in the general hospitals above the county level. CONCLUSION: The distribution of ophthalmic organizations and human resource is uneven. It may be the main cause to restrict the activities for prevention of blindness in China.

Expression of neural cell adhesion molecule L1 in the brain of rats exposed to X-irradiation in utero.

To gain insight to the cellular and molecular mechanisms involved abnormal neuronal migration induced by irradiation, we investigated expression of neuronal cell adhesion molecule L1 and neuronal migration in the brains through comparison between rats prenatally exposed to X-ray and controls. To observe the pattern of neuronal migration, bromodeoxyuridine (BrdU) was chosen as a marker to label migrating cells. The results showed some of the labeled cells remained in the lower of the cortical plate in the irradiated rats, suggesting that neuronal migration was disrupted by X-ray. To study change of expressing neural cell molecule L1, rat brains were analyzed by SDS-PAGE after isolation of L1 by immunoaffinity chromatography. In the all brain membrane fraction, immunoaffinity purified L1 had bands at 200, 180, 140 and 80 kDa. However, the bands in the irradiated group were very weak when compared with the control. Taking these results into account, abnormal neuronal migration and reduction of expression L1 found in the irradiated brain indicated that migration of neural cells may be largely dependent on radial glial fiber as well as neural cell molecules like L1. A decrease in L1 expression may be one of reasons of abnormal neuronal migration.

[Clinical analysis of pregnancy in second and third trimesters complicated severe acute respiratory syndrome].

OBJECTIVE: To explore and analyze the clinical manifestation, diagnosis, management and maternal-perinatal prognosis of severe acute respiratory syndrome (SARS)-complicated pregnancy in second and third trimesters. METHODS: Clinical data of 5 inpatients with SARS-complicated pregnancy in second and third trimesters from 4(th) February to 17(th) March 2003 were analyzed retrospectively. RESULTS: Five patients were all primigravida (including 2 twins). Two were infected in second trimester while the other 3 in third trimester with 2 hospital-acquired infections and 3 community-acquired infections. All patients had fever (5/5), 3 chills or rigor, 4 cough; 2 with decreased lymphocyte, 2 decreased platelet, 3 elevated alanine aminotransferase (ALT), 4 hypoalbuminemia, 5 abnormal chest radiographs. All 5 patients were cured with 1 requiring intensive care. 5 neonates including 1 twins have been followed up without evidence of SARS infections up to now. In a twin-pregnancy 1 fetus was lost while the pregnant's situation is stable. CONCLUSIONS: Common diagnostic criteria were Suitable for SARS-complicated pregnancy in second and third trimesters, but attention should be paid to the interaction between SARS and special pathological changes during pregnancy. Patients should be isolated and monitored intensively with timely cesarean section in severe cases, which could significantly decrease the maternal-perinatal mortality. The use of corticosteroids and psychological supports need further study.

[Effect of occupational stress on mental health].

OBJECTIVE: To study the effect of job psychological demands and job control on mental health and their interaction. METHODS: 93 male freight train dispatchers were evaluated by using revised Job Demand-Control Scale and 7 strain scales. Stepwise regression analysis, Univariate ANOVA, Kruskal-Wallis H and Modian methods were used in statistic analysis. RESULTS: Kruskal-Wallis H and Modian methods analysis revealed the difference in mental health scores among groups of decision latitude (mean rank 55.57, 47.95, 48.42, 33.50, P < 0.05), the differences in scores of mental health (37.45, 40.01, 58.35), job satisfaction (53.18, 46.91, 32.43), daily life strains (33.00, 44.96, 56.12) and depression (36.45, 42.25, 53.61) among groups of job time demands (P < 0.05) were all statistically significant. ANOVA showed that job time demands and decision latitude had interaction effects on physical complains (R(2) = 0.24), state-anxiety (R(2) = 0.26), and daytime fatigue (R(2) = 0.28) (P < 0.05). Regression analysis revealed a significant job time demands and job decision latitude interaction effect as well as significant main effects of the some independent variables on different job strains (R(2) > 0.05). CONCLUSION: Job time demands and job decision latitude have direct and interactive effects on psychosomatic health, the more time demands, the more psychological strains, the effect of job time demands is greater than that of job decision latitude.

Independent control of ion transmission in a jet disrupter dual-channel ion funnel electrospray ionization MS interface.

