Human apo-lipoprotein B from normal plasma contains oxidised peptides.

Oxidation of low density lipoprotein (LDL) may be atherogenic, but radical-initiated oxidation of its apoprotein B-100 (apoB) has been little studied. Transition metal ions iron and copper are candidates for mediating radical oxidation of LDL in vivo. Therefore, we studied the copper-ion-induced oxidation of apoB in human LDL. Using HPLC methods developed in our recent work, we studied the destruction of native and the generation of six oxidised amino acids; we also assessed the release of peptides from the LDL particle by FPLC. We observed time-dependent losses of apoB histidine, lysine and glycine. Long-lived reactive species, the reductant DOPA, and the oxidant hydroperoxides of valine and leucine (measured as hydroxides after reduction), were generated. Their relative abundance (mol/mol of parent amino acid) was DOPA > o- and m-tyrosine > dityrosine, valine-hydroxides, leucine hydroxides. Low molecular weight fragments were also released from the LDL in a time-dependent manner, contained hydroperoxides sensitive to GSH peroxidase, and generated radicals on reaction with iron-EDTA. The fragments contained peptides active in the quinone redox cycling procedure, comprising 0.25% of the supplied LDL amino acids. Characteristic peptides were present in each FPLC fraction containing the fragments, as judged by further HPLC fractionation. Some fragments were present in the unoxidised LDL preparations, and when these were largely removed by FPLC, copper oxidation could still generate fragments, suggesting that those present in the starting material might indicate prior oxidation. Concordantly, we found that fresh plasma LDL apoB contained 43% of total plasma protein-bound oxidised amino acids, and with the same relative abundance. We conclude that plasma proteins including apoB are subject to physiological oxidation, similar to that inflicted by copper ions; the latter may contribute to intimal LDL oxidation, which could be the source of oxidised plasma apoB.

Augmentation of phenylbutyrate-induced differentiation of myeloid leukemia cells using all-trans retinoic acid.

Despite preliminary evidence of clinical activity of the putative differentiating agent sodium phenylbutyrate (PB) in the treatment of myeloid neoplasms, it has proven difficult to maintain therapeutic levels of PB above 0.5 mM, well below the ED50 of 1-2 mM. We have studied the impact of combining PB with all-trans retinoic acid (ATRA) on the ML-1 myeloid leukemia cell line. ATRA augmented PB-induced differentiation, cell-cycle arrest, and apoptosis. ATRA augmented PB induction of the myelomonocytic marker CD11b at all doses of ATRA tested (0.0025-1 microM). Although ATRA did not significantly affect the ED50 of PB, the combination of ATRA (1 microM) and PB (0.5 mM) augmented PB-induced CD11b expression eight-fold. Compared to PB alone, this combination of ATRA and PB induced greater cell cycle arrest (S-phase 14% vs 38%; G0/G1-phase cells 72% vs 52%) and greater apoptosis (24% vs 16% by TUNEL assay). Treatment with ATRA (0.5 microM) in combination with PB (0.5 mM) led to significantly greater inhibition of colony formation (4.8% vs 48% inhibition). ATRA combined synergistically with PB to augment CD11b expression and inhibit colony formation. This combination also showed significant interaction in terms of S-phase inhibition. However, this interaction varied as a function of ATRA concentration: antagonistic at low concentrations of ATRA, synergistic at higher concentrations of ATRA. These data suggest that retinoids may significantly augment the cytostatic and differentiating activity of PB, leading to increased potency of the latter drug at clinically achievable doses.

Phenylbutyrate-induced G1 arrest and apoptosis in myeloid leukemia cells: structure-function analysis.

