Myocardial homing and neovascularization by human bone marrow angioblasts is regulated by IL-8/Gro CXC chemokines.

In the adult, new blood vessel formation can occur either through angiogenesis from pre-existing mature endothelium or vasculogenesis mediated by bone marrow-derived endothelial precursors. We recently isolated endothelial progenitor cells, or angioblasts, in human adult bone marrow which have selective migratory properties for ischemic tissues, including myocardium, to where they home and induce vasculogenesis. Here we show that myocardial production of the IL-8/Gro-alpha CXC chemokine family is significantly increased after acute ischemia, and that this provides a chemoattractant gradient for bone marrow-derived endothelial progenitors, or angioblasts. This chemokine-mediated homing of bone marrow angioblasts to the ischemic heart regulates their ability to induce myocardial neovascularization, protection against cardiomyocyte apoptosis, and functional cardiac recovery. Together, our results indicate that CXC chemokines play a central role in regulating vasculogenesis in the adult, and suggest that manipulation of interactions between chemokines and their receptors on autologous human bone marrow-derived angioblasts could augment neovascularization of ischemic myocardial tissue.

[Codon usage of Chinese hamster ovary cells.].

BACKGROUND: To investigate the optimal codons of dihydrofolate reductase (dhfr) minus Chinese hamster ovary cells (CHO dhfr-). METHODS: A cDNA library of CHO dhfr- containing high abundence mRNA was constructed and protein-coding sequences were obtained after identification and analysis. Codon frequence of CHO dhf- was compared with that of Chinese hamster in CUTG database. Then codon usage variation among cDNA was investigated using correspondence analysis (COA). RESULTS: Fifty qualified cDNAs from CHO dhfr- were selected, which encodes proteins of high abundence. Comparing with the codon frequence of Chinese hamster, the highest frequence of synonymous codons for amino acids in CHO dhfr- cells were the same as Chinese hamster except that of Arg and Pro. This method of COA identifies the first main factor which can account for the largest fractions (14.7%) of variation among cDNAs. Twenty-two synonymous codons were identified as the optimal codons of CHO cell. CONCLUSION: CHO dhfr- cell has its own optimal codons, it is suggested that codon bias is one of reasons for functional diversity of different mammal cells and it is an effective stratagy to modification of the codon usage of the foreign gene according to the optimal codons of CHO dhfr- to increase the production of foreign gene.

Determination of acidic and basic species by headspace gas chromatography.

This paper reported a novel headspace gas chromatographic (GC) technique on quantification of acidic and basic species. It is based on an acid-base reaction between the measured species and bicarbonate in an aqueous solution, which generates carbon dioxide in a closed headspace sample vial. By operating at 60 degrees C, carbon dioxide is completely released to the headspace and thus can be measured by GC with a thermal conductivity detector. Bicarbonate concentrations of 0.030 and 0.0025 mol/L are recommended for general applications and very small species content, respectively. This method is able to accurately measure small sample sizes (down to few milligrams or microliters). The present method is simple, accurate, and automatic.

Chemical defenses of Trifolium glanduliferum against redlegged earth mite Halotydeus destructor.

Trifolium glanduliferum Boiss. was resistant to redlegged earth mite, Halotydeus destructor Tucker (Acari: Penthaleidae), feeding compared with Trifolium subterraneum L. Extracts from T. glanduliferum leaves, obtained with dichloromethane and methanol, acted as feeding deterrents to the redlegged earth mite. Bioassay-directed fractionation of the extracts led to the isolation of several metabolites, coumarin, alpha-and beta-ionone, linalool, medicarpin, that were shown to be deterrent in laboratory experiments. The compounds contributing to the defense of T. glanduliferum were different from those influencing mite feeding on T. subterraneum leaves.

Treatment of short gut syndrome with early living related small bowel transplantation.

AIM: To investigate the results of treating short bowel syndrome with an early living related small bowel transplantation (SBT). METHODS: A 17-year-old boy with a 20-cm-long residual intestine due to necrotic volvulus received an early living related SBT from his mother. Donor-specific blood transfusion was performed for 8 weeks before transplantation, each time for 50 mL every week. Cytomegalovirus status in both donor and recipient was negative. A 160-cm distal ileal segment was removed from the donor. The graft ilecolic artery and vein were anastomosed to the recipient's infrarenal aorta and caval vein. The proximal end of the graft was anastomosed end-to-end to the residual recipient jejunum; the distal anastomosis, between the distal end of the graft and transverse colon. An ileostomy was also performed. Immunosuppression, infection prophylaxis, and antithrombotic and nutrition support were given postoperatively. RESULTS: The donor had an uneventful recovery. No technical complications were observed. The recipient was alive and well at 31 weeks after the operation. No graft rejection or infection was observed. He was off TPN 8 weeks after the operation and took low-fat food. The D-xylose test in the recipient was almost normal. CONCLUSIONS: Early living related small intestine transplantation is a good treatment for short bowel syndrome.

