The challenge of the so-called electron configurations of the transition metals.

Quite different meanings are attached by chemists to the words element, atom, orbital, order of orbitals or configurations. This causes conceptual inconsistencies, in particular with respect to the transition-metal elements and their atoms or ions. The different meanings will here be distinguished carefully. They are analyzed on the basis of empirical atomic spectral data and quasi-relativistic density functional calculations. The latter are quite reliable for different average configuration energies of transition-metal atoms. The so-called "configurations of the chemical elements", traditionally displayed in periodic tables, are the dominant configurations of the lowest spin-orbit levels of the free atoms. They are chemically rather irrelevant. In many-electron systems the ns and np AOs are significantly below the more hydrogen-like nd ones. Even (n+1)s is below nd for all light neutral atoms from C onwards, but only up to the first elements of the respective long rows! The most common orbital order in transition-metal atoms is 3p << 3d < 4s etc. The chemically relevant configuration in group g is always d(g) instead of d(g-2) s(2). Conceptually clear reasoning eliminates apparent textbook inconsistencies between simple quantum-chemical models and the empirical facts. The empirically and theoretically well-founded Rydberg (n-deltal) rule is to be preferred instead of the historical Madelung (n+l) rule with its large number of exceptions.

The electrochemical corrosion of bulk nanocrystalline ingot iron in acidic sulfate solution.

The corrosion properties of bulk nanocrystalline ingot iron (BNII) fabricated from conventional polycrystalline ingot iron (CPII) by severe rolling were investigated by means of immersion test, potentiodynamic polarization (PDP), electrochemical impedance spectroscopy (EIS) tests, and scanning electron microscopy (SEM) observation. These experimental results indicate that BNII possesses excellent corrosion resistance in comparison with CPII in acidic sulfate solution at room temperature. It may mainly result from different surface microstructures between CPII and BNII. However, the corrosion resistance of nanocrystalline materials is usually degraded because of their metastable microstructure nature, and the residual stress in nanocrystalline materials also can result in degradation of corrosion resistance according to the traditional point of view.

Preparation and electrochemical corrosion behavior of bulk nanocrystalline ingot iron in HCl acid solution.

Bulk nanocrystalline ingot iron (BNII) was produced by the severe rolling technique. The corrosion behaviors of BNII and as-received conventional polycrystalline ingot iron (CPII) in 1 M HCl solution were investigated by potentiodynamic polarization tests, electrochemical impedance spectroscopy measurement, and immersion tests at room temperature. For BNII, the anodic dissolution process is inhibited, but the cathodic process is enhanced. The corrosion current and average corrosion rate of BNII are 0.479 and 0.391 those of CPII, respectively. The resistance of the charge transfer of BNII is about 1.59 times higher than that of CPII. These results indicate that the corrosion resistance of BNII is improved in comparison with CPII.

DNA biosensors based on self-assembled carbon nanotubes.

DNA biosensors based on self-assembled multi-walled carbon nanotubes (MWNTs) were described in this paper, in which the probe DNA oligonucleotides were immobilized by forming covalent amide bonds between carboxyl groups at the nanotubes and amino groups at the ends of the DNA oligonucleotides. Hybridization between the probe and target DNA oligonucleotides was confirmed by the changes in the voltammetric peak of the indicator of methylene blue. Our results demonstrate that the DNA biosensors based on self-assembled MWNTs had a higher hybridization efficiency compared to those based on random MWNTs. In addition, the developed DNA biosensors also had a high selectivity of hybridization detection.

A novel multi-walled carbon nanotube-based biosensor for glucose detection.

The bioelectrochemical characteristics of a novel multi-walled carbon nanotube (MWNT)-based biosensor for glucose detection are studied and compared with those of glassy carbon (GC)-based biosensor. The MWNT-based biosensor exhibits a strong glucose response at applied potentials of 0.65 and 0.45 V versus Ag/AgCl, respectively, while GC-based biosensor shows a weak glucose response at 0.65 V and no response at 0.45 V. Besides, the MWNT-based biosensor shows a high stability of 86.7% of the initial activity to glucose after four-month storage, much higher than 37.2%, the corresponding value for a GC-based biosensor. The detection mechanism of the MWNT-based biosensor is also discussed in detail.

Effect of DBP/DEHP in vegetable planted soil on the quality of capsicum fruit.

