Structural studies of the streptavidin binding loop.

The streptavidin-biotin complex provides the basis for many important biotechnological applications and is an interesting model system for studying high-affinity protein-ligand interactions. We report here crystallographic studies elucidating the conformation of the flexible binding loop of streptavidin (residues 45 to 52) in the unbound and bound forms. The crystal structures of unbound streptavidin have been determined in two monoclinic crystal forms. The binding loop generally adopts an open conformation in the unbound species. In one subunit of one crystal form, the flexible loop adopts the closed conformation and an analysis of packing interactions suggests that protein-protein contacts stabilize the closed loop conformation. In the other crystal form all loops adopt an open conformation. Co-crystallization of streptavidin and biotin resulted in two additional, different crystal forms, with ligand bound in all four binding sites of the first crystal form and biotin bound in only two subunits in a second. The major change associated with binding of biotin is the closure of the surface loop incorporating residues 45 to 52. Residues 49 to 52 display a 3(10) helical conformation in unbound subunits of our structures as opposed to the disordered loops observed in other structure determinations of streptavidin. In addition, the open conformation is stabilized by a beta-sheet hydrogen bond between residues 45 and 52, which cannot occur in the closed conformation. The 3(10) helix is observed in nearly all unbound subunits of both the co-crystallized and ligand-free structures. An analysis of the temperature factors of the binding loop regions suggests that the mobility of the closed loops in the complexed structures is lower than in the open loops of the ligand-free structures. The two biotin bound subunits in the tetramer found in the MONO-b1 crystal form are those that contribute Trp 120 across their respective binding pockets, suggesting a structural link between these binding sites in the tetramer. However, there are no obvious signatures of binding site communication observed upon ligand binding, such as quaternary structure changes or shifts in the region of Trp 120. These studies demonstrate that while crystallographic packing interactions can stabilize both the open and closed forms of the flexible loop, in their absence the loop is open in the unbound state and closed in the presence of biotin. If present in solution, the helical structure in the open loop conformation could moderate the entropic penalty associated with biotin binding by contributing an order-to-disorder component to the loop closure.

Variability of femoral muscle attachments.

Analytical and experimental models of the musculoskeletal system often assume single values rather than ranges for anatomical input parameters. The hypothesis of the present study was that anatomical variability significantly influences the results of biomechanical analyses, specifically regarding the moment arms of the various thigh muscles. Insertions and origins of muscles crossing or attaching to the femur were digitized in six specimens. Muscle volumes were measured; muscle attachment area and centroid location were computed. To demonstrate the influence of inter-individual anatomic variability on a mechanical modeling parameter, the corresponding range of muscle moment arms were calculated. Standard deviations, as a percentage of the mean, were about 70% for attachment area and 80% for muscle volume and attachment centroid location. The resulting moment arms of the m. gluteus maximus and m. rectus femoris were especially sensitive to anatomical variations (SD 65%). The results indicate that sensitivity to anatomical variations should be analyzed in any investigation simulating musculoskeletal interactions. To avoid misinterpretations, investigators should consider using several anatomical configurations rather than relying on a mean data set.

Cancer in Circumpolar Inuit. Background information for cancer patterns in Canadian Inuit.

The cancer pattern among Inuit in the Circumpolar area is remarkably different from those of other populations in the world. The current paper summarizes the most important risk factors in Canadian Inuit residing in the Northwest Territories, northern Quebec (Nunavik) and Labrador, particularly during the time period 1969-1988 covered by the study. Factors considered include: the geographic area and physical environment; population and human environment, including fertility and life expectancy; lifestyle and diet, including tobacco and alcohol use; other lifestyle factors, and health conditions; and health services and cultural accessibility. Development of the cancer registry and population databases supporting the analysis of cancer rates is described. The information in the present paper is needed to interpret cancer incidence patterns and differences among the Circumpolar Inuit of Canada, Alaska and Greenland.

Cancer incidence by ethnic group in the Northwest Territories (NWT) 1969-1988.

Clear differences occurred in the cancer patterns among the population sub-groups in the NWT. When compared to those for the total Canadian population, rates for all cancers tended to be higher than expected among Inuit, lower than expected among Status Indians, and at expected levels for the Other NWT population. Among Inuit, traditional patterns still persist. Cancers of the lung, cervix, nasopharynx and salivary gland, and choriocarcinoma, occurred more often, and cancers of the breast, prostate, uterus and colon less often than in the total Canadian population. Among Status Indians, small numbers precluded definitive conclusions. However, several cancer sites occurred less often than expected, including colon, bladder and prostate among males, and uterus in females. While no cancer was significantly elevated in either males or females, SIRs for cervix and lung were above 1.0 for females, and kidney cancer was significantly higher when data for both sexes were combined (SIR = 2.0). For the Other NWT group--comprising about 50% of the population--most types of cancers occurred at about the expected rate, except that lung cancer was significantly elevated in females. Nevertheless, the generally high rates for lung and cervical cancer, which were particularly evident among the Inuit, are clearly targets for prevention programmes. It is hoped the cancer registry data now available for twenty years for the NWT, as well as for the overall Canadian Inuit population, can be used by researchers for studies to further determine the etiology of cancers, especially where distinctive patterns occur in these populations.