The aromatic fatty acid phenylbutyrate (PB) induces cytostasis, differentiation, and apoptosis in primary myeloid leukemic cells at clinically achievable concentrations. In the present study, we have investigated the structural and cellular basis for PB-induced cytostasis, using the ML-1 human myeloid leukemia cell line as a model system. PB induced a dose-dependent increase in cells in G1 with a corresponding decrease in cells in S-phase of the cell cycle. At comparable doses, PB induced expression of CD11b, indicating myeloid differentiation. At higher doses, the drug induced apoptosis. The antitumor activity was independent of the aromatic ring, as butyric acid (BA) was of equal or greater potency at producing these biological changes. In contrast, shortening of the fatty acid carbon chain length, as demonstrated with phenylacetate (PA), significantly diminished drug potency. Consistent with their effects on cell cycle, PB and BA, but not PA, induced the cyclin-dependent kinase inhibitor, p21(WAF1/CIP1), and led to the appearance of hypophosphorylated Rb, suggesting a role for p21(WAF1/CIP1) in PB-induced cytostasis. Therefore, it appears that the fatty acid moiety of PB, rather than its aromatic ring, is critical for its activity in myeloid leukemic cells. These data provide a potential mechanistic basis for the increased potency of PB over PA previously demonstrated in primary leukemic samples, and support the further clinical development of PB in the treatment of hematologic malignancies.

Are reactive oxygen species involved in the pathogenesis of murine cerebral malaria?

To investigate the involvement of oxidative tissue damage in the pathogenesis of murine cerebral malaria (CM), brain levels of protein carbonyls, 3,4-dihydroxyphenylalanine (DOPA), o-tyrosine, and dityrosine were measured during Plasmodium berghei ANKA (PbA) and P. berghei K173 (PbK) infections. During PbA infection in a CM model, brain levels of the substances were similar to those in uninfected mice. The role of phagocyte-derived reactive oxygen species in the pathogenesis of CM was examined in gp91phox gene knockout mice. The course of CM in these mice was the same as in their wild type counterparts. To examine whether superoxide production in the central nervous system could have occurred via increased xanthine oxidase activity, brain concentrations of urate were measured in CM mice and in mice infected with PbK (which does not cause CM). Brain urate concentration increased significantly in both groups of mice, suggesting that purine breakdown is not specific to CM. These results indicate that reactive oxygen species probably do not contribute to the pathogenesis of murine CM.

Absence of the derived allele at the DYS199 locus in 3 NE Chinese populations.

The allelic states of DYS199 locus were studied by allele-specific PCR. We found that all of the 107 samples chosen from three populations of northeast China had DYS199 C alleles. The result excluded the possibility that this mutation first identified in native American populations came from the three populations of northeast China.

[A study on polymorphism of mitochondrial DNA D loop in the Han nationality in China].

OBJECTIVE: To investigate the polymorphism of mitochondrial DNA D loop in the Han nationality in China and the mean nucleic acid substitute frequency on each site. METHODS: A 465bp-long DNA fragment in mtDNA D loop was amplified by PCR technique and the polymorphism was analyzed by restriction fragment length polymorphism (RFLP) using 6 restriction enzymes (on 10 sites). Some samples were analyzed by direct sequencing of PCR products. RESULTS: RFLP analyses with 6 restriction enzymes on the 10 sites generated 14 restriction types. The calculated mean nucleic acid substitute frequency (based on Nei formula) on each site of mtDNA D loop in the Han nationality in China was 0.029. CONCLUSION: The mean nucleic acid substitute frequency on each site in the D loop region was 10 times as high as that in mtDNA.

[The relationship between apolipoprotein E gene polymorphism and coronary heart disease and arteriosclerotic cerebral infarction].

OBJECTIVE: To detect the relationship between ApoE gene polymorphism and coronary heart disease(CHD), arteriosclerotic cerebral infarction(ACI). METHODS: Exon 4 of ApoE gene was analyzed in normal group(87 cases), CHD group(78 cases) and ACI group(55 cases) by a combination of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and direct AgNO(3) staining technique. RESULTS: Between the control group and CHD group, ACI group, chi(2) test showed no significant difference in genotype of E3/E3 (P>0.5, P>0.1, respectively); while mu test showed no significant difference in allele epsilon(2)(P>0.25, P>0.1, respectively), epsilon(4)(P>0.1, P>0.25, respectively) of ApoE. CONCLUSION: No relationship between ApoE gene polymorphism and CHD and ACI was noted in this study.

