Immunoreactive ACTH/beta-endorphin neurons in the tubero-infundibular hypothalamus of rats.

In the preinfundibular portion of male rat hypothalamus, the arcuate-median eminence region was examined after staining with anti-porcine ACTH 1-39 serum. In several cases, anti-beta-endorphin serum was also employed. Both sera stain the same cell bodies. The cell bodies of the immunoreactive neurons are scattered in the subependymal layer, arcuate nucleus and lateral tuberal region. Fibers originating from these cell bodies are distributed extensively throughout these regions. They make synaptic contacts on immunonegative fibers in the arcuate nuclei, or terminate directly in the perivascular space in the internal layer of the median eminence. The findings suggest that the ACTH/beta-endorphin neuronal system may serve neuronally and humorally as an intrahypothalamic constituent of the hypothalamic pituitary regulation system.

Establishment of a human renal pelvic cancer cell line producing tissue thromboplastin and plasminogen activator.

A new epithelial cell line derived from undifferentiated carcinoma of human renal pelvis, designated KP 1, was established in vitro. The cell line has been passaged 190 times in vitro for 5 years and 9 months. The predominant cell in KP 1 was a tear-drop-shaped cell. Doubling time of the cell line was 35 h. The malignant epithelial character of this line was verified by carcinogenicity in the subcuticular layer of nude mice and by karyotypic analysis which revealed the cells to be completely aneuploid with a model chromosome number in the hypertriploid range. KP 1 cells were shown to produce both tissue thromboplastin and plasminogen activator which was immunologically identical to urokinase, the plasminogen activator in urine.

Human renal cell carcinoma: establishment and characterization of two new cell lines.

Characterization studies have been carried out on 2 cell lines (KPK 1 and KPK 13) established from human renal adenocarcinoma. KPK 1 and KPK 13 have been passaged 178 times in vitro for about 6 years and 7 months and 78 times for about 3 years an 2 months, respectively. Although morphologic differences exist between the 2 lines, each has an epithelial morphology and exhibits multilayering. Doubling time of KPK 1 and KPK 13 cells was 29 hours and 51 hours, respectively. Both KPK 1 and KPK 13 induced tumors at the site of subcutaneous injection, closely resembling the original tumor from which they were derived. Chromosome number of both cell lines was 100 per cent aneuploid and the presence of Y chromosomes was confirmed by G banding in KPK 13 cells. KPK 1 was found to have high thromboplastic and high fibrinolytic activities, whereas KPK 13 was shown to have comparatively low thromboplastic and no detectable fibrinolytic activities. These activities were detected in the serum free supernatant fraction from KPK 1 cells but were not detected in that from KPK 13 cells.

Immunohistochemical demonstration of LHRH-neurons in young rat hypothalamus: light and electron microscopy.

Localization and ultrastructural appearance of immunoreactive LHRH neurons were studied in 2-, 8- and 21-day-old rats. The lower half of the diencephalon including the septo-preoptic area was serially cut at 10 microns in the frontal plane, and all sections stained with an anti-LHRH serum were examined light microscopically. Although the antiserum could not demonstrate LHRH-immunoreactive perikarya in adult rats, they were stained in these young rats. The immunoreactive perikarya were scattered in the following regions: nucleus tractus diagonalis, medial septum, medial preoptic area, lateral septum, lateral preoptic area, tuberculum olfactorium, suprachiasmatic area, supraoptic area, pericommisural area, anterior commissure, vascular organ of the lamina terminalis (OVLT), lateral hypothalamic region and lateral tuberal region. The first three regions contained many cells but others showed only sporadical cells. The cells were most frequently observed in 8-day-old rats. Most of the cells appeared to be bipolar showing two immunoreactive processes. Electron microscopy observation was made using the section prepared by a pre-embedding technique. In the immunoreactive perikarya, secretory granules, ribosomal particles, and the particles on the outer surface of endoplasmic reticulum appeared to be immunoreactive, but the Golgi area was immunonegative. The secretory granules appeared along the cytoplasmic membrane but were surprisingly few in number. By contrast, the immunoreactive nerve terminals in the median eminence and in the OVLT contained many granules. These findings suggest that LHRH produced in the perikarya is prompted to be transported and stored in the axonal terminals.

Electron microscopic study of immunoreactive LHRH perikarya with special reference to neuronal regulation.

In early postnatal rats, immunoreactive LHRH perikarya in the preoptic area were studied by light and electron microscopy. Synaptic junctions were found between the immunoreactive perikaryon or its process, and the immunonegative nerve fibers. The significance of these synapses is discussed in relation to possible mechanisms by which the activities of LHRH neurons are regulated.

Effects of long term enteral hyperalimentation on pancreatic secretion in dogs.

Acute and chronic experiments in adult mongrel dogs were performed to investigate the influences of a low residue diet (Group I) and an elemental diet (Group II) on pancreatic function. 1) In the acute experiment, intraduodenal administration of the low residue diet produced a significant increase in amylase output in pancreatic juice, while the elemental diet die not. 2) Intragastric administration of the low residue diet or elemental diet to conscious dogs showed little difference between the two groups regarding secretin and gastrin release, at the same pH. 3) Long term enteral hyperalimentation showed that it is the low residue diets which enhance pancreatic secretions. 4) In both groups, carbohydrate metabolism, liver function and electrolyte balance were satisfactorily maintained during long term enteral hyperalimentation and morphologic changes of the liver, pancreas and small intestine were not evident. 5) From the viewpoint of nutritional management, low residue diets should be prescribed in some cases of alimentary surgery.