A new atmospheric pressure ionization mass spectrometer (API-MS) interface has been developed to allow the control of ion transmission through the first vacuum stage of the mass spectrometer. The described interface uses a dual-heated capillary and a dual-inlet ion funnel design. Two electrosprays, aligned with the dual-capillary inlet, are used to introduce ions from different solutions independently into the MS. The initial design was specifically aimed at developing a method for the controlled introduction of calibrant ions in highly accurate mass measurements using Fourier transform ion cyclotron resonance mass spectrometer (FTICR). The dual-channel ion funnel has different inlet diameters that are aligned with the dual capillaries. The large diameter main channel of the ion funnel is used for analyte introduction to provide optimum ion transmission. The second, smaller diameter channel inlet includes a jet disrupter in the ion funnel to modulate the ion transmission through the channel. The two inlet channels converge into a single-channel ion funnel where ions from both channels are mixed, focused, and transmitted to the mass analyzer. Both theoretical simulations and experimental results show that the transmission of different m/z species in the small diameter channel of the ion funnel can be effectively modulated by varying the bias voltage on the jet disrupter. Both static and dynamic modulations of ion transmission are demonstrated experimentally by applying either a constant DC or a square waveform voltage to the jet disrupter. High ion transmission efficiency, similar to the standard single-channel ion funnel, is maintained in the main analyte channel inlet of the ion funnel over a broad m/z range with negligible "cross talk" between the two ion funnel inlet channels. Several possible applications of the new interface (e.g., for high-accuracy MS analysis of complex biological samples) are described.

Stereo- and enantioselective alkene epoxidations: a comparative study of D4- and D2-symmetric homochiral trans-dioxoruthenium(VI) porphyrins.

The mechanism of stoichiometric enantioselective alkene epoxidations by the D4- and D2-symmetric homochiral trans-dioxoruthenium(VI) porphyrins, [RuVI(D4-Por*)O2] (1) and [RuVI(D2-Por*)O2] (2a), in the presence of pyrazole (Hpz) was studied by UV/Vis spectrophotometry and analysis of the organic products. The enantioselectivity of styrene oxidations is more susceptible to steric effects than to substituent electronic effects. Up to 72% ee was achieved for epoxidation of 3-substituted and cis-disubstituted styrenes by employing 1 as the oxidant, whereas entantioselectivities of only 20-40% were obtained in the reactions with 2-substituted and trans-disubstituted styrenes. Complex 2a oxidized 2-substituted styrenes to their epoxides in up to 88% ee. Its reactions with trans-alkenes are more enantioselective (67% ee) than with the cis-alkenes (40% ee). Based on a two-dimensional NOESY NMR study, 2a was found to adopt a more open conformation in benzene than in dichloromethane, which explains the observed solvent-dependent enantioselectivity of its reactions with alkenes. The oxidation of aromatic alkenes by the chiral dioxoruthenium(VI) porphyrins proceeds through the rate-limiting formation of a benzylic radical intermediate; the observed enantioselectivity (eeobs) depends on both the facial selectivity of the first C-O bond formation step and the diastereoselectivity of the subsequent epoxide ring closure. To account for the observed facial selection, "side-on" and "top-on" approach transition state models are examined (see: B. D. Brandes, E. N. Jacobsen, Tetrahedron Lett. 1995, 36, 5123).

Modulatory effects of compound polysaccharide erweikang on the immune function of mice.

In order to study the modulatory effects of compound polysaccharide Erweikang on the immune function of the immunosuppressive mice, the positive immune modulation of Erweikang at different doses and different time points was compared when normal saline and cyclophosphamide served as controls. The results showed that different doses of Erweikang could effectively reverse the decreased induction levels of NK, IL-2, gamma-IFN and the contents of TNF and IgF of the immunosuppressive mice after administration of Erweikang for 7 to 14 days. It was suggested that compound polysaccharide Erweikang presents positive immune modulation in dose- and time-dependent manners.

Anisotropy in domain engineered 0.92Pb(Zn1/3 Nb2/3)O3-0.08PbTiO3 single crystal and analysis of its property fluctuations.

The orientation dependence of slowness and electromechanical coupling coefficients of 0.92Pb(Zn1/3 Nb2/3)O3-0.08PbTiO3 (PZN-8%PT) domain engineered single crystal was analyzed based on the measured complete set of elastic, piezoelectric, and dielectric constants. There exist one quasi-longitudinal, one quasi-shear, and one pure shear wave in each of the [100]-[010], [010]-[001], and [001]-[110] planes. The slowness of the quasi-shear wave exhibits strong anisotropy in all three planes, and the coupling coefficient k33 and k31 reach their maximum in [001] and [110] directions of cubic axis, respectively. Because the composition of PMN-8%PT system is very close to the morphotropic phase boundary, the extraordinary large piezoelectric coefficients d31 and d33, and high coupling coefficient k33 are very sensitive to compositional variation. We have performed error analysis and proposed an improved characterization scheme to derive a complete data set with best consistency.

[Relationship between the immunohistopathological changes of hepatitis B virus carrier mothers' placentas and fetal hepatitis B virus infection].

