A case of Sweet's syndrome with early gastric cancer.

The case of a 49-year-old man with Sweet's syndrome (acute febrile neutrophilic dermatosis) is described, who had coincidently gastric cancer. The therapeutic effectiveness of colchicine is also presented as one treatment for Sweet's syndrome, because rapid regression of the lesions was noticed.

[Radiation-induced malignant skin tumors in the hands of five doctors].

Radiation-induced malignant skin tumors in the hands in 3 surgeons, one physician, and one dentist are reported. The latent period of the irradiation ranged between 16 years and 34 years. Clinically, small keratotic lesions were seen in 4 doctors and a large tumor the remaining doctor. On histological examination, one of the tumors was a squamous cell carcinoma, 3 were carcinomas in situ, and the fifth was a basal cell carcinoma with a squamous cell carcinoma. While the occupational hazard of developing malignant skin tumors following irradiation have decreased recently, this paper emphasizes precautions should be taken when diagnosing and caring for cancer patients.

Serodiagnosis of cancer, using porcine antigens recognized by human monoclonal antibody, HB4C5.

Antigens, recognized by human monoclonal antibody (HB4C5) generated from a lung cancer patient, were found to occur in porcine pancreas. The antigens-I and -II were purified from crude trypsin of porcine pancreas, only by Mono Q column chromatography, and were eluted at 260 and 300 mM NaCl in 10 mM Tris-HCl buffer, pH 7.4, respectively. These antigens differed from trypsin in molecular weight, elution pattern from the Mono Q column, and their reactivity with HB4C5. The molecular weights of the two antigens were almost the same at around 35000. These were used for serodiagnosis with an assay system based on 96-well immunoplates. The reactivities of antigens-I and -II with various sera were similar. When the reactivity of IgG in serum with antigen-II was measured, absorbance at 415 nm in the case of normal and lung cancer patients was 0.178 +/- 0.056 and 0.492 +/- 0.136 (p less than 0.005). The rates of positive reaction in ovary, larynx, uterus, lung and liver cancers were more than 50%, but the rates in stomach and breast cancers were less than 30%. Positive reaction was hardly detected in pancreas cancer and normal controls.

[Detection of Chlamydia trachomatis antigen for diagnosis of respiratory infection I. Cross reaction with bacteriae consisting normal flora respiratory tract].

We examined the cross reaction of Chlamydia trachomatis (C. trachomatis) by antigen detection method using EIA against the aerobic bacteria which colonize in the respiratory tract of children. Chlamydiazyme showed a cross reaction among 5 out of 7 species of Gram negative bacteria (= 71.4%) and Neisseria and Branhamella catarrhalis showed a cross reaction even in low concentrations of 1 X 10(3-4) CFU/ml. There were no Gram positive bacteria which showed cross reaction. IDEIA Mark III did not react to Gram negative bacteria even in high concentrations of 1 X 10(7-10) CFU/ml. One strain of coagulase positive staphylococcus (CPS) showed positive at a concentration of 1 X 10(8): however, other Gram positive bacteria including two other strains of CPS exhibited no cross reaction. The detectable concentration of C. trachomatis (D/UW 3 strain) using IDEIA Mark III was 1 X 10(3) IFU/ml. Low cross reaction rate and sensitivity suggest IDEIA Mark III is preferable for diagnosis of respiratory infection of C. trachomatis though further clinical studies are necessary.

Immunohistological characterization of human monoclonal antibody against lung cancer.

Using immunoperoxidase staining method, a human monoclonal antibody (MAb), termed HB4C5, which was generated by fusion of NAT-30 cells with regional lymph node lymphocytes from a lung cancer patient, was examined for tumor specificity. The determinant recognized by HB4C5 MAb was well preserved in usual formalin-fixed paraffin-embedded tissue sections. Experiments with four major histological types of the lung cancer showed that HB4C5 MAb reacted with adenocarcinoma (19/27), squamous cell carcinoma (2/15), and large cell carcinoma (4/6), but not with small cell carcinoma (0/7). The immunoreaction was seen in the cytoplasm. In addition, this MAb also reacted with some gastric and colon adenocarcinomas. Further study on normal lung and nonpulmonary tissues revealed that HB4C5 MAb bound only type II pneumocytes and proximal tubules of normal kidney.

Specificity of anti-polynucleotide monoclonal antibodies from human-human hybridomas.