Heterotransplantation of human upper urinary tract tumors in nude mice.

Eight human upper urinary tract tumors were transplanted subcutaneously into congenitally athymic nude mice. Subcutaneous growth was obtained in three (NKPP 1, NKPP 2, and NKPU 1) at initial transplantation and they were all serially passaged up to 29, 28, and 5 passages, respectively. Their histologic appearances were similar to those of the originals. These established strains with high transplantability and differing degrees of thromboplastic and fibrinolytic activities will provide a useful model system for studying anticancer therapies and biological characteristics of upper urinary tract tumors.

Immunohistochemical and histochemical investigations on in vivo thrombolysis with urokinase in rabbits.

Using human urokinase in vivo thrombolysis was studied in autologous artificial thrombi in the pulmonary circulation of rabbits by immunofluorescence and Todd's fibrinolysis autography techniques. As compared to the control thrombi of the untreated rabbits, a small increase of thrombolysis was found in the rabbits treated with urokinase. Urokinase fluorescence was observed in the leukocytes, but not along the fibrin fibrils in the thrombi of the rabbits treated with urokinase. By Todd's fibrinolysis autography, lytic areas were observed around aggregated leukocytes in the thrombi of the rabbits treated with urokinase. The small lysis of autologous artificial thrombi of rabbits by human urokinase may be caused by (1) low affinity of human urokinase for rabbit plasminogen, (2) weak adsorption of human urokinase to rabbit fibrin, (3) phagocytosis of human urokinase by leukocytes and (4) high level of antifibrinolytic activity of rabbit plasma.

Synaptic junctions in the sinus gland of the freshwater prawn Palaemon paucidens.

Two types of neurosecretory fibers, designated as Type 5 and Type 6 axons, in the sinus gland of the freshwater prawn, Palaemon, establish contact with other neurosecretory axons by means of synaptic junctions. This finding strongly supports the view that release of some neurohormones from the eyestalk may be regulated by neurosecretory neurons through synaptic transmission.

Immunofluorescence study on thrombolysis with special reference to the patterns of distribution and the contents of fibrin, plasminogen, alpha2-macroglobulin and urokinase in artificial thrombi.

By the immunofluorescence technique, fibrin, plasminogen and alpha2-macroglobulin demonstrated a similar basic pattern of distribution in the artificial thrombi, rimming the platelet aggregates in the white part and forming a network structure in the red part. The content of plasminogen seemed to be higher in the red part than in the white part. After superfusion with urokinase solution, urokinase was found in a similar pattern of distribution of that of fibrin. The content of urokinase seemed to be higher in the red part, especially in its outer area, than in the white part. It is concluded that urokinase diffused into the artificial thrombi can convert plasminogen, trapped onto fibrin in threads to plasmin. Poor lysibility of the white part might be due to thick strands of fibrin, a low content of plasminogen and a low diffusibility of plasminogen activator, in addition to the antifibrinolytic activity of platelets.

Scanning and transmission electron microscopic study of human pulmonary pneumocystosis.

Histopathologic examinations were performed in 13 autopsy cases with Pneumocystis pneumonia, five of whom were examined with scanning as well as transmission electron microscope. Findings by scanning electron microscopy were compared with those by light microscopy and transmission electron microscopy. By scanning electron microscopy, two principal structural varieties of Pneumocystis organisms, smooth-surfaced organisms and rough-surfaced organisms, could be classified. It was indicated that rough-surfaced organisms and smooth-surfaced organisms correspond respectively to trophozoite-type organisms and thick-walled cysts, recognized with transmission electron microscope. Intraalveolar honeycombed material was mainly composed of dilated trophozoite-type organisms, containing thick-walled cysts scatteredly.

Neurosecretory cell types in the eyestalk of the freshwater prawn Palaemon paucidens: an electron microscopic study.

In the medulla terminalis ganglionic X-organ (MTGX) of the eyestalk of the freshwater prawn, Palaemon paucidens, six peptidergic neurosecretory cell types (A-, B-, C-, D-, E-, and F-cells) are distinguishable on the basis of the different morphology of their elementary granules and rough endoplasmic reticulum (rER). All of these cell types seem to correspond to Type-IIIa cells or dispersing Type-IV cells, that have previously been differentiated at the light microscopic level (Hisano, 1974), as judged from the dimensions of their cell bodies and nuclei. Two other peptidergic neurosecretory cell types that are apparently comparable to the Type-II and Type-IIIb cells (Hisano, 1974), respectively, are recognized in parts of the optic ganglia other than MTGX, and these are now designated as G- and H-cells, respectively. All the remaining cell types, designated as Type-I, cluster-forming Type-IV, Type-V and Type-VI cells in our previous light microscopic study, have small cored-vesicles in their cytoplasm. It remains undecided whether these, possibly aminergic, neurons are neurosecretory or not.