OBJECTIVE: To investigate the role of placenta in intrauterine transmission of hepatitis B virus (HBV), and the relationship between the fetal HBV infection and the placental infection. METHODS: Cord blood or peripheral blood was obtained from 61 newborn infants of HBsAg positive mothers. Neonates (28 of 61) infected by HBV were assigned to fetal infected group, and 33 infants without infection were assigned to control group. Histopathological changes of placentas from 61 HBsAg positive mothers were observed by pathological examination and classification. RESULTS: (1) The positive rate of HBsAg/HBcAg detected in the placentas of fetal infected group was 82% (23/28), which was significant higher than that of control group (55%), P < 0.05. (2) Among various types of cells in placental tissue, the amnionic cell showed higher HBsAg/HBcAg positive rate in fetal infection group, which was 36% (10/28), than that of control group (6%) (P < 0.01). (3) The syncitial cell was the cell with the higher detecting HBsAg/HBcAg positive rate (49%) comparing with other types of cells in placental barrier (P < 0.05). (4) The incidence of fibrinoid necrosis and chorionic hyperemia in fetal infection group were 29% and 50%, respectively, which were higher than those in control group (9%, 15%). The detecting rate of Hofbauer cell in fetal infection group was 46%, significantly lower than that in control group (79%) (P < 0.05). CONCLUSIONS: HBV infection of fetus is associated with placental infection. HBV infection of amnionic cell is an important factor of intrauterine infection. Placental barrier can protect the fetuses from infection to some extent. Some histopathological changes of placental tissue, for example fibrinoid necrosis, chorionic hyperemia and decreasing number of Hofbauer cells, may play a role in fetal HBV infection.

Normal and abnormal neuronal migration in the developing cerebral cortex.

Neuronal migration is the critical cellular process which initiates histogenesis of cerebral cortex. Migration involves a series of complex cell interactions and transformation. After completing their final mitosis, neurons migrate from the ventricular zone into the cortical plate, and then establish neuronal lamina and settle onto the outermost layer, forming an "inside-out" gradient of maturation. This process is guided by radial glial fibers, requires proper receptors, ligands, other unknown extracellular factors, and local signaling to stop neuronal migration. This process is also highly sensitive to various physical, chemical and biological agents as well as to genetic mutations. Any disturbance of the normal process may result in neuronal migration disorder. Such neuronal migration disorder is believed as major cause of both gross brain malformation and more special cerebral structural and functional abnormalities in experimental animals and in humans. An increasing number of instructive studies on experimental models and several genetic model systems of neuronal migration disorder have established the foundation of cortex formation and provided deeper insights into the genetic and molecular mechanisms underlying normal and abnormal neuronal migration.

[An experimental study of therapeutic effect of basic fibroblast growth factor on experimental retinal ischemia/reperfusion injury].

OBJECTIVE: To evaluate the therapeutic effect of basic fibroblast growth factor (bFGF) injected into the vitreous cavity on experimental retinal ischemia/reperfusion injury. METHOD: The Wistar rat model of experimental retinal ischemia/reperfusion injury was made by increasing the intraocular pressure. The rats were divided into normal, ischemia and treatment groups randomly. At the beginning of reperfusion, normal saline was injected into the vitreous cavity in ischemia group and 2 micro g of bFGF was injected into the treatment group. The histological and ultrastructural changes in retina of different time after reperfusion were observed. The retinal ganglion cell number was counted by using microscope. The thickness of inner layer of retina was measured by using Image Diagnosis System. RESULT: In the early period of retinal ischemia/reperfusion injury, the edematous status of retina of treatment group was lighter than that of the ischemia group. The retinal inner layer thickness and the RGC number of treatment group were greater than that of the ischemia group during all the post-reperfusion stages. At 168 hours after reperfusion, the thickness of the retinal nerve fiber layer and the RGC numbers of ischemia groups were obviously lower than that of the normal groups. But the differences of thickness of the nerve fiber layer and RGC number between the treatment and normal group were of no statistical importance. The nuclear membranes of RGCs were edematous and the mitochondrial cristae were unclear at 24th hour after reperfusion, the apoptotic body can be found, and the microtubules in nerve fibers became unclear and even disappeared in the ischemic group. While in the treatment group, the nucleus was clear, only part of the nuclear membrane was edematous, the cell organs were abundant and the structures of mitochondria and microtubules were clear. CONCLUSION: Injection of bFGF into the vitreous cavity has the therapeutic effect on experimental retinal ischemia/reperfusion injury.

[Analysis about clinical data of intrauterine infection of hepatitis B virus].

