Stimulation of adrenal medullary cells in vivo and in vitro induces expression of c-fos proto-oncogene.

The nuclear proto-oncogene, c-fos, has been implicated in the coordinated regulation of gene expression during cell proliferation and differentiation. In this study, we have demonstrated the induction of the c-fos gene products in differentiated cells of the adrenal medulla by non-mitogenic signals. Activation of adrenal medullary cells in vivo by insulin-induced hypoglycemia, and in vitro by nicotine or angiotensin resulted in the rapid and transient elevation of c-fos mRNA levels. Induction of the c-fos mRNA by angiotensin and nicotine were accompanied by the appearance of the c-fos protein. The increase in c-fos protein occurred initially in the cytoplasm and, later, in the nucleus, and it was co-localized with tyrosine hydroxylase. Nuclear expression of the c-fos protein was also induced by veratridine, forskolin and the calcium ionophore A231287. The role of calcium in the regulation of the c-fos gene by angiotensin with nifedipine and inhibition of the effects of angiotensin with nifedipine and sphingosine, a protein kinase C inhibitor. Activation of the c-fos gene may play a role in the coordinated induction of genes involved in the long-term adaptation of adrenal medullary cells to increased functional demands.

Studies on the phenomenon of latent propagated sensation along the channels. II. Investigation on the lines of LPSC on the twelve main channels.

Applying an electric impulse at the Jing points and then tapping on the skin surface along the lines perpendicular to the channels, latent PSC of 12 channels was examined respectively on 200 patients at an acupuncture clinic. Of the cases examined, 68.5% were found to have positive latent PSC. No prominent difference was found among the 12 main channels. In patients with higher sensitivity of latent PSC and in those with marked PSC as well, the traces of latent PSC can be located simply by tapping (i.e. without electric impulsation at the Jing points), and the width of the sensitive lines is about 0.5 cm. Furthermore, in cases with marked PSC, the latent sensation lines were always found to be a continuation of the marked ones. These facts indicate that the latent PSC is a common meridian phenomenon in a large number of individuals.

Studies on live attenuated mumps vaccine. II. Follow-up study on the efficacy of Biken Vaccine.

Clinical and serological follow-ups were made on 18 children for 3 years and on another 18 children for 4 years, after immunization against mumps by attenuated mumps vaccine; Biken vaccine. To evaluate the protective efficacy of the vaccine, matched controls were studied during the same period. Serological examinations revealed that 77% of the vaccinees and 93% of the controls were infected with mumps after close contacts with mumps patients. Regular mumps was contracted by 88% of the controls, but none of the vaccinees. There was no substantial decrease in the antibody titers in unexposed vaccinees after vaccination.

Effects of microtubule inhibitors-taxol, vinblastine and estramustine on the growth and p53 gene expression in the hormone independent human prostatic JCA-1 cells.

Treatment of the JCA-1 prostate cells with different concentrations of microtubule inhibitors-taxol (paclitaxel) vinblastine, and estramustine-was accompanied by reduced cell growth and, correspondingly, characteristic morphological changes. The reduction of cellular proliferation was unlikely to be attributable to an inhibitory effect of the antimicrotubule agents on cell cycling, but was correlated with the decreased expression of the p53 gene. These results suggest that antimicrotubule agents may be considered as potentially complementary treatment modalities for patients with hormonally independent prostate cancer.