Field experiment was conducted to investigate the di-n-butyl phthalate (DBP) and di-2-ethylhexyl phthalate (DEHP) contamination in Capsicum annum fruit grown in DBP and DEHP contaminated soil, and to evaluate the effect of DBP and DEHP on the quality of capsicum fruit. The top layer soil (0-10 cm) of plots was treated with a mixture of DBP and DEHP (1:1 w/w) and capsicum seedlings were transplanted. After 90 days, capsicum fruit, shoot and root samples were collected. DBP and DEHP concentration in various parts of the samples were determined by gas chromatography. Vitamin C and capsaicin contents in fruit were determined using 2,4-dinitrophenylhydrazine colorimetric analysis and sodium nitrite-sodium molybdate colorimetric analysis, respectively. The results showed that DBP concentration in fruit, shoot and root increased with the increase of soil-applied DBP/DEHP concentration, but DEHP was not detected in all samples. When the soil-applied DBP/DEHP concentration was 5, 10, 20, 40, 80 and 160 mg kg(-1) soil, compared with control, vitamin C and capsaicin content in capsicum fruit decreased by 1.6%, 5.9%, 10.6%/o, 18.2%, 19.2%, 22.6% and 1.6%, 2.5%, 12.9%, 20.1%, 22.2%, respectively. Pearson correlation analysis demonstrated that the decrease of vitamin C and capsaicin content was negatively correlated to the increase of DBP concentration in capsicum fruit, which suggested that DBP uptake by the plant might be mainly responsible for quality degradation of capsicum fruit.

Expression of telomerase activity in cholesteatoma otitis media.

Telomerase maintains the length of telomeres in immortal cells and is also often associated with cell proliferation. Cholesteatoma epithelium is characterized by a dysregulation with hyperproliferative growth. The study evaluated the telomerase activity in cholesteatoma and normal retro-auricular skin to discover the relationship between telomerase expression and clinical findings. Twenty-two samples of cholesteatoma and 15 samples of retro-auricular skin were obtained from patients undergoing middle-ear surgery. The telomerase activity was detected by the telomerase repeat amplification protocol (TRAP) assay method. Seventeen of the 22 (77.3 per cent) cholesteatoma cases expressed telomerase activity, whereas none of the 15 retro-auricular normal skin (0 per cent) detected telomerase activity. There was no significant difference between telomerase expressions and clinical findings, including hearing level, duration of disease, and the degree of extension (p>0.05). The high expression of telomerase in cholesteatoma suggests that the activation of telomerase may be related to the proliferative nature of cholesteatoma.

[Study on improvement of cell affinity of polymer materials--modified poly(D,L-lactide) by anhydrous ammonia gaseous plasma].

OBJECTIVE: To modify the surface of poly(D,L-lactide) film by anhydrous ammonia gaseous plasma treatment. METHODS: The changes of contact angles were measured and surface energy were calculated. Mouse 3T3 fibroblast cells were cultured on plasma modified and control poly(D,L-lactide) films. RESULTS: It was found that the hydrophilicity and surface energy of the materials have been increased after plasma treatment. Cell culture results showed that ammonia plasma treatment could promote the cell attachment and cells growth. After 4 days culture, the cells on the plasma treated films were 2-folds quantitatively compared with that of the control films. CONCLUSION: Ammonia plasma treatment can improve the cell affinity to poly(D,L-lactide).

[Research and application of drug controlled release technique in tissue engineering].

OBJECTIVE: To investigate protection of biological activity and controlled release of growth factor by means of drug controlled release technique in tissue engineering. METHODS: Using drug controlled release technique that to embed or microcapsulate the biological drug with biodegradable polymer. RESULTS: The aliphatic polylactone could be used as drug carrier for each drug including the biological matter. And the release behavior of the drug could be controlled by adjusting the molecular structure of the carrier and the controlled release method. The successful example, that to realize regeneration of rat's sciatic nerve with 5, 10, 15 and 20 mm of gap by using polylactide as nerve guide and the embedding growth factor, had been obtained. CONCLUSION: It is possible to realize protection of biological activity and sustained release of growth factor by using aliphatic polylactone as drug carrier.

A hybrid between Na+,K+-ATPase and H+,K+-ATPase is sensitive to palytoxin, ouabain, and SCH 28080.

