Amino acid sequences of double-headed proteinase inhibitors from the seeds of Canavalia lineata.

The amino acids of two Bowman-Birk type proteinase inhibitors (CLTI-I and -II) from the seeds of Canavalia lineata were sequenced by a manual Edman degradation using the DABITC/PITC double coupling method after enzymatic digestions with Achromobacter lyticus lysyl endopeptidase, Staphylococcus aureus V8 protease, and chymotrypsin. CLTI-I contains 75 amino acid residues. CLTI-II has an identical sequence to CLTI-I except an extra Asp residue attached at the C-terminus. The inhibitors showed a homology (40-70%) to other Bowman-Birk inhibitors. The reactive-site peptide bonds were estimated to be Lys21-Ser22 and Leu48-Ser49 against trypsin and chymotrypsin, respectively. An inhibitory active fragment containing only the chymotrypsin-reactive site was also described.

Syalil-SSEA1(SLX) levels in supernatant of cultured human lung carcinoma cell lines.

SLX levels in the culture supernatant of the following 50 cell lines were measured by RIA: pulmonary carcinoma cell lines derived from 46 patients, cell lines of other human cancers derived from 4 patients, and 2 passaged human fibroblast cells as control. Of the 46 pulmonary carcinoma cell lines, 17 (37%) were SLX positive. When the SLX-positive rate was analyzed in relation to the histological type of pulmonary carcinoma, the positive rate was 71% (10/14) for adenocarcinoma, 27% (3/11) for squamous cell carcinoma, 33% (2/6) for large cell carcinoma, 0% (0/11) for small cell carcinoma and 50% (2/4) for adenosquamous cell carcinoma. Analysis of the relationship between tumor cell proliferation and SLX level in 20 patients revealed that the SLX level in the supernatant of SLX-producing cell lines becomes higher in proportion to the increase in the number of these cells. The SLX-positive rate did not differ significantly among different stages of pulmonary carcinoma at the time of tissue collection. There was no significant correlation between SLX production and prognosis. SLX production by each cell line was not correlated with the doubling time of the same cell line in vitro or in vivo (in nude mice). SLX production also showed no correlation with the duration of tumor cell passage.

Experimental study of lung transplantation from non-heart-beating donor following brain death in canine model of left lung allotransplantation.

Thirty mongrel dogs were divided into 3 groups. Group I was a normal control group (n = 4) without transplantation procedure. Group II was a transplantation control group (n = 5) with left lung allotransplantation from heart-beating donor. Group III consisted of animals (n = 8) which received lung allografts from non-heart-beating donor following brain death. In Group III, brain death was brought about by intracranial hypertension with the inflation of balloon in the subdural space of donor. After 6 hours, the mechanical ventilation was discontinued followed by cardiac arrest. Left lung was excised twenty minutes after the cardiac arrest and was washed out with cold Ep4 solution for subsequent orthotopic allotransplantation. Right pulmonary arterial occlusion test (RPAO) were carried out under right thoracotomy to evaluate graft function immediately and 7 to 14 days after the surgery. PaO2, PAP and CO were measured before and 20 minutes after RPAO. All animals in Group II and 7 of 8 animals in Group III survived more than 7 days after surgery. No significant differences in the value of PaO2, mean PAP and TPVR with RPAO among three groups at each time of assessment, showing the possibility of lung transplantation from non-heart-beating donor followed by brain death.

Identification of a rat protein tyrosine phosphatase gene preferentially expressed in the embryonal brain.

We have identified a novel protein tyrosine phosphatase gene from an embryonal rat brain by reverse transcription-based polymerase chain reaction. Its transcription is specific to brain and is developmentally regulated, as it is expressed at high levels in embryonal and neonatal stages but scarcely in an adult. Southern blot analysis indicates that the gene exists as a single copy and is conserved among various species. Homology search of the deduced amino acid sequence suggests that this gene belongs to the same family of the membrane-type tyrosine phosphatase gene of Drosophila (DPTP10D), whose expression is specific to an central nervous system of fly embryo.

Expression of protein tyrosine phosphatase genes in the developing brain of mouse and rat.

