Amounts of fibrous proteins and matrix substances in hairs of different races.

Fibrous proteins (FPs) and matrix substances (MSs) were extracted as S-carboxymethylated (SCM) derivatives from scalp hair of five individuals each of the Mongoloid, Negroid and Caucasoid races. The ratio of the extracted SCM FPs to SCM MSs in dry weight was calculated for each hair specimen. The ratios obtained from the Mongoloid hair were 0.45 +/- 0.03; those from the Negroid, 0.18 +/- 0.02; and those from the Caucasoid, 0.29 +/- 0.02. It has thus been suggested that the ratios of FP to MS contents in the hair may be involved in the determination of the macroscopic racial variations in the form of hair.

A familial occurrence of sporotrichosis.

A grandfather and granddaughter suffering from sporotrichosis were reported. It is thought that they might have been infected from the same source of Sporothrix schenckii (S. schenckii).

Two-dimensional polyacrylamide gel electrophoresis of human hair matrix proteins using isoelectric focusing electrophoresis as the first dimension.

S-carboxymethylated (SCM) matrix proteins from fifteen normal human scalp hair specimens were analyzed using two-dimensional polyacrylamide gel electrophoresis (PAGE) with usual isoelectric focusing electrophoresis as the first dimension and then with usual sodium dodecyl sulfate-PAGE as the second dimension. On the electrophoregrams obtained, one low-mobility smear of SCM proteins embedding one SCM protein band was always seen. Many higher-mobility SCM protein spots were also always seen. However, some variations were seen in these SCM protein compositions; in type I, there were eighteen SCM protein components and, in type II, seventeen components.

Proliferating trichilemmal tumor with apocrine sweat glands.

A 43-year-old man with a proliferating trichilemmal tumor is described. Since the tumor had appeared on a pre-existing alopecia and accompanied ectopic apocrine sweat glands, it is supposed that the pathogenesis of the tumor of our patient might be similar to that of an organoid nevus.

[A case report of spotted fever group rickettsiosis first encountered in Shimane Prefecture, Japan].

Rickettsial infections of a spotted fever group have recently been experience in Japan. Although there have been 50 cases reported by this time, they were exclusively distributed in the areas along the Pacific coast. We report in this paper the first case of spotted fever group rickettsiosis in Shimane prefecture, an area faced to the Japan Sea. A 57 year-old man with high fever, general fatigue and rash was admitted to the hospital of Shimane Medical University at the end of September 1987, who had been treated with cefaclor for a few days without effect before his visit to the University Hospital. Physical examination revealed erythematous eruption, eschar on the left side of the abdomen, and generalized lymphadenopathy. Hepatosplenomegaly was not detected. Laboratory studies showed normal leukocyte counts with relative lymphopenia, high ESR and increased CRP. Transaminase levels were slightly elevated. Paul-Bunnel and Weil-Felix tests resulted in negative. Specific immunofluorescence tests demonstrated that IgG and IgM antibodies of acute-phase serum to Rickettsia japonica, a spotted fever group rickettsia isolated from patients in Japan, were not detected, while both IgG and IgM antibody titers of convalescent-phase serum increased to 1:320. Convalescent-phase serum reacted at significantly lower titers with R.typhi as well as other pathogenic spotted fever group rickettsiae. The patient was treated with ofloxacin for six days and then with combination of ofloxacin and minocycline. It should be noted that clinical symptoms such as high fever and general fatigue disappeared during a single therapy with ofloxacin.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of cutaneous localized cryptococcosis].

A 65-year-old Japanese male with cutaneous localized cryptococcosis, which was developed as an erythematous infiltrated plaque on the right side of the face, was reported. The biopsy specimen taken from the lesion showed a granulomatous change with many spores in the dermis. Culture of the biopsy specimen gave Cryptococcus neoformans serotype D. There were no cryptococcal infections in the other organs. The skin lesion was improved by a systemic administration of miconazole. This patient was unusual since the cryptococcal lesion was limited to the skin, nevertheless he was under a severe cellular immunity deficiency. The reason for this may be that the infected C. neoformans strain belonged to serotype D.

Two-dimensional gel electrophoresis of ribosomal proteins from Propionibacterium acnes and granulosum.