Na(+),K(+)-ATPase is inhibited by cardiac glycosides such as ouabain, and palytoxin, which do not inhibit gastric H(+),K(+)-ATPase. Gastric H(+),K(+)-ATPase is inhibited by SCH28080, which has no effect on Na(+),K(+)-ATPase. The goal of the current study was to identify amino acid sequences of the gastric proton-potassium pump that are involved in recognition of the pump-specific inhibitor SCH 28080. A chimeric polypeptide consisting of the rat sodium pump alpha3 subunit with the peptide Gln(905)-Val(930) of the gastric proton pump alpha subunit substituted in place of the original Asn(886)-Ala(911) sequence was expressed together with the gastric beta subunit in the yeast Saccharomyces cerevisiae. Yeast cells that express this subunit combination are sensitive to palytoxin, which interacts specifically with the sodium pump, and lose intracellular K(+) ions. The palytoxin-induced K(+) efflux is inhibited by the sodium pump-specific inhibitor ouabain and also by the gastric proton pump-specific inhibitor SCH 28080. The IC(50) for SCH 28080 inhibition of palytoxin-induced K(+) efflux is 14.3 +/- 2.4 microm, which is similar to the K(i) for SCH 28080 inhibition of ATP hydrolysis by the gastric H(+),K(+)-ATPase. In contrast, palytoxin-induced K(+) efflux from cells expressing either the native alpha3 and beta1 subunits of the sodium pump or the alpha3 subunit of the sodium pump together with the beta subunit of the gastric proton pump is inhibited by ouabain but not by SCH 28080. The acquisition of SCH 28080 sensitivity by the chimera indicates that the Gln(905)-Val(930) peptide of the gastric proton pump is likely to be involved in the interactions of the gastric proton-potassium pump with SCH 28080.

Preliminary study of oral polylactide microcapsulated insulin in vitro and in vivo.

AIM: Although the oral route for insulin delivery is the most convenient, directly administered oral insulin is degraded by proteolytic enzymes in the gastrointestinal (GI) tract. Polylactide was prepared in order to microcapsulate the insulin to avoid the enzymes in the GI. The physical characteristics and therapeutic possibilities of polylactide microcapsulated insulin (PLA-MCI) were studied in vivo and in vitro. METHODS: PLA-MCI was prepared by the two-step method of emulsion and solvent extraction. Its morphologic character was observed by scanning electron microscopy (SEM). The insulin release profile was determined in vitro by insulin measurement and in vivo by blood glucose measurement after the force-feeding of 66 diabetic rats. RESULTS: When the microcapsule was spherical in shape (diameter 1.5-2.0 microm) the entrapment efficiency of insulin was 90% and the loading rate was 10% (W/W). The PLA-MCI (which contained 3.0 units of insulin/mg of PLA) had peak release rates of 65-74% over 6-8 h in phosphate buffer. The same dose of PLA-MCI (insulin 2.5 mg) led to decreased responses (from 28% to 68% of control blood glucose levels) in the level of blood glucose in 32 rats which had not fasted after they had been force-fed. When 1.2, 1.8, 2.2 and 3.0 mg of insulin + PLA-MCI was administered to eight diabetic rats, their blood glucose levels decreased by 28%, 36%, 54% and 78%, respectively. CONCLUSIONS: PLA microcapsules are capable of protecting insulin from degradation by the proteolytic enzymes in the GI and of alleviating hyperglycaemia for a prolonged period of time in diabetic rats. It may therefore be considered as a new carrier for oral insulin.

Topical inhalant steroid (budesonide, Pulmicort nasal) therapy in intubation granuloma.

Intubation granuloma of the larynx is an iatrogenic disease which is induced by endotracheal intubation. It has basically been managed by conservative medical treatment with observation. Surgical excision can be considered as a last resort due to the high recurrence rate which subjects the patients to repeated anaesthesia. The purpose of this study is to evaluate the therapeutic effect of topical steroid in intubation granuloma, comparing the results of conservative medical treatment with, or without, surgery (Group I, 14 patients) and inhalant therapy with topical budesonide (Group II, 20 patients). In Group I, complete disappearance of granuloma occurred in six cases within a year (42.8 per cent) with conservative therapy only. Microlaryngeal surgery was performed on the eight cases of persisting granuloma after conservative therapy for a year, resulting in two cases of recurrence. In Group II, the granuloma disappeared completely in 85 per cent within six months and in 95 per cent within 12 months without any remarkable side-effects. We concluded that intubation granuloma of the larynx could be treated with topical inhalant steroid as the first choice of therapy rather than other medical treatment or surgical intervention.

