Basic helix-loop-helix protein sequences determining differential inhibition by calmodulin and S-100 proteins.

Basic helix-loop-helix (bHLH) proteins are a group of transcription factors that are involved in differentiation and numerous other cellular processes. The proteins include the widely expressed class A bHLH proteins (E proteins) and the tissue-specific class B proteins. Previous studies have shown that calmodulin can inhibit the DNA binding activity of certain E proteins but not their heterodimers with class B proteins. Here we show that calmodulin binds to the DNA-interacting basic sequence within the bHLH domain of E proteins. The strength of the binding of bHLH proteins to calmodulin correlates directly with the calmodulin sensitivity of their DNA binding. The basic sequence of MyoD, a class B protein, can also interact with calmodulin. This interaction, however, is blocked by MyoD sequences directly N-terminal of the basic sequence. We further demonstrate that S-100 proteins can interact with and differentially inhibit the DNA binding of bHLH proteins through interaction with the basic sequence. Both the binding to the basic sequence and the effect of the directly N-terminal sequence vary for different S-100 proteins and bHLH proteins. The results suggest the involvement of both calmodulin and S-100 proteins in the differential regulation of bHLH proteins.

Comparison between intraluminal multiple electric impedance measurement and manometry in the human oesophagus.

Conventional oesophageal manometry and intraluminal electrical impedance measurement were simultaneously applied in eight healthy volunteers to study the effect of wet and semisolid bolus viscosities on oesophageal motility and bolus transit. Contraction front velocity measured by electrical impedance and manometry were identical for wet and semisolid swallows and highly associated. Bolus front velocity as measured by electrical impedance was significantly faster than contraction front velocity in both wet and semisolid swallows. Bolus front velocity during semisolid swallows was significantly slower compared to wet swallows. It is concluded that intraluminal electrical impedance measurement is a reliable technique to detect oesophageal motility as well as to differentiate between transit of wet and semisolid bolus consistencies.

Tetrasomy 18p de novo: parental origin and different mechanisms of formation.

We have used eight PCR-based DNA polymorphisms to determine the parental origin and mechanisms of formation in 9 patients with de novo nonmosaic tetrasomy 18p. The 9 patients, 4 girls and 5 boys, had clinical features characteristic of i(18p) syndrome. The supernumerary marker chromosome was identified by fluorescence in situ hybridization (FISH) analysis using centromeric probes and a flow-sorted 18p-specific library. The isochromosome was of maternal origin in all 9 cases. The formation of tetrasomy 18p cannot be explained by a single model. In 6 cases, meiosis II nondisjunction, followed by subsequent postzygotic misdivsion, and in 1 case postzygotic nondisjunction and postzygotic misdivision were the most likely mechanisms of formation. Alternative mechanisms are suggested in the remaining 2 cases.

Chronic diphenyl hydantoin encephalopathy in mentally retarded children and adolescent with severe epilepsy.

An account is given of chronic diphenyl hydantion (DPH) encephalopathy in 21 mentally retarded epileptics with increasing psychomotor deterioration, choreiform hyperkinesia, deposits of immunoglobulins in the skin, and changes in serum immunoglobulins. Three months after withdrawal of DPH the condition proved partially reversible, from the clinical as well as laboratory point of view. Eleven patients have been followed for 1 year after discontinuation of DPH, and the findings were largely unchanged from the 3-month follow-up examination. Before the drug was withdrawn, seven patients exhibited deposits of immunoglobulins at the dermo-epidermal junction and in vessel walls. At the end of 1 year such deposits were found in only three patients, all of whom were on another antiepileptic drug.

[Morpho-histology of the maxilla in cleft palate].

The author's morphohistological study on 5 maxilla specimens without clefts from neonates and on 1 specimen with a median cleft of the palate was designed to detect possible differences which might be of importance to the anatomical and functional rehabilitation of the cleft patient. The results obtained permit to draw the following conclusions: 1. The changes confirmed in the structures of the hard and soft tissues exert, in themselves, no unfavourable effects on the surgical closure of the cleft palate if the date of operation and the operating method are chosen with regard to the general primary underdevelopment of the cleft maxilla. 2. An anatomically correct "reconstruction" of the musculature of the soft palate cannot be expected from the closure of the cleft. By avoiding extended scars, it is possible to inhibit further negative effects on from and function. 3. The functionally "inferior" musculature in the region of the soft palate needs intensive postoperative activation to achieve the closure of the nasopharynx which is necessary for undisturbed speech.

Calcium regulation of basic helix-loop-helix transcription factors.

The basic helix-loop-helix (bHLH) family of transcription factors is essential for numerous developmental and growth control processes. The regulation of bHLH proteins occurs at many levels, including tissue specific expression, differential oligomerization and DNA binding specificities, interaction with negatively acting HLH proteins and post-translational modifications. This review focuses on what is emerging as another level of bHLH protein regulation, calcium regulation through interaction with Ca2+ loaded calmodulin and S-100 proteins. The mechanism and implications of these Ca2+ regulated interactions are discussed.
