Effects of temperature, salinity, desiccation and chemical treatments on egg embryonation and hatching success of Benedenia seriolae (Monogenea: Capsalidae), a parasite of farmed Seriola spp.

The effects of temperature and salinity on the embryonation period and hatching success of eggs of Benedenia seriolae were investigated. Temperature strongly influenced embryonation period; eggs first hatched 5 days after laying at 28 degrees C and 16 days after laying at 14 degrees C. The relationship between temperature and embryonation period is described by quadratic regression equations for time to first and last hatching. Hatching success was >70% for B. seriolae eggs incubated at temperatures from 14 to 28 degrees C. However, no B. seriolae eggs embryonated and hatched at 30 degrees C and <2% of eggs hatched when incubated at 24 degrees C after transfer to 30 degrees C for 48 h. Embryonation period was similar for eggs incubated in sea water at 25, 30 and 35 per thousand salinity, but increased for eggs incubated at higher or lower salinities. When incubated at salinities ranging from 25 to 45 per thousand, more than 70% of B. seriolae eggs embryonated and hatched. Hatching success was lower at 20 and 50 per thousand salinity and few or no eggs hatched at 10 and 15 per thousand. Hatching of B. seriolae eggs can be prevented by desiccation for 3 min, by immersion in water at 50 degrees C for 30 s or by treatment with 25% ethanol for 3 min.

Synthesis and characterization of segmented polyurethanes based on amphiphilic polyether diols.

Segmented polyurethanes (SPUs) based on polyethylene glycol (PEG), polypropylene glycol (PPG) and a series of Pluronics with different ethylene oxide/propylene oxide ratios (EO/PO) and molecular weights were prepared. Different diisocyanates were used for making SPUs: 4,4-diphenylmethane diisocyanate (MDI), 4,4-dicyclohexylmethane diisocyanate (MDCI), hexamethylene diisocyanate (HMDI) and isophorone diisocyanate (IPDI). 1,4-Butane diol (BD) and ethylene diamine (ED) were used as chain extenders. The polymers obtained were characterized by infrared spectroscopy (IR), nuclear magnetic resonance (NMR) and differential scanning calorimetry (DSC). The microphase morphology (phase separation and phase mixing) is discussed in more detail.

Stimulation of thyroid function by several pituitary hormones results in an increase in plasma thyroxine and reverse triiodothyronine in tilapia (Tilapia nilotica).

In this study, intravenous injection of several doses of porcine follicle stimulating hormone (pFSH: 0.002, 0.01, 0.05, and 0.5 micrograms/g body wt), bovine TSH (bTSH: 0.5 micrograms/g body wt), and ovine growth hormone (oGH: 0.04, 0.02, and 0.4 microgram/g body wt) stimulated an increase in plasma thyroxine (T4) and reverse triiodothyronine (rT3) in tilapia. This effect occurred in a dose-dependent manner. pFSH was the most potent in stimulating thyroid function. The dose of 0.002 microgram pFSH/g body wt increased plasma levels of T4 over control levels (2.59 +/- 0.16 ng/ml) about 2.5-fold within 4 hr, whereas a concentration of 0.5 micrograms/g body wt caused a great and prolonged increase of T4 and rT3 levels. Control levels (2.59 +/- 0.16 ng/ml for T4 and 40.37 +/- 8.60 pg/ml for rT3) were increased 19- and 22-fold respectively, over 24 hr. An increase of T4 and rT3 levels occurred also after injection of total hypophyseal extract and Con A II glycoprotein fraction of a tilapia pituitary homogenate, whereas the protein fraction failed to alter plasma concentrations of T4 and rT3. rT3 levels were also significantly increased at 2 hr, but not at 1 hr, after injection of T4. Basal T3 levels (1.90 +/- 0.22 ng/ml) were reduced by half over 24 hr in all experiments. These results suggest the existence, in tilapia, of a 5-D pathway deiodination of T4 which is pituitary independent. Stimulation of T4 release is always followed by an increase in plasma rT3 levels.

Chronological appearance of the different hypophysial hormones in the pituitary of sea bass larvae (Dicentrarchus labrax) during their early development: an immunocytochemical demonstration.

Antisera raised against chum salmon prolactin (PRL), rainbow trout growth hormone (GH), mammalian adrenocorticotropic hormone (ACTH), thyroid-stimulating hormone (TSH), and luteinizing hormone (LH) were used to study the chronological appearance of immunoreactivity for PRL, GH, ACTH, TSH, LH, and melanocyte-stimulating hormone (MSH) in the pituitary of sea bass larvae (Dicentrarchus labrax) during the first 26 days after hatching. The anti-ACTH gives positive immunostaining in the ACTH cells as well as in the MSH cells; however, the two cell types can easily be distinguished by their different localization in the pituitary: ACTH in the rostral pars distalis, MSH in the pars intermedia. The first day after hatching cells immunoreactive for TSH, GH and ACTH could already be noticed, ACTH reacted strong in the pars intermedia but very weak in the rostral pars distalis. Cells immunopositive for PRL became visible between Days 9 and 15. With anti-LH, no positive reaction could be obtained during the first 26 days after hatching.

Circadian variations in plasma osmolality, electrolytes, glucose, and cortisol in carp (Cyprinus carpio).

Male carps (Cyprinus carpio) of about 1.5 kg were used in all experiments performed in December and February. Blood was taken by heart-puncture within 1 min or less every 4 hr from different fishes during a 24-hr period to establish circadian rhythmicities of all parameters. The water temperature was 11-12 degrees. In December circadian variations could be calculated for cortisol, Na+ and plasma osmolality in partly fed carps and for cortisol, Ca2+, glucose, and plasma osmolality in food deprived animals. No difference in cortisol level, amplitude, or acrophase was present between both experiments but plasma Na+ and osmolality was more elevated when feeding. Also the acrophase of the osmolality rhythm differed between both groups. In February carps, cortisol levels were still comparable to December ones, but a 4 hr shift in acrophase had occurred (from +/- 02 hr to 06 hr). Levels of all other parameters also were comparable to December, except for glucose with only 1/5th of the December values. There was also a shift in acrophase present from the morning to around midnight. The level of cortisol in February carps acclimated to 18 degrees for 1 week was twice the one found in the 11 degrees group. At the same time a significant increase in amplitude and shift in acrophase to 22 hr was seen. Other parameters, except for glucose of which the level was significantly lower than in the 11 degrees group, remained unchanged. Also no correlation between the individual 24-hr data of all parameters could be found. It is therefore concluded that cortisol is not responsible for the observed circadian rhythmicities of Na+, Ca2+, glucose, or plasma osmolality.

Surface plasmon resonance: a technique for characterizing biomaterial surfaces.

Surface plasmon resonance is an optical technique that allows the study of surface phenomena in real time, without preliminary labelling. The technique is useful in biomaterial science to determine affinity constants, concentrations, kinetics, film thickness and dielectric properties of material surfaces. This article examines the technology and some of its applications.
