Distribution of bemitil in organs and tissues of rats after single or repeated administration.

After single and repeated peroral administration of bemitil to rats this drug was found in the liver, brain, kidneys, spleen, heart, skeletal muscles, lungs, adipose tissue, and testicles. After single treatment accumulation of bemitil was most pronounced in the liver. After repeated treatment the decrease in bemitil concentration in the liver was probably associated with increased elimination of the drug from liver tissue due to intensification of its biotransformation. We conclude that bemitil can accumulate in the blood, but not in tissues.

[Pharmacoprophylaxis of sea-sickness syndrome with the help of homeopathic drug "Avia-more"].

Anti-sea-sickness effect of a new homeopathic drug [see text] and its influence on operator's working capacity during Coriolis acceleration was evaluated in blind placebo-controlled investigation conducted in practically healthy volunteers with low vestibular resistance. For comparison "Dimenhydrinate" was used. The investigation has demonstrated that [see text] has rather strong anti-sea-sickness effect, is comparable with dimenhydrinate effectiveness and prevents the development of disorders in sensory-and-motor and cognitive functions during the influence of static-and- kinetic loads. According to the data obtained the protective action of [see text] is most likely connected with adaptive effect on organs and systems that form the functional protective system against sea-sickness. The negative influence of the drug on CNS complex functions was not observed that allows recommending its prophylactic use in subjects with low sea-sickness resistance including the operators.

Neuroprotective activity of proproten in rats with experimental local photothrombosis of the prefrontal cortex.

Proproten (ultralow doses of antibodies to S100 protein) exhibited neuroprotective activity in rats with experimental photochemical thrombosis of the prefrontal cortex. Proproten was more potent than standard neuroprotectors piracetam and vinpocetine in alleviating the signs of memory disorders produced by ischemic injury. Pathomorphological study of the damaged area confirmed the neuroprotective effect of Proproten.

Pharmacological regulation of functional activity of stem cells in restoration of the myocardium during the postinfarction period.

The effect of granulocytic CSF on myocardial tissue recovery after acute myocardial infarction was studied on rats. A course of granulocytic CSF after ligation of the left coronary artery normalized ECG parameters and morphological picture of the myocardium 1 month after treatment. It was shown on mouse model of myocardial infarction that this process was associated with more intensive mobilization and homing of mesenchymal stem cells in the heart.

Mechanisms of hepatoprotective effect of preparation containing superlow doses of antibodies to granulocytic colony-stimulating factor.

The hepatoprotective effects of superlow dose preparation of antibodies to granulocytic CSF were studied on a model of CCl4-induced hepatitis. The preparation exhibited high antiinflammatory and antisclerotic activities determined by stimulation, mobilization, and determined homing of mesenchymal stem cells into damaged liver with subsequent differentiation of these cells into mature hepatocytes.

Mechanisms of the effects of granulocytic CSF on tissue reparation during chronic CCl4-induced damage to the liver.

Hepatoprotective effects of granulocytic CSF were studied using experimental model of CCl4-induced hepatitis. It was found that treatment with granulocytic CSF increased the content of stromal precursors and mesenchymal stem cells in the bone marrow and peripheral blood with subsequent increase in the number of hepatic precursor cells in the liver. These findings attest to mobilization of progenitor mesenchymal cells and their migration into damages liver tissue, which accelerates its regeneration related to changes in the parenchyma, but not connective tissue development.

A randomized, open-label, comparative, 6-month trial of oral ultra-low doses of antibodies to tumor necrosis factor-alpha and diclofenac in rheumatoid arthritis.

Artrofoon (oral ultra-low doses of antibodies to TNF-alpha is a novel drug approved by the Russian Ministry of Health for the treatment of rheumatoid arthritis (RA). The aim of this study was to assess clinical efficacy and safety of artrofoon in RA compared with diclofenac. In a 6-month, randomized, open-label, comparative trial, 60 patients with active RA (eight men and 52 women aged 23 to 62, mean disease duration 10 years) received artrofoon (8 tablets daily, n = 30) or diclofenac (100 mg daily, n = 30). RA signs and symptoms as well as serum levels of inflammatory markers were evaluated before treatment and at months 1, 3 and 6. Most patients in the artrofoon group showed a 20% improvement in major RA symptoms by the end of the study. The clinical effect rose gradually reaching maximum at month 6. In the artrofoon group, 57% of the patients achieved an American College of Rheumatology (ACR) 20% criteria (ACR20) by month 6 versus 20% of those receiving diclofenac. In some patients in the artrofoon arm, serum proinflammatory cytokine levels significantly decreased (> or = 25% reduction). Diclofenac produced a less pronounced clinical effect, and no changes in cytokine profile. Unlike conventional nonsteroidal anti-inflammatory drugs, artrofoon produced no adverse effects and the overall tolerability and safety were excellent. A half-dose treatment with artrofoon (4 tablets daily) was able to sustain clinical improvements over a 6-month follow-up period. To conclude, artrofoon is a safe and effective treatment for rheumatoid arthritis that acts by influencing the inflammatory process.