[Mutation analysis of p16 gene in non-small cell lung carcinomas].

OBJECTIVE: To investigate the role of p16 gene in the tumorigenesis of non-small cell lung carcinomas (NSCLC). METHODS: Homozygous deletion and mutation of exon 2 of p16 genes in 40 NSCLC tissues were analyzed using PCR and dsDNA direct sequencing technique. RESULTS: In 2 NSCLC tissues, homozygous deletions of p16 gene were detected; in 14 NSCLC tissues, 19 point mutations and a frameshift were detected. CONCLUSION: Point mutations of p16 gene were frequent and might play a role in the initiation and progression of NSCLC. Nt380 in this gene was a hot spot of mutation.

Oxalate: from crystal formation to crystal retention.

Idiopathic calcium oxalate stone formation is a multifactorial disease. It is therefore unlikely that a single underlying condition will be responsible for entire spectrum of the disease; however, it appears that one important factor in the pathogenesis is an abnormality in oxalate metabolism. Whatever the cause, two critical parameters for stone formation are crystal formation and crystal retention in the renal tubules. Although crystal formination and role of oxalate in crystal formation have been evaluated extensively, it is only recently that crystal retention has been addressed. Previous studies from our laboratories demonstrated that oxalate exposure to renal epithelial cells in culture resulted in initiation of a program of events including DNA synthesis and cell death. The present studies evaluated effects of oxalate on cell proliferation and damage to distal tubular (Madin-Darby canine kidney cells) and proximal (LLC-PK1 cells) cells. Effects of oxalate exposure on calcium oxalate monohydrate (COM) crystal adherence to these cells were also evaluated. Results presented herein demonstrate that proximal tubular cells are more sensitive to oxalate than distal tubular cells. Furthermore, oxalate exposure to proximal tubular cells resulted in reinitiation of DNA synthesis, whereas no such effect was observed in distal tubular cells. Higher levels of oxalate (> 1 mM) resulted in cell loss of both proximal and distal tubular cells, as observed by crystal violet staining. Despite these differences, oxalate exposure to both proximal and distal tubular cells resulted in increased COM crystal adherence. Thus, oxalate exposure may promote crystal adherence to renal epithelial cells either secondarily to cell death and proliferation or by a yet unidentified mechanism. These studies provide the first direct evidence for the role of oxalate in promoting COM crystal retention by the urothelium.

[Comparative genomic hybridization analysis of primary gastric carcinomas].

OBJECTIVE: To investigate whether unknown genes are involved in the tumorigenesis of gastric cancer. METHODS: Fourty-three primary gastric carcinomas were analyzed by comparative genomic hybridization(CGH). RESULTS: A gain in chromosome 3p(8/43), 8q(8/43), 20[20(9/43), 20p(7/43), 20q(4/43)], 12q (16/43), and 13q(12/43) was observed while a loss of 19[19(15/43), 19p (13/43)], 17[17(8/43), 17p(10/43)], 16(10/43) and 1p(11/43) was discovered. CONCLUSION: There were characteristic changes in 3p, 8q, 20, 12q, 13q, 19, 17, 16, and 1p in gastric carcinoma, and some unknown genes located in the above regions might be of importance to gastric carcinoma pathogenesis.

[An anatomical study of the nasolabial fold region].

OBJECTIVE: This study of the nasolabial fold is to provide the morphological basis for maxillofacial plastic and cosmetic surgery. METHODS: Twenty fresh adult cadavers' heads were dissected with microsurgical technique and the structure of the nasolabial region was observed and measured. RESULTS: 1. The data about the length, width and thickness of each mimetic muscle were obtained and presented in the paper. 2. The medial and lateral subcutaneous fat of the nasolabial groove was 1.3 mm and 4.5 mm in thickness respectively. 3. There existed muscle fiber attachments in the medial dermis of nasolabial groove, and in the lateral dermis there were only a few muscle bundle attachments. 4. Among the superficial facial muscles there were not only fasciae but also aponeuroses. The muscles, fasciae and aponeuroses composed a three-dimensional reticular structure. CONCLUSION: This study gave a further support to the theoretical hypothesis of SMAS intermediate tendon.