Nine human-human hybridoma clones, secreting monoclonal antibodies reactive with nucleic acids, were generated by fusing with lymphocytes of lung cancer or systemic lupus erythematosus patients. These hybridoma antibodies were classified into 5 types, in terms of reactivities with DNA, RNA, various synthetic nucleic acids and cardiolipin. Hybridoma clone SU-1 secreted antibody reacting with dsDNA, ssDNA and RNA (type I). Clone HL-321 did not react with these, but with poly (dT), poly (I) and poly (G) (type II). Clone HL-349 was reactive with almost all nucleic acids tested and also with cardiolipin (type III). Clones HF-4, HF-7, HB-7 and HL-259 reacted with ssDNA, poly (A), poly (G) and cardiolipin, but not with RNA (type IV). HB-5 and SH-9 antibodies were reactive only with poly (dT) (type V).

Recombinant vaccinia virus LC16m0 or LC16m8 that expresses hepatitis B surface antigen while preserving the attenuation of the parental virus strain.

Thymidine kinase-negative recombinant vaccinia virus LC16m0 or LC16m8 expressing hepatitis B surface antigen preserved almost the same pathogenicity as their parental thymidine kinase-positive attenuated virus strains. The results demonstrate the potential for using the LC16m0 or LC16m8 virus strain as a vector for recombinant vaccine.

Genetic analysis of vaccinia virus Lister strain and its attenuated mutant LC16m8: production of intermediate variants by homologous recombination.

We prepared vaccinia virus variants by introducing part of the HindIII D fragment of the DNA of the parental Lister (LO) strain (temperature-resistant and forming large plaques and pocks) into the attenuated LC16m8 strain (temperature-sensitive and forming small plaques and pocks) by the use of a homologous recombination technique in vivo. Special attention was paid to the HindIII D fragment, since this fragment has an extra XhoI site in LC16m8 which is absent from LO. After HindIII D of LO was introduced as a calcium phosphate precipitate into rabbit kidney (RK13) cells which had been infected with LC16m8, five virus variants (LOTC-1 to LOTC-5) forming much larger plaques than LC16m8 were cloned. In LOTC-2, LOTC-4 and LOTC-5, the introduction of at least part of HindIII D of LO into the corresponding HindIII D region of the LC16m8 genome was apparent as judged by the disappearance of the XhoI site, whereas variants LOTC-1 and LOTC-3 retained the site. The biological characteristics of all the LOTC variants were similar to each other. Their plaque size and pock size were similar to those of LO, whereas they were rather akin to LC16m8 with regard to temperature sensitivity and neurovirulence. The present results strongly suggested that part of the HindIII D fragment was involved in determining biological characteristics affecting plaque size and pock size, but had little influence on temperature sensitivity and neurovirulence.

HO-323, a human B-lymphoblastoid cell line useful for making human-human hybridomas.

A 6-thioguanine-resistant human B-lymphoblastoid cell line, HO-323, was isolated for making human-human hybridomas with high efficiency. Fusions with peripheral blood lymphocytes of systemic lupus erythematosus (SLE) patients and lymphocytes isolated from lymph nodes of lung and breast cancer patients yield constantly more than one hybridoma clones per 10(5) HO-323 cells plated. HO323 cells also fused with lymphocytes from normal peripheral blood to give hybridomas in the same efficiency. The HO-323 cells were diploid with 46 chromosomes and non-secretors of immunoglobulins. This parent cells doubled every 15 hr and could proliferate in serum-containing medium, even if they were plated at low cell density of less than 10(3) cells/ml. The cells could grow in serum-free medium as well as in serum-containing medium, and the resultant human-human hybridomas could also grow in the same media.

Incorporation of isoleucine into protein by a soluble fraction from spermatozoa.

A heat-labile, non-dialyzable factor(s) in soluble fractions from porcine, bull, rabbit and cock spermatozoa was found to incorporate the radioactivity of [14C]isoleucine into a 95 degrees C CCl3COOH-insoluble fraction. The incorporation required ATP, Mg2+, casein and 2-mercaptoethanol. Trypsin and alpha-chymotrypsin inhibited the incorporation, while RNase A and DNase I did not. A mixture of 19 amino acids other than isoleucine had no effect on the incorporation. The reaction product was identified as protein. The incorporated moiety was the isoleucyl moiety of isoleucine and it retained a free alpha-amino group in the product protein. Some other characteristics of this incorporation are also described.