We have examined the expression of protein tyrosine phosphatase genes in the embryonic brain of mouse and rat with reverse transcription-polymerase chain reaction. Several receptor and cytoplasmic types of tyrosine phosphatase genes were detected. Among them a novel gene was identified from mouse and rat brain, respectively. The partial amino acid sequences reveal that the new genes found in the developing brain of mouse and rat are homologous each other. Since they retain conserved phosphatase sequences, they may represent a family of protein tyrosine phosphatase gene that is commonly expressed in rodent brain.

[Effects of Pseudomonas aeruginosa on transport of protein and liquid from alveolar spaces in anesthetized rabbits].

We determined the effects of Pseudomonas aeruginosa on transport of protein and liquid across the alveolar epithelium in anesthetized rabbits. Following the injection of 3 microCi of 131I-albumin in anesthetized rabbits as a vascular marker, we instilled 3 ml/kg of 5% albumin solution with 10(7) or 10(9) CFU P. aeruginosa, 3 microCi of 125I-albumin and 3 mg of Evans blue into the alveolar spaces of the right lung. The rabbits were ventilated with 60-100% oxygen and 1-2% halothane over 8 h. We removed the lungs and aspirated the alveolar fluid 8 h after instillation. We measured protein concentration in instilled and aspirated alveolar fluid, and radioisotope counts in alveolar fluid, homogenized lung and plasma. Then we calculated the influx and efflux of radioisotope across the alveolar epithelium. Liquid clearance from the alveolar spaces and the lungs was calculated using protein concentration of alveolar fluid and extravascular lung water. In the experiments with 10(9) CFU P. aeruginosa, we found that 125I-albumin remaining in the lungs decreased and 125I-albumin in plasma increased. Lung liquid clearance decreased by 15%. However, protein and liquid clearance in the experiments with 10(7) CFU P. aeruginosa or autologous serum were not different from those in the control experiments. P. aeruginosa was cultural from blood, right pleural effusion, and liver in the experiments with 10(9) CFU P. aeruginosa, but not in the experiments with the alveolar epithelium, but decreases clearance of liquid from the alveolar spaces.(ABSTRACT TRUNCATED AT 250 WORDS)

[Effect of amurinone on human pulmonary artery in vitro].

We studied the effect of amurinone, a phosphodiesterase inhibitor, on human pulmonary arterial smooth muscle in vitro. Amurinone caused dose-dependent relaxation of pulmonary arterial strips precontracted with 60 mM KCl. Preincubation with either meclofenamate (31 microM), a cyclooxygenase inhibitor, or L-NG-nitroarginine (100 microM), an endothelium-derived relaxing factor (EDRF) production inhibitor, failed to inhibit amurinone-induced pulmonary vasodilation. The cyclic AMP (cAMP) levels in the supernatant of the lung vessel homogenates significantly increased after incubation with amurinone. These results indicate that amurinone causes relaxation of human pulmonary artery in vitro, and suggest a role for cAMP in the mechanisms of amurinone-induced pulmonary vasodilation.

[Flow cytometric analysis of nuclear DNA content of lung cancer--correlation between nuclear DNA content and tumor doubling time].

A total of 65 resected cases with primary lung cancer were examined on correlation between nuclear DNA content by flow cytometry and other prognostic factors, including tumor doubling time (DT). 1) DNA diploidy was found in 18 cases (28%) and DNA aneuploidy in 47 cases (72%). Percent aneuploidy was 79.5% in cases with adenocarcinoma, which tended to be higher than that in cases with squamous cell carcinoma (57.1%). 2) Percent aneuploidy in cases with advanced carcinomas tended to be higher than that in cases with earlier carcinomas, and percent aneuploidy in cases with rapid growing carcinomas tended to be higher than that in cases with slow growing carcinomas. 3) The geometric mean of DT of DNA aneuploid tumor was 130.0 day, which tended to be shorter than that of DNA diploid tumor (171.9 days), but the difference was not significant. There was not a significant correlation between DT and DNA Index. 4) The five-year survival rate of 47 patients with DNA aneuploid tumor was 45%, which was significantly lower than that of 18 patients with DNA diploid tumor (78%).

[Assessment of nucleolar organizer regions (Ag-NORs) in primary lung cancer--correlation between Ag-NORs and tumor doubling time].