Factors involved in the neuronal death during postischemic reperfusion: experimental study in rabbits.

OBJECTIVE: To explore the main pathogenic factors in the development of neuronal death during normothermic reperfusion in rabbits. METHODS: Ninety-six New Zealand rabbits were randomly allocated into two groups: group I served as non-ischemic controls; group II served as postischemic normothermic reperfusion models. Complete cerebral ischemia was induced by the four-vessel model for 30 minutes. After ischemia, rabbits in group II were further divided into three subgroups according to the duration of reperfusion: subgroup A, 30 minutes; subgroup B, 180 minutes and subgroup C, 360 minutes. Twenty-eight biochemical parameters in the brain were measured, and neuronal changes were observed by histomorphological assessment. Neurons of 12 regions were differentiated into four types: type A (normal), type B (mildly damaged), type C (severely damaged) and type D (necrotic). Bivariate correlate analysis between the levels of biochemical parameters and the percentages of each type of neurons was carried out. RESULTS: The main parameters involved in the progressive decrement of type A neurons were VIP, beta-EP, PGI2, T3, T4 and Na+, K(+)-ATPase; in the increment of type B were beta-EP and TXB2; in the increment of type C were GLU and TXB2/PGI2 respectively; in the stepwise increment of percentages of type D neurons were T4, Na+, K(+)-ATPase, GLU, T3 and VIP (P < 0.05). CONCLUSION: The main factors involved in the development of neuronal death during postischemic normothermic reperfusion in rabbits include hypermetabolism, deactivation of Na+, K(+)-ATPase, release of excitatory amino acids and disorder of neuropeptides.

Correlation of cell proliferation inhibition and apoptosis induction with expression of human beta 5 integrin on hematopoietic cells.

OBJECTIVE: To investigate the function of the alpha v beta 5 integrin in hematopoietic cells. METHODS: Tissue culture, integrin expression vectors, gene transfer, polymerase chain reaction (PCR), apoptosis analyses and cytometic analysis were made on hematopoietic cells. RESULTS: The beta 5 integrin cDNA was not expressed in hematopoietic cells following exposure to the beta 5 integrin retrovirus vector pG beta 5CHT. Unbalanced expression of the alpha v beta 3 and alpha v beta 5 integrins occurred during apoptosis induced by serum depletion and upon differentiation. The treatment of hematopoietic cells with anti-alpha v beta 5 monoclonal antibody inhibited apoptosis induced by serum depletion. Inducible expression of the beta 5 integrin cDNA in the hematopoietic cell line K562 caused cellular proliferation inhibition. CONCLUSION: The alpha v beta 5 integrin cDNA in hematopoietic cells can inhibit the proliferation of the hematopoietic cell, cause the differentiation of the hematopoietic cells and induce the apoptosis of the hematopoietic cells.

[A novel human DNA sequence with tumor metastasis suppressive activity].