The vitellogenin gene of the mosquito Aedes aegypti is a direct target of ecdysteroid receptor.

In the female mosquito Aedes aegypti, vitellogenin (Vg), the major YPP, is activated by 20-hydroxyecdysone (20E) at the transcriptional level. We used cell transfection assays in the Drosophila S2 cells to investigate whether 20E acts directly on the Vg gene via its functional receptor, the heterodimer composed of the ecdysteroid receptor (EcR) and the ultraspiracle (USP) proteins. We demonstrated that the Vg 5'-regulatory region contains a functional ecdysteroid-responsive element (VgEcRE1) that is necessary to confer responsiveness to 20E. VgEcRE binds directly to EcR-USP produced in vitro and extracted from the vitellogenic fat body nuclei. The binding intensity of the EcR-USP-EcRE1 complex from nuclear extracts corresponds to the levels of ecdysteroids and of the Vg transcript during the vitellogenic cycle. Given the modest level of 20E-dependent activation, it is likely that the EcR-USP receptor acts synergistically with other transcription factors to bring about the high level of Vg gene expression.

Primary leiomyosarcoma of bone.

We report a case of primary leiomyosarcoma in the right femoral diaphysis. Conventional radiographs revealed an indistinct osteolytic lesion with a very subtle periosteal reaction. MR imaging revealed an intramedullary lesion with a well-defined margin, cortical breakthrough and soft tissue extension. The lesion was isointense relative to muscle on T1-weighted imaging, iso-hypointense relative to fat on fast spin echo T2-weighted imaging, and hyperintense to muscle on fast spin echo T2-weighted imaging with fat saturation. A peripheral rim and intratumoral spoke-wheel-like enhancement in the soft tissue component of the tumor was noted on gadolinium-enhanced T1-weighted imaging with fat saturation. Such an enhancing pattern, which has not been described in the literature previously but has been seen in leiomyosarcomas elsewhere by both MR imaging and computed tomography, deserves further pathological correlation.

Phonological studies of the new gas-induced agitated reactor using computational fluid dynamics.

An ozone-induced agitated reactor has been found to be very effective in degrading industrial wastewater. However, the cost of the ozone generation as well as its short residence time in reactors has restricted its application in a commercial scale. An innovated gas-induced draft tube installed inside a conventional agitated reactor was proved to effectively retain the ozone in a reactor. The setup was demonstrated to significantly promote the ozone utilization rate up to 96% from the conventional rate of 60% above the onset speed. This work investigates the mixing mechanism of an innovated gas-induced reactor for the future scale-up design by using the technique of computational fluid dynamics. A three-dimensional flow model was proposed to compute the liquid-gas free surface as well as the flow patterns inside the reactor. The turbulent effects generated by two 45 degrees pitch-blade turbines were considered and the two phases mixing phenomena were also manipulated by the Eulerian-Eulerian techniques. The consistency of the free surface profiles and the fluid flow patterns proved a good agreement between computational results and the experimental observation.

Circulating hepatitis B virus nucleic acids in chronic infection : representation of differently polyadenylated viral transcripts during progression to nonreplicative stages.

PURPOSE: Beside the established maturation of hepatitis B virus (HBV) transcripts at a polyadenylation signal downstream of the HBV x protein open reading frame, maturation at an internal polyadenylation signal has been observed in the chronically infected liver. In the present study, it was the aim to identify the respective circulating full-length and truncated transcripts in plasma/serum of carriers. EXPERIMENTAL DESIGN: Nucleic acids extracted from sera were analyzed using established PCR and reverse transcription-PCR procedures targeted to HBV x protein gene regions. Amplification products were cloned and sequenced. RESULTS: Base substitution patterns were determined, which indicated infection stages advanced to different degrees regardless of the transcript type analyzed. HBV full-length RNA (fRNA) showed a high correlation with hepatitis B e antigen and viral DNA, indicative for a replicative infection. In contrast, truncated RNA (trRNA) appeared to be independent of hepatitis B e antigen and showed only a weak association with circulating viral DNA. No correlation was observed between the levels of trRNA and the apparent liver damage as reflected by alanine transaminase levels. An age-dependent representation of fRNA and trRNA was observed: fRNA decreased progressively to low levels, whereas trRNA remained at comparably high values. trRNA and RNA not polyadenylated at either of the two polyadenylation signals were detected even in the absence of any other conventional HBV seromarker, including viral DNA. This was shown for patients with cryptogenic cirrhosis and hepatitis C virus carriers. CONCLUSIONS: The identification of HBV RNA in human serum has a diagnostic potential for apparent and for inapparent infection stages.