Hypothalamic modulation of growth hormone secretion in the rhesus monkey: evidence from intracerebroventricular infusions of glucose, free fatty acid, and ketone bodies.

To evaluate the hypothalamus as a possible site of metabolic modulation of GH secretion, we studied the GH response to insulin hypoglycemia (IHG) and nicotinic acid (NA)-induced FFA depression in the absence and presence of third ventricular (ivt) infusions of glucose, oleic acid (Ol-Ac), or beta-hydroxybutyrate (beta OHB). Four rhesus monkeys had been prepared for chronic remote iv and ivt infusions as well as blood sampling from the adjacent room. Statistical evaluation used a two-way analysis of variance and individual comparisons with Tukey's Studentized range test. The GH response (area under the curve +/- SE) to IHG was significantly reduced by a concomitant ivt glucose infusion (control, 1.0 +/- 0.1; IHG, 12.1 +/- 3.3; IHG plus ivt glucose, 7.0 +/- 1.2 microgram/L.120 min). The GH response to FFA depression was significantly reduced by ivt Ol-Ac or beta OHB infusion (control, 6.0 +/- 1.0; NA, 51.5 +/- 4.1; Na plus Ol-Ac, 81.2 +/- 1.3; NA plus beta OHB, 38.6 +/- 3.5 microgram/L.300 min). Introcerebroventricular infusions of glucose, Ol-Ac, or beta OHB alone had no effect on plasma GH, glucose, FFA, or beta OHB concentrations. These results provide evidence for a hypothalamic site of metabolic modulation of GH secretion in the rhesus monkey. This does not exclude an additional effect directly at the pituitary gland.

Cancer patterns in the Inuit population of Canada 1970-1984.

1. Rates for lung cancer in Canadian Inuit are high and increasing for both men and women; in fact they are the highest reported rates for lung cancer among any Inuit population. 2. Cervical cancer in Canadian Inuit are high but rates appear to be stable, unlike the rapidly increasing trend reported in both Alaska and Greenland. 3. Rates for traditional Inuit cancers such as those of the nasopharynx and salivary gland do not appear to be declining in Canadian Inuit. 4. Rates for colorectal cancer in Canadian Inuit are similar to those expected for the Canadian population as a whole and do not appear to be increasing. 5. Finally, cancers traditionally reported to be rare in Inuit are still rare in the Canadian Inuit population; these include breast, prostate, bladder, and endometrial cancer. Cancer is a disease which can be controlled through prevention, early detection or treatment. The following future directions arising from this work reflect this paradigm. First, the Inuit cancer registry developed for this analysis will be used as a base for further research into environmental and genetic factors influencing cancer rates in the Inuit. Second, given that at least 40% of cancers in Canadian Inuit are cancers of the lung and cervix, health promotion programs should be developed to encourage tobacco-free environments to halt the epidemic of lung cancer, and also to make available Pap smear programs for early detection of cervical cancer. Third, health care services and programs, including diagnosis and treatment, should be culturally accessible to the Inuit population (6).(ABSTRACT TRUNCATED AT 250 WORDS)

[Bronchobiliary fistula as sequela of the stenosis of hepaticojejunostomy].

This is a case report on a bronchobiliar fistula after frequent recurrences of cholangites following an occlusion of a hepaticojejunostoma. Sufficient drainage and correction of the stenosis as well as balancing of water and electrolyte metabolism were without any therapeutic success. The patient died in consequence of an acute liver dystrophy.

Tissue engineering of articular cartilage under the influence of collagen I/III membranes and low oxygen tension.

The objective of this study was to study the matrix production and phenotype stability of articular chondrocytes cultured on collagen I/III membranes (CM) under the influence of low oxygen tension (Po(2)). Primary bovine and osteoarthritic human chondrocytes were cultured for 2 weeks under 5-21% Po(2) on CM, in alginate, or as monolayers. Dedifferentiated cells were produced by 2-week monolayer culture under 21% Po(2). Collagen (Coll) type II and I expression was demonstrated immunohistochemically, by Western blotting (Coll II), and by semiquantitative RT-PCR; proteoglycan synthesis was demonstrated histochemically (toluidine blue); and biosynthetic activity was indicated by radiolabel incorporation ([(3)H]proline and [(35)S]sulfate). Bovine chondrocytes on CM showed an increase in Coll II expression and proteoglycan synthesis under low Po(2) conditions, whereas Coll I decreased. This oxygen-dependent phenotype-stabilizing effect was even more pronounced in alginate cultures. Biosynthesis of bovine and human chondrocytes was also increased by low Po(2), except for proline incorporation, which decreased in bovine CM cultures (low-oxygen effects were significantly higher in alginate than in CM cultures). Dedifferentiated chondrocytes reexpressed Coll II protein when cultured under low Po(2) on CM or in alginate only, but not under high Po(2) or in monolayer culture. We conclude that CM and, even more, alginate foster phenotype stability and cartilage-specific matrix production of bovine chondrocytes, especially when cultured under in vivo-like oxygen conditions.