OBJECTIVE: To isolate human tumor metastasis suppressive DNA sequence and to study the molecular mechanisms regulating tumor metastasis. METHODS: A mouse lung adenocarcinoma cell clone 12 derived from its parent cell line LM2, which had been transduced with normal human genomic DNA, was previously reported. Compared with LM2, the metastatic potential of clone 12 was very much decreased. Clone 12 was used in this study to amplify the human DNA fragments by Inter Alu PCR technique. The human DNA fragments obtained were then transfected into LM2 cells and their malignant phenotype was observed in vitro and in vivo, and compared with that of the untransfected LM2 cells. RESULTS: Three human DNA fragments of 700, 500 and 300 bp were isolated. DNA sequencing revealed that the 700 bp fragment does not show homology with hitherto reported genes and was accepted by the GenBank (pt 712 U67835). In vitro proliferation and colony formation in soft agar of the 700 bp fragment-transfected LM2 cells were significantly inhibited as compared to the untransfected LM2 cells. Upon subcutaneous inoculation to syngeneic T739 mice, the 700 bp-transfected LM2 cells grew more slowly and smaller tumors developed compared to the un-transfected ones. Moreover, lung metastasis was not found in 6 of 10 mice inoculated with the 700 bp-transfected LM2 cells, while it was found in 9 of 10 mice inoculated with the un-transfected LM2 cells. The difference was statistically significant (P < 0.001). The frequency of lymph node metastasis was also statistically different between the 2 groups of mice. CONCLUSION: The newly isolated 700 bp human DNA fragment may be a metastasis suppressor gene of malignant tumor.

[Biomechanical experimental study on effective fraction of radix salviae miltiorrhizae on healing of bone fracture].

OBJECTIVE: To assess the effect of Danshen-9403 (DS-9403, an effective fraction of Radix Salviae Miltiorrhizae) on healing of bone fracture. METHODS: Standardized radial fracture was performed in 120 Wistar rats. The model animals were randomized into four groups: group A was fed with DS-9403, group B injected with Staphylococcus Aureus, group C with normal saline, and group D with Salviae injection. The treatment began at the first day of fracture. The rats were sacrificed on the 25th, 39th and 50th day separately in batches and their intact radii were removed by dissection for detecting load and stress of three-point bending test with autograph universal material testing machine (Shimazu, Japan). RESULTS: The parameters of load in DS-9403 treated groups on the 39th day and that of stress at 25th, 39th and 50th day were (6.20 +/- 1.32) N, (5.71 +/- 3.58) N/mm2, (8.27 +/- 2.42) N/mm2 and (66.25 +/- 26.21) N/mm2 respectively, which were significantly higher than those in other groups, P < 0.05. CONCLUSION: DS-9403 has the action of increasing the strength of fracture healing site.

[Studies on relationship between occupation and pregnancy outcome].

OBJECTIVE: To study the effects of maternal occupation on reproductive outcomes, including pregnancy outcome and neonatal status, and complications of pregnancy and delivery. METHODS: Data were extracted from 12 224 obstetric records of eight hospitals in Beijing, and all the subjects recruited in the study were interviewed before discharge during from May 1992 to April 1993. Pregnancy and reproductive outcomes were analyzed according to their occupational classifications, adjusted for the potential confounding factors, such as age, smoking and drinking during pregnancy and history of spontaneous abortion. RESULTS: Incidence rates of pregnancy induced hypertention, low birth weight, intrauterine growth retardation and postpartum hemorrhage were higher in the workers than that of the average levels. Incidence rates of threatened abortion, gestational anemia, stillbirth and uterine atony in professional persons, of prolonged pregnancy and neonatal asphyxia in farmers, of low birth weight in housewives, of gestational anemia and of uterine atony in administrative persons all were higher than the average levels. CONCLUSION: Occupation undertaken by pregnant women was associated with the adverse pregnancy and reproductive outcomes to certain extent.

[Effect of growth period, storage time and varieties on the contents of main active constituents of Curcuma longa L. in rhizome].

OBJECTIVE: To provide criteria for selection, storage and cultivation of the rhizome of Curcuma longa. METHOD: Field observation and sample analysis on the rhizome. RESULT: The contents of curcuminoids and essential oils in mother and daughter rhizome went up with the progress of plant development. The contents of curcuminoids reached the maximum in early September and early October. The contents of essential oils in mother rhizome reached the maximum in early September. The contents of curcuminoids and essential oils in the rhizome varied with the species and went down with the increase of storage years. CONCLUSION: The rhizome should be collected when leaves of the plant have just withered. The suitable storage time is three years.