Dose-dependent effects and specificity of action of antibodies to endogenous regulators in ultralow doses.

We studied the effects of antibodies against interferon-gamma, erythropoietin, and tumor necrosis factor-alpha in ultralow doses on the production of endogenous interferon-gamma. Course peroral treatment with potentiated antibodies to interferon-gamma in various dilutions produced similar changes in experimental animals. The treatment stimulated spontaneous production of endogenous interferon-gamma by mouse lymphocytes. It should be emphasized that antibodies to other cytokines in ultralow doses did not stimulate interferon-gamma production. These data illustrate the specificity of action of antibodies to endogenous regulators in ultralow doses.

[Efficacy of therapeutic and prophylactic actions of ultralow doses of antibodies to gamma-interferon in experimental murine model of herpes virus].

The process of the disease due to herpes simplex virus types 1 and 2 (HSV-1 and 2) was studied on white uninbred mice weighing 10 to 12 g. The animals were infected intracerebrally or intraperitoneally. Intraperitoneal contamination of the animals with MS strain of HSV-2 was used for the experimental model of the herpes simplex infection. The prophylactic antiherpes action of ultralow doses of the human gamma-interferon antibodies (ULD of anti-IFN-gamma) at a course of its intragastral administration was evaluated. The preparation was shown to have a significant (p < 0.05) protective effect in a dose of 10 LD50, evident from a 10-fold decrease of the HSV-2 accumulation in the brain, a lower percentage of the animal deaths and an increase of the average lifespan of the animals by 3.3 days. The study of the therapeutic action of ULD of anti-IFN-gamma at a course of its intragastral administration showed that the preparation had no significant positive effect on the disease process in the animals infected with HSV-2 in a dose of 10 LD50. However, a positive effect associated with delayed virus replication in the brain was observed in the study on the therapeutic effect of ULD of anti-IFN-gamma after its intragastral administration to the mice infected with a sublethal dose of the virus.

[Antiviral activity of oral ultralow doses of antibodies to gamma-interferon: experimental study of influenza infection in mice].

Course intragastric administration of ultralow doses of human gamma-interferon antibodies (ULD anti-IFN-gamma) to intact mice resulted in an increase of endogenous IFN-gamma production by the animal lymphocytes. Oral prophylactic administration of ULD anti-IFN-gamma significantly lowered the influenza virus concentration in the animal lungs at the initial stage of the aerogenous infection: in 2 (p = 0.05) and 3 (p = 0.07) days after the contamination. The therapeutic antiviral effect of ULD anti-IFN-gamma in mice with influenza was evident from a significant decrease of the influenza virus concentration in the lungs of the animals on the 4th (p = 0.05) and 5th (p = 0.07) days after the contamination. The antiviral effect of ULD anti-IFN-gamma after the prophylactic and therapeutic use is likely provided by induction of endogenous IFN-gamma.

Efficiency of potentiated antibodies to tumor necrosis factor-alpha (Artrofoon) in the therapy of patients with rheumatoid arthritis.

Antiinflammatory activity of a new preparation Artrofoon (antibodies against tumor necrosis factor-alpha in ultralow doses) and nonsteroid antiinflammatory drug diclofenac in patients with rheumatoid arthritis was compared in an open randomized trial. The course of treatment with Artrofoon more significantly improved clinical and laboratory signs in patients with the articular form of rheumatoid arthritis than diclofenac.

Antidepressant properties of Proproten and amitriptyline: comparative experimental study.

Antidepressant properties of Proproten (antibodies to S100 protein in ultralow doses) were studied on outbred albino rats. The animals were subjected to the Porsolt's test of behavioral helplessness and Nomura's test of forced swimming in a reservoir with freely rotating wheels. Proproten in a dose of 2.5 ml/kg produced a strong antidepressant effect. It was observed after single and repeated (5 days) peroral treatment with the preparation. Proproten decreased the immobility time (Porsolt's test) and increased the number of wheel turns (Nomura's test). The activity of Proproten compared well with the standard preparation amitriptyline. As differentiated from amitriptyline, Proproten did not produce the sedative effect.