Ribosomal proteins (r-proteins) from a type I strain (ATCC 11827) and a type II strain (ATCC 11828) of Propionibacterium acnes (P. acnes) and from a Propionibacterium granulosum (P. granulosum) strain (ATCC 11829-2) were analyzed using two-dimensional polyacrylamide gel electrophoresis (two-dimensional PAGE), for the purpose of genetical analysis of the P. acnes type I and type II strains and the P. granulosum strain. Among the fifty r-proteins in all, almost all of the proteins were electrophoretically different between both of the two types of P. acnes strains and the P. granulosum strain, whereas only five proteins were different between the type I and type II strains of P. acnes. Such differences of r-protein composition between the type I and type II strains of P. acnes indicate that the type I strain is genetically different from, but very closely related to, the type II strain. However, the P. granulosum strain is not so closely related to both the two types of P. acnes strains. This study thus suggests that the two-dimensional PAGE of r-proteins is an excellent tool for the taxonomy of P. acnes and the related bacteria.

A case of tick-bite from Ixodes ovatus Neumann--a report from west Japan.

A 70-year-old male forestation worker had a tick-bite on the lower abdominal skin. The bite was treated by surgical excision of the tick together with the surrounding skin. The tick was identified as Ixodes ovatus Neumann through investigation of its morphological characteristics. This patient is the fourth case of a tick-bite from Ixodes ovatus Neumann in western Japan.

A case of mosquito hypersensitivity showing abnormality of peripheral monocyte function.

We report here an 11-year-old, Japanese girl with an ulcer-forming type of mosquito hypersensitivity. Since the lesions might have arisen from an Arthus type of hypersensitivity, we used a phorbol myristate acetate (PMA)-triggered chemiluminescence method to investigate the function of her peripheral monocytes, which should play an important role in any Arthus reaction in association with polymorphonuclear leucocytes (PMNs). The chemiluminescence of the patient's monocytes was significantly reduced from that of the normal control. To our best knowledge, no such functional abnormality of the monocytes has been seen previously in patients with an ulcer-forming type of mosquito hypersensitivity.

Comparison of human hair and nail low-sulfur protein compositions on two-dimensional electrophoresis.

Compositions of human normal hair and nail low-sulfur proteins were compared using two-dimensional electrophoresis of their S-carboxymethylated (SCM) derivatives. Six SCM low-sulfur protein components with molecular weights (MWs) of 76,000, 73,000, 72,000, 64,000, 61,000 and 55,000 were common to the hair and nail. One component with a MW of 61,000 was specific to hair, and two components, both with a MW of 50,000, were specific to nail.

Eosinophil-infiltrating folliculitis in childhood--report of a case.

A 5-year-old Japanese girl developed follicular pustules with erythematous plaques on the lower extremities. The histopathology was compatible with eosinophilic pustular folliculitis (EPF). However, the clinical findings were not typical of EPF; the pustules did not appear in crops, the pustules became ulcers, and the skin lesions appeared exclusively on non-seborrheic areas.

Amino acid compositions of human hair fibrous protein components purified with two-dimensional electrophoresis.

Normal human S-carboxymethylated (SCM) hair fibrous protein (HFP) components were purified with two-dimensional electrophoresis, and their amino acid compositions were examined. As previously reported, the SCM cysteine and glycine contents of the crude HFPs were characteristically high and low, respectively, as compared with those reported for the stratum corneum fibrous proteins (SCFPs). However, the SCM cysteine and glycine contents of the purified SCM HFP components were not as high or as low, respectively, but were rather very similar to those of the SCFPs. This suggests that, with respect to cysteine and glycine content, fibrous protein components similar to those of the stratum corneum exist in normal human hair.

Idiopathic calcinosis of the scrotum presenting as a solitary pedunculated tumour.

Idiopathic calcinosis of the scrotum usually develops in the form of scrotal calcified nodules varying in number from 1 to over 100 and from pinhead to walnut in size. In this paper, we describe a case of this skin disorder which had developed as a solitary pedunculated tumour. To the best of our knowledge, no such patient has been previously described.

Hypotrichosis with keratosis pilaris: electrophoretical study of hair fibrous proteins from a patient.

S-carboxymethylated (SCM) fibrous proteins from the scalp hair of a patient with hypotrichosis with keratosis pilaris (HTKP) and from that of a normal individual were analyzed using two-dimensional electrophoresis. One SCM fibrous protein component was different electrophoretically in the HTKP patient. It is suggested that the brittleness of the HTKP hair might result from this alteration of the fibrous protein composition of the hair.