Valine 904, tyrosine 898, and cysteine 908 in Na,K-ATPase alpha subunits are important for assembly with beta subunits.

A 26-amino acid sequence in an extracellular loop of the Na,K-ATPase alpha subunit between membrane-spanning segments 7 and 8 has been shown to bind to the beta subunit of Na,K-ATPase and to promote alphabeta assembly (Lemas, M. V., Hamrick, M., Takeyasu, K., and Fambrough, D. M. (1994) J. Biol. Chem. 269, 8255-8259) When this 26-amino acid sequence of the rat Na,K-ATPase alpha3 subunit was replaced by the corresponding sequence of the rat gastric H,K-ATPase alpha subunit, the chimeric alpha subunit assembled preferentially with the rat gastric H,K-ATPase beta subunit (Wang, S.-G., Eakle, K. A., Levenson, R., and Farley, R. A. (1997) Am. J. Physiol. 272, C923-C930). In the present study, these 26 amino acids (Asn886-Ala911) of rat Na,K-ATPase alpha3 were replaced by the corresponding amino acids Asn908-Ala933 of rat distal colon H, K-ATPase. Site-directed mutagenesis of the chimeric alpha subunits and Na,K-ATPase alpha3 showed that Val904, Tyr898, and Cys908 in the Na,K-ATPase alpha3 subunit are key residues in alphabeta subunit interactions. The V904Q mutation in Na,K-ATPase alpha3 reduced the Bmax for ouabain binding and the ATPase activity of alpha3beta1 complexes by approximately 95%, and Y898R reduced the Bmax and ATPase activity by approximately 60%. The complementary mutations Q904V and R898Y increased the amount of ouabain bound by yeast membranes expressing the chimera with the colon H,K-ATPase sequence. The amount of ouabain bound by complexes assembled between Na, K-ATPase alpha3 containing the Y898R,C908G mutations and gastric H, K-ATPase beta was less than 10% of wild type Na,K-ATPase alpha3 expressed with the same beta subunit. The R898Y,G908C mutations in the chimeric alpha subunits also increased ouabain binding.

Na+-K+-ATPase alpha-subunit containing Q905-V930 of gastric H+-K+-ATPase alpha preferentially assembles with H+-K+-ATPase beta.

Amino acids N886-A911 of the rat Na+-K+-ATPase alpha3-subunit were replaced by the corresponding region (Q905-V930) of the rat gastric H+-K+-ATPase alpha-subunit. The chimera (NGH26) was expressed in yeast with the rat Na+-K+ -ATPase beta1-subunit (rbeta1), the rat H+-K+-ATPase beta-subunit (HKbeta), the chimeric beta-subunit NHbeta1 (containing the carboxy-terminal ectodomain of HKbeta), or the chimeric beta-subunit HNbeta1 (containing the carboxy-terminal ectodomain of rbeta1). Increased resistance to trypsin digestion indicated that NGH26 preferentially assembled with HKbeta and NHbeta1 rather than with rbeta1 or HNbeta1. Ouabain binding also indicated that more functional complexes were assembled when NGH26 was expressed with HKbeta or NHbeta1. These results suggest that the sequence Q905-V930 interacts with the HKbeta-subunit on the extracellular side of the cell membrane. The NGH26 + HKbeta complex is less stable than alpha3 + HKbeta when heated and also has a lower binding affinity for ouabain [dissociation constant (Kd) = 63 nM] compared with alpha3 + rbeta1 or alpha3 + HKbeta (K(d) = 5-10 nM). In contrast, the NGH26+NHbeta1 complex is thermally as stable as alpha3 + rbeta1 complexes, and its ouabain binding affinity (K(d) = 10 nM) is the same as the wild type. These results indicate that the amino acids Q905-V930 of the rat gastric H+-K+-ATPase alpha-subunit preferentially associate with the extracellular domain of H+-K+-ATPase beta-subunit to form functional pump complexes and that the cytoplasmic and/or transmembrane region of the beta-subunit influences the stability of the alpha beta complexes.