OBJECTIVE: To investigate a practical diagnostic method in clinic for fetuses infecting with hepatitis B (HBV) and study the mutual effects between fetal infection and clinical factors. METHODS: Venous blood was drawn from 144 cases of HBV carrier mothers and their neonates. HBV DNA was detected by polymerase chain reaction (PCR) and hybridization, HBV M was detected by enzyme linked immunoadsorbent assay (ELISA), and aspartate aminotransferase/alanine aminotransferase (AST/ALT) was detected by IFCC. Umbilical blood and femoral blood was taken from 40 of 144 neonates for HBV DNA detection. Clinical data, neonatal AST and ALT level were compared between fetal infection group and control group. RESULTS: (1) The fetal infectious rate was 22.9% (33/144). Comparing with peripheral venous blood sample, the sensitivity and positive predictive value of HBV DNA detected in cords was 100.0%, 80.0% respectively. Following up the infants, HBV DNA was found persistently positive in 7 of 28 intrauterine infectious infants 6 approximately 9 months after birth. HBsAg was found changing to be negative 1 month later in the infants with HBsAg positive at birth. (2) The fetal infectious rate in mothers with HBeAg (+) or HBV DNA (+) was 70.5%, 61.1% respectively which was significantly higher than that in mothers with HBeAg (-) or HBV DNA (-). P < 0.01. There was no significantly difference in mothers' age, gestational age, delivery way, birth weight (BW), body length (BL), Apgar score between fetal infectious group and control group. (3) The mean value of AST, ALT in fetal infectious group was (61.2 +/- 31.3) IU, (24.7 +/- 14.9) IU respectively, which was significantly higher than that in control group [(55.2 +/- 37.1) IU, (19.0 +/- 10.1) IU]. P < 0.01. CONCLUSIONS: (1) Detection of HBV DNA in cord blood is a sensitive index for diagnosing fetal infection, however detection of peripheral venous blood is with the significance of making correct diagnosis. (2) HBsAg or HBV DNA positive in mothers is one of the risk factors of intrauterine infection. There is no relationship among fetal infection and mothers' age, gestational age, delivery way, neonates' sex, BW, BL. (3) The liver function of neonates infected with HBV intrauterinely maybe impaired to some extent.

[Study on granulocytes derived from induction of committed differentiation of hematopoietic stem/progenitor cells ex vivo].

To evaluated the feasibility of preventing infection after high dose chemotherapy and radiotherapy using the granulocytes derived from differentiated from hematopoietic stem/progenitor cells ex vivo, human CD34-positive cells were isolated from umbilical cord blood by using a high-gradient magnetic cell sorting system (MACS), and the cells committedly differentiated with hematopoietic cytokines (SCF + IL-3 + IL-6 + G-CSF) in a liquid culture system. The expanded cell number, ratio of the viable cells, chromosome and phenotype of the differentiated cells and safety analysis of expanded cells were detected by using cell count, trypan blue exclusion test, karyotype analysis, flow cytometry and tumorigenic model of nude mice, respectively. The results showed that the combination of cytokines increased cell number by (1006.4 +/- 103.2) folds and flow cytometric analysis showed myeloid marker CD11b expressed in the about 60% cells. The growth peak of differentiated cells was at 14 days of culture and decreased at about 33 days. No abnormality was found in the karyotype analysis of expanded cells. No tumor was found in the nude mice injected with expanded cells after 35 days and the expanded cells had the ability of phagocytizing bacteria. It is concluded that the cells, differentiated from CD34(+) cells, expanded ex vivo possess the function of granulocyte and it was safe for clinical trial.

[Expression of telomerase during induction of committed differentiation of human cord blood hematopoietic stem/progenitor cells in vitro].

To investigate the expression of telomerase in cord blood hematopoietic stem/progenitor cells during their committed differentiation in vitro and provide an index of monitoring the proliferating potential of the hematopoietic stem/progenitor cells and security for clinical application. Human CD34 positive cells were isolated from umbilical cord blood by using magnetic cell sorting system (MACS), and were induced to differentiation with hematopoietic growth factors (SCF + IL3 + IL6 + GCSF and SCF + IL3 + IL6 + EPO) in a liquid culture system. The telomerase activity and the cytalytic subunit of telomerase (hTERT) of the cells were analysed during different periods of culture by using TRAP-PCR, TRAP-ELISA, Western blot and RT-PCR techniques, respectively. The results showed that a peak of cell growth was achieved on day 14 - 21 during induction of differentiation in vitro. Total cell number could increase 1006.4 +/- 103.2 times and could not increase there after. Telomerase activity and hTERT expression were low in freshly isolated cord blood CD34(+) cells and increased after about 7 days of culture in addition of cytokine combinations of SCF + IL3 + IL6 + GCSF and SCF + IL3 + IL6 + EPO, respectively. The telomerase activity and hTERT decreased after 14 days of culture and were not detected after 28 days of culture. It was concluded that the hematopoietic stem/progenitor cells can be expanded in large number in vitro and do not have the character of immortality and the telomerase activity could be a useful index in hematopoiesis regulation.
