Radical differences in CYP1A1 genotype and function.

The cytochrome P450 1A1 (CYP1A1) gene may be of critical importance in determining individual cancer susceptibility to aromatic hydrocarbons such as those in tobacco smoke. We compared the frequencies of CYP1A1 haplotypes, and complete genotypes, taking into account polymorphisms at 3 sites, including an African-specific polymorphism. No concordance was observed in Africans or African-Americans between any of the 3 polymorphisms, (Msp1 restriction fragment length polymorphism (RFLP)--'M', exon 7--'E', new RFLP--'A') in contrast to the concordance seen between the M and E polymorphisms in Caucasians and Asians. We observed an effect of the E polymorphism on enzyme activity and mRNA induction in Asians and Caucasians.

Cancer in Mali, 1987-1988.

Results from the population cancer registry in Bamako, Mali, for the years 1987 and 1988, are presented. The age-standardized incidence for all cancers is high compared with rates reported elsewhere in West Africa (119.6 per 10(5) in males and 88.3 per 10(5) in females), but the leading cancers in each sex are the same (liver cancer in males, cervix cancer in females). The incidence of stomach cancer is the highest recorded in Africa, while rates for lung cancer, although low, exceed those in earlier series from registries in the region.

Effect of active and passive immunization on the development of experimental Pseudomonas aeruginosa pyelonephritis in mice.

Experimental pyelonephritis was produced in mice by the intravenous injection of Pseudomonas aeruginosa. Immune response to infection was studied by passive hemagglutination antibody titers. Vaccination of mice with live P. aeruginosa or culture filtrates (Pseudomonas antigen) induced antibodies and resulted in a high degree of protection against death and pyelonephritis following subsequent hematogenous challenge with the homologous strain. Transfer of immune serum protected mice against death following infection with the homologous strain and with a heterologous strain. However, immune serum failed to protect mice from kidney infection by the heterologous strain. These data indicate that immune serum seemed to protect against early, overwhelming bacteremia but did not prevent a chronic course of kidney infection by a heterologous strain.

[Ectopic spleens. About a spleen of the right iliac fossa (author's transl)].

A rare case of the presence of the spleen in the right iliac fossa is reported. The frequency and signs of ectopic and supernumerary spleens are described, the latter requiring differentiation from peritoneal splenosis. An embryological explanation for the presence of the spleen in the right iliac fossa is suggested.

[Histopathological observations of experimental hematogenous infection with Pseudomonas aeruginosa in mice (author's transl)].

The effect of intravenous injection of Pseudomonas aeruginosa in mice was studied in various conditions : dose of bacteria injected (2.5 X 10(6) to 4.5 X 10(7)) , moment of necropsy (one hour to 4 months after infection), number of injections (one, two and eight). Gross and microscopic examinations of tissues included kidney, lung, spleen and liver. The frequency, the type and the time of appearance of the lesions depend upon the dose of organisms, upon the individual susceptibility of the mouse and upon the number of injections. Abscesses preferentially localised in kidneys appeared in mice that received only one injection of bacteria. They were visible to the naked eye as soon as two days after inoculation with a large dose. Granulomatous inflammatory reaction was observed in the different tissues, however it was predominantly seen in the kidney and in animals that received several injections. In the liver and the spleen hyperplasia and hypertrophy of lymphoid, myeloid, megacaryocytic and mononuclear phagocytic cells were observed. This model of experimental pyelonephritis due to P. aeruginosa seems to us useful to study the factors promoting localisation and multiplication of this organism in a tissue.

Distribution of composite CYP1A1 genotypes in Africans, African-Americans and Caucasians.

We present the genotype distribution of the CYP1A1 gene in a sample of over 300 subjects of various ethnic origins. Genotypes are presented as composites of eight possible alleles, taking into account the three major polymorphisms, including a recently described African-American-specific MspI RFLP. A new nomenclature system is presented for clarifying the various haplotypes. Interesting interracial differences in allelic frequencies and admixture rates were observed for the three polymorphisms. Because of the importance of the CYP1A1 gene (which encodes the aromatic hydrocarbon hydroxylase) as a biomarker of genetic susceptibility to environmental carcinogens such as polycyclic aromatic hydrocarbons, these data may provide a useful reference for future studies of relationships between CYP1A1 genotype and disease susceptibility.