The influence of beta subunit structure on the stability of Na+/K(+)-ATPase complexes and interaction with K+.

Heterologous expression of the beta subunit of H+/K(+)-ATPase (HK beta) with alpha subunits of Na+/K(+)-ATPase (NK alpha) in yeast leads to the formation of ouabain binding complexes, indicating assembly of the two subunits into active ion pumps (Eakle, K. A., Kim, K. S., Kabalin, M. A., and Farley, R. A. (1992) Proc. Natl. Acad. Sci. U. S. A. 89, 2834-2838). Complexes of NK alpha and HK beta are less sensitive to inhibition of ouabain binding by K+, suggesting that HK beta lowers the affinity of K+ binding sites. This effect is particularly pronounced when HK beta is combined with the alpha 3 isoform of NK alpha. In this case, titration with K+ yields a biphasic curve, suggesting that there are two nonequivalent sites for K+ binding. Attempts at purifying complexes formed with either alpha 1 + HK beta or alpha 3 + HK beta using SDS extraction of microsomal membranes resulted in the loss of ouabain binding. Controls show that alpha 1 + beta 1 and alpha 3 + beta 1 complexes still retain ouabain binding after SDS extraction under the same conditions. This suggests that the HK beta subunit forms a less stable complex with NK alpha subunits. We have created chimeric beta subunits comprised of the amino-terminal cytoplasmic and transmembrane regions of HK beta combined with the carboxyl-terminal extracellular region of Na+/K(+)-ATPase beta 1 (HN beta 1) and the complementary chimera with amino-terminal cytoplasmic and transmembrane regions of beta 1 combined with the carboxyl-terminal extracellular region of HK beta (NH beta 1). When NH beta 1 is combined with either alpha 1 or alpha 3, the complexes show profiles of K+ inhibition of ouabain binding that are very similar to HK beta combined with either alpha 1 or alpha 3. The data suggest that the extracellular region of HK beta is primarily responsible for the effect on apparent K+ affinity. When the HN beta 1 subunit is expressed with the alpha 3 subunit, less than 5% of the amount of ouabain binding complexes are formed compared with HN beta 1 + alpha 1. This observation suggests that the HN beta 1 subunit either assembles poorly or forms an unstable complex with alpha 3. After SDS extraction, complexes of alpha 1 + NH beta 1 and alpha 3 + NH beta 1 retain ouabain binding, while alpha 1 + HN beta 1 complexes are sensitive to SDS extraction.(ABSTRACT TRUNCATED AT 400 WORDS)

[Observation on blood flow changes in 34 cases of progressive systemic scleroderma treated with Chinese herbal medicine].

The blood flow change of 34 progressive systemic scleroderma (PSS) patients were examined. The amplitude proved to be lowered markedly than healthy subjects. All patients were treated with the basic prescription of PSS as the principal method, combined with infusion of Mailuoning injection in 500 ml of 5% glucose. The course of treatment lasted three months to one year. The result of treatment showed that the abnormal blood flow of extremities of all patients were improved remarkably. Marked improvement rate and total effective rate were 70.5% and 100% respectively. Significant improvements in clinical and laboratory parameters were observed. It revealed that there was a close relationship between the occurrence and development of PSS and blood circulation. It is assumed that the pathogenic mechanism of PSS is Deficiency of vitality and Excess of pathogenic factor (Stasis of Blood), and the Qi tonifying and Blood activating, hard lump softening and mass dissolving medicinal herbs according to Syndrome Differentiation of TCM should be used.

[Surgical treatment of contusive dislocation of the lens].

54 of 60 consecutive cases of contusive lens dislocation (incomplete 45 cases and complete 15 cases) were treated surgically by (1) loop delivery of the lens in 39 cases, (2) double needle fixation (Barraquer's method) in 4 cases, (3) intracapsular cryo- or silicon ball extraction in 5 cases, (4) extracapsular cortical aspiration in 3 cases, and (5) VISC lensectomy and vitrectomy in 3 cases. Surgical treatment is indicated in cases of (1) lens dislocation into the anterior chamber or incarcerated in the pupil; (2) secondary glaucoma induced by pupil block; (3) secondary uveitis; (4) visual impairment due to turbid or eccentric lens; and (5) presence of other complications that require surgical intervention, such as retinal detachment.