A total of 86 resected cases with primary lung cancer were examined on relationships between argyrophil nucleolar organizer regions (Ag-NORs) and other prognostic factors and correlation between Ag-NORs and tumor doubling time (DT). Survival rates were compared between patients with low Ag-NOR counts and patients with high Ag-NOR counts. 1) After logarithmic conversion of mean Ag-NOR counts in lung cancer, a small skewness (0.00486) and a small kurtosis (-0.7859) showed a normal distribution. Mean Ag-NOR counts was found to have a log-normal distribution. 2) There was a significantly inverse correlation between mean Ag-NOR counts and DT (correlation coefficient -0.705, p < 0.001). By plotting log (DT) on X axis and log (Ag-NORs) on Y axis, a formula representing a linear correlation was obtained: Y = 1.17-0.312 X, correlation coefficient -0.886. 3) The five-year survival rate (35%) of 46 patients with mean Ag-NOR counts which were more than or equal to 3.0 significantly lower than that (74%) of 40 patients with mean Ag-NOR counts of less than 3.0.

[Lung transplantation from non-heart-beating donor following brain death in canine model].

Candidates for pulmonary transplantation have been limited because of extreme susceptibility to lung infections and pulmonary edema in the brain-dead donor. To ameliorate the donor shortage, two possibilities for expanding the donor source have been presented, i.e. xenotransplantation and transplantation from cardiac-dead donors. The present study was conducted to evaluate the possibility of lung transplantation from a non-heart-beating donor following brain death, using a canine model. Six mongrel dogs were put into a state of brain death by elevating intracranial pressure with a balloon catheter. Following the intravenous administration of methylprednisolone and heparin used for 6-hour management following brain death mechanical ventilation was discontinued leading to cardiac arrest with in a few minutes. Excision of the left lung was scheduled for twenty minutes after cardiac arrest, followed by washing out the pulmonary vasculature with cold Ep4 solution, and orthotopic transplantation into the recipient animal. Immunosuppression was achieved with methylprednisolone and azathioprine. A right pulmonary arterial occlusion test (RPAO) was performed to assess graft function immediately and 7 days postoperatively. All but one animal survived and three animals had an uneventful postoperative course, with the transplants alone according to immediate and 7 day postoperative, respectively, RPAO results. These outcomes indicate the possible feasibility of lung transplantation from non-heart-beating donors following brain death.

[Lung 3H-thymidine incorporation in monocrotaline (MCT)-treated rats--role of platelet-activating factor (PAF)].

Monocrotaline (MCT) causes chronic pulmonary hypertension associated with pulmonary vascular thickening in rats. Since components of the pulmonary vascular thickening are reflected in increased DNA synthesis in medial smooth muscle cells, and since platelet-activating factor (PAF) has been reported to contribute to the pulmonary hypertension induced by MCT, we examined the effect of WEB 2170, a specific PAF receptor blocker, on MCT-induced pulmonary vascular thickening and in vitro 3H-thymidine incorporation into lung tissue, as an index of stimulation of cell proliferation. In MCT-treated rats, pulmonary hypertension, right ventricular hypertrophy, and pulmonary vascular thickening developed at 3 weeks after injection. Also, in MCT-treated rat lung tissue, there was a significant increase in the in vitro 3H-thymidine incorporation rate. In WEB 2170-treated MCT rats, these changes were significantly less severe than those observed in rats receiving MCT alone. We conclude that PAF plays a role in the inflammatory process that contributes to the development of pulmonary hypertension and vascular remodeling associated with increased lung cell proliferation in MCT-treated rats.

[A case of bronchial neurinoma treated with bronchofiberscopic electrosurgery and right S6 sleeve segmentectomy].

A 30-year-old woman was admitted for cough and fever over a period of two months. Her chest X-ray film showed atelectasis of the right middle and lower lobes. Bronchofiberscopic examination revealed complete obstruction of the truncus intermedius by polypoid tumor. Its punched biopsy specimen was diagnosed as the benign tumor. Bronchofiberscopic resection was performed by high frequency snare, however the tumor remained at the membranous portion of the truncus intermedius and the right B6 orifice. Therefore, right S6 sleeve segmentectomy was carried out after two weeks. The pathological diagnosis of the exturpated tumor was neurinoma. There were no complications and she is well without any evidence of recurrence after surgery. Neurinoma of the tracheobronchial tree is extremely rare and only 84 cases have been reported.