Membrane bioreactor process of organic wastewater from brassylic acid manufacturing plant.

The wastewater treatment from brassylic acid manufacturing plant using membrane bioreactor (MBR) was studied. The membrane bioreactor consisted of batch-operation biological aeration tank and ultrafiltration evaluation tank. The content of test included the affection of variation operation conditions on ultrafiltration separation, the general characteristics of MBR process, and the difference comparing with the conventional biological treatment. The results are as follows: (1) among the test membrane material, polyether sulphone (PES) membrane is more suitable for the wastewater treatment; (2) when the cutoff molecular weight is among 10,000-50,000, the higher the cutoff molecular weight, the bigger the water flux is in the test; (3) under the operation pressure, water flux increases accompanying with the increasing of operation pressure; (4) the paper filtered COD concentration has more affection on the water flux than the suspended solid concentration; (5) as the volume loading of MBR increases, the accumulation of high molecule organic substance and colloid increases, the membrane permeate COD concentration and paper filtered COD concentration increase too, meanwhile the water flux reduces; (6) when the sludge retention time of activated sludge of MBR increases, the accumulation of high molecule organic substance and colloid reduces, the membrane permeate COD concentration and paper filtered COD concentration reduce too, and the water flux increases; (7) comparing with the conventional biological process, the microbial activity is higher, but the microbial species is less.

[Genescan analysis of the loss of heterozygosity on the long arm of chromosome 6 in non-small cell lung cancer].

To investigate if there are microsatellite loci in the long arm of chromosome 6 that have close relationship with non-small cell lung cancer, Multiple PCR approach was used to analyze the 36 loci in the long arm of chromosome 6. The PCR products were analyzed in PAGE and then the electrophoresis maps were analyzed with Genescan and Genotyper. There is different LOH frequency in different loci. The total frequency of LOH in 41 lung cancers was 78%(32/41), with the highest frequency of LOH was detected on the locus D6S302(75%). There are 14 loci which have LOH frequency more than 20% and the loci are mainly located in 2 regions: 6q16.3-q21 [6 loci D6S458 (21.43%), D6S1694 (26.92%), D6S1717 (35.71%), D6S1565 (40%), D6S302 (75%), D6S1706 (36.36%) and 6q26-q27 (5 loci D6S1550 (38.46%), D6S264 (20%), D6S1585 (25%), D6S446 (33.33%), D6S281 (30.77%)], There may be tumor suppressor genes located in the two regions, which have a close relationship with non-small cell lung cancer.

[Cloning of HAL1 gene and characterization for salt tolerance tomato].

The HAL1 gene was cloned by PCR strategy and confirmed by sequencing. Its open read frame is 879 bp, encoding a peptide of 294 amino acids (32 kD Protein). A chimaeric construct of HAL1 and Npt II (neomycin phosphotransferase II) was constructed and introduced into commercial cultivars of tomato (Zhong SU No. 5: Lycopersicon escullentum) by Agrobacterium tumefacien-mediated gene transformation. Transformants were selected for their ability to grow and root on media containing kanamycin. Transformation was confirmed by analysis of PCR, Southern blot and RT-PCR. The salt tolerance of transgenic tomato is evaluated by comparing the fresh weight, dry weight, Na+, K+ content of transgenic tomato and control tomato. It is concluded that the over-expressing of HAL1 in tomato could enhance the salt tolerance of the transgenic tomato.

Differential expression and regulation by 20-hydroxyecdysone of mosquito ultraspiracle isoforms.

Ultraspiracle (USP), the insect homologue of the vertebrate retinoid X receptor, is an obligatory dimerization partner for the ecdysteroid receptor (EcR). Two USP isoforms, USP-A and USP-B, with distinct N-termini, occur in the mosquito Aedes aegypti. In the fat body and ovary, USP-A mRNA is highly expressed during the pre- and late vitellogenic stages, corresponding to a period of low ecdysteroid titer, while USP-B mRNA exhibits its highest levels during the vitellogenic period, correlating with a high ecdysteroid titer. Remarkably, 20-hydroxyecdysone (20E) has opposite effects on USP isoform transcripts in in vitro fat body culture. This steroid hormone upregulates USP-B transcription and its presence is required to sustain a high level of USP-B expression. In contrast, 20E inhibits activation of USP-A transcription. Although EcR.USP-A recognizes the same ecdysteroid-responsive elements, EcR.USP-B binds them with an affinity twofold higher than that of EcR.USP-A. Likewise, EcR.USP-B transactivates a reporter gene in CV-1 cells twofold more strongly than EcR.USP-A. These results suggest that USP-B functions as a major heterodimerization partner for EcR during the vitellogenic response to 20E in the mosquito.