Cytogenetic effects of antibodies to gamma-interferon in ultralow doses.

Single and course administration of ultralow doses of antibodies gamma-interferon did not increase the incidence of cytogenetic abnormalities in bone marrow cells from BALB/c mice and produced no genotoxic effect on Drosophila melanogaster wing cells in the test of somatic mosaicism.

Immunotropic effects of potentiated antibodies to human interferon-gamma.

We studied immunotropic properties of antibodies to human interferon-gamma in homeopathic doses. The effects of these antibodies on the ratio between the counts of T helper (CD4+) and T suppressor cells (CD8+), production of interleukin-1 and interleukin-2, and functional activity of natural killer cells were evaluated. Potentiated antibodies slightly decreased the number of CD8+ cells in the spleen of experimental animals. After the addition to cultured human mononuclear cells the preparation produced a moderate comitogenic effect, stimulated proliferation of T and B lymphocytes, increased spontaneous and induced production of interleukin-1, and enhanced functional activity of natural killer cells (under certain conditions).

Effects of ultralow doses of antibodies to prostate-specific antigen on morphological and functional state of rat prostate.

Morphological and morphometric assays showed that administration of antibodies to the prostate-specific antigen in ultralow doses for 1.5 months delayed the development of atrophic and sclerotic processes in rats with chronic aseptic prostatitis. The concentration of zinc ions playing an important role in binding of androgens increased in the prostate of rats receiving the preparation. Studies of copulatory behavior showed that male rats with chronic prostatitis receiving antibodies to the prostate-specific antigen displayed increased sexual activity compared to control and intact animals.

Clinical efficiency of Epigam in patients with exacerbation of peptic ulcer disease of the stomach and duodenum.

Clinical efficiency and safety of Epigam containing antibodies to histamine in ultralow doses were studied during the therapy of patients with exacerbation of peptic ulcer disease of the stomach and duodenum associated with Helicobacter pylori infection. We examined 20 patients (18-50 years) with ulcerative lesions of the mucosal layer in the stomach and duodenum and H. pylori infection. Epigam (1 tablet, 6 times a day) or H2 receptor blocker ranitidine (150 mg, 2 times a day) were given in combination with amoxicillin (500 mg, 3 times a day, 14 days) and metronidazole (500 mg, 2 times a day, 14 days) for 28 days. The efficiency of treatment was determined before and 1, 2, 3, and 4 weeks after the start of therapy. The symptoms of peptic ulcer disease and time of ulcer healing underwent similar changes in patients of both groups. However, after ranitidine therapy pain syndrome disappeared more rapidly than in patients receiving Epigam. Epigam did not cause undesirable side effects. Our results indicate that Epigam is an efficient and safe preparation that may be used for combination therapy of patients with peptic ulcer disease of the stomach and duodenum.

Anxiolytic effect of Proproten under conditions of punished and unpunished behavior.

The anxiolytic effect of Proproten containing potentiated antibodies to brain-specific S100 protein (2.5 ml/kg) in male outbred albino rats was studied under conditions of punished (Vogel's conflict situation with pain stimulation) and unpunished behavior (elevated plus-maze and open-field tests). Proproten significantly increased the incidence of punished drinking in the conflict situation with pain stimulation, number of entries, and time spent in the open arms of the elevated plus-maze and decreased the rate of defecation and urination. In the open-field test Proproten induced entries of rats into the center of an illuminated area. Proproten was efficient after single administration and course of treatment (2 times a day, 5 days). These results show that Proproten produces the anxiolytic effect under conditions of punished and unpunished behavior.

GABAergic system in the anxiolytic effect of Proproten: experimental study.

The medicinal preparation Proproten contains ultralow doses of antibodies to S100 protein that acts as an important regulator of integrative activity in the brain and synaptic processes. Intracerebroventricular administration of Proproten, diazepam, and mexidol in doses of 2.5 ml/kg, 2 mg/kg, and 100 mg/kg, respectively, produced a strong anxiolytic effect on male outbred rats in the conflict situation and markedly increased the incidence of punished drinking. Antagonists of GABAergic transmission bicuculline (GABAA receptor blocker) and picrotoxin (chlorine channel blocker) produced the pro-conflict anxiogenic effect, which was accompanied by a decrease in the number of punished drinking in control animals. The anti-conflict effect of Proproten was less pronounced during blockade of GABAA receptors or chlorine channels. Bicuculline and picrotoxin similarly modulated the anxiolytic effect of diazepam and mexidol. Our results suggest that the GABAergic system plays a role in the anxiolytic effect of diazepam, mexidol, and Proproten.
