Amino acids and peptides. XXVIII. Preparation of poly(ethylene glycol) hybrids of Gln-Val-Val-Ala-Gly analogs and their inhibitory effect on papain.

Gln-Val-Val-Ala-Gly is a sequence which is common in cysteine protease inhibitors. Poly(ethylene glycol) hybrids of Gln-Val-Val-Ala-Gly analogs were prepared by the solid phase method by fluorenylmethyloxycarbonyl chemistry. The hybrid formation of Gln-Val-Val-Ala-Gly resulted in improvement of the solubility and also in enhancement of the inhibitory effect of the peptide on papain. In addition, the hybrid formation of Gln-Val-Val-Ala-Gly analogs resulted in the enhancement of the inhibitory effect of the peptide analogs.

Amino acids and peptides. XLIV. Synthesis of stereoisomeric pentapeptides of thiol proteinase inhibitor.

Stereoisomers of thiol proteinase inhibitor (TPI) were synthesized by a conventional solution method. Among them, iNoc-D-Gln-Val-Val-Ala-Ala-pNA weakly inhibited the amidolytic activity of papain, although iNoc-Gln-Val-Val-Ala-Ala-pNA inhibited papain activity fairly potently. However, the other five D-amino acid-containing peptides did not show any inhibitory effects on papain. The circular dichroism (CD) spectra of the enzyme-peptides mixtures were measured in order to study the relationship between the peptide anilides-papain interaction and the inhibitory activity.

Severe cryoglobulinemia in a patient with asymptomatic hepatitis C virus infection.

A patient with severe type II cryoglobulinemia after ten years of asymptomatic hepatitis C virus (HCV) infection is reported. Laboratory data showed hypergammaglobulinemia with polyclonal IgG and monoclonal IgM, blood hyperviscosity, high level of cryocrit (60%), HCV viremia, and normal levels of serum transaminases. Due to cold exposure, acrocyanosis and cardiac ischemic attack occurred; he recovered after being artificially warmed. Administration of prednisolone and natural interferon-alpha for three months resulted in significant decreases in the levels of serum IgM and cryocrit.

A protective effect of glutathione-dextran macromolecular conjugates on acetaminophen-induced hepatotoxicity dependent on molecular size.

Glutathion (GSH) was covalently attached to dextrans with various molecular weights of 2, 5, 10, 40, and 70 kDa by the cyanogen bromide activation method. The conjugates obtained synthetically were white or pale yellowish powders containing 6-10% (w/w) of GSH. The average molecular weights of the conjugates were estimated to be larger and the molecular weight distribution was a little broader than that of each original dextran. The conjugates significantly stabilized GSH and liberated it gradually under physiological conditions (t1/2 = 0.99-1.6h). Mice depleted of GSH by treatment with buthionine sulfoximine, a potent inhibitor of gamma-glutamylcysteine synthetase, exhibited a significant increase in hepatic GSH level after intravenous injection of the conjugates. In mice given a hepatotoxic dose of acetaminophen, the survival rate increased progressively with coadministration of the conjugates, whereas a small improvement was found when free GSH was given. The conjugate of GSH attached to dextran with the molecular weight of 40 kDa exhibited the highest prophylactic effect on acetaminophen-induced hepatotoxicity in mice. The prolonged retention of the conjugates of larger molecular weight in the circulation would cause a higher hepatic accumulation. These results suggested that molecular size would be the most critical factor in the delivery of GSH, as a dextran conjugate, into the liver.

The disposition of serum proteins as drug-carriers in mice bearing Sarcoma 180.

The tumor distribution and the disposition of serum proteins, such as albumin, fetuin, transferrin, and IgG, were investigated in mice bearing Sarcoma 180. Serum proteins labeled with fluoresceinisothiocyanate (FITC) were administered to the mice. The FITC-labeled proteins acylated with glutaric anhydride were also administered to the mice in order to investigate the effect of chemical modification. The plasma concentration of each glutarylated serum protein was significantly lower, about 15 to 46-fold, in comparison to that of the non-acylated protein at 24 h after administration. The tissue distributions of the glutarylated serum proteins were also decreased compared to those of the non-acylated proteins. Especially, the hepatic distribution of albumin and IgG was significantly reduced with glutarylation. The urinary excretion of albumin and transferrin, and fecal excretion of IgG, were significantly increased with glutarylation. The serum proteins were accumulated effectively in the tumor tissue in mice bearing Sarcoma 180. It was found that the tumor distributions were not impaired by the glutarylation, except involving fetuin. It was suggested, therefore, that the glutarylated serum proteins were valuable for relative tumor-selectivity and might be utilized in a macromolecular carrier system for antitumor drugs.

Novel 5-HT3 antagonists. Isoquinolinones and 3-aryl-2-pyridones.

Synthesis and pharmacological evaluation of a series of 1,2-dihydro-1-[(5-methyl-1-imidazol-4-yl)methyl]-2-oxopyridine 5-HT3 antagonists are described. The key pharmacophoric elements were defined as a basic nitrogen, a linking group capable of hydrogen bonding interactions, and an aromatic moiety. 1,2-Dihydro-2-oxopyridine moiety could be a good linking group because of its nicely planar structure. The steric limitations of the aromatic moiety were investigated by X-ray analysis and computer analysis and shown to be optimal when the aromatic moiety was constrained within an arched planar system, which could be successfully replaced by 3-(2-thienyl)-2-oxopyridine function or 6-amino-7-chloro-1-isoquinolinone function without any loss of the activity. Among the synthesized compounds, 42 showed the most potent activity in the inhibition of Bezold-Jarisch reflex in rats. Compounds 44a and 64 were orally active in the protection against cisplatin-induced emesis in dogs or ferrets. Structure-activity relationships are discussed.

Amino acids and peptides. XXXIII. Synthesis of N-terminal epitope peptides of mammalian metallothioneins (MTs).

In order to determine the fine structure of the mammalian metallothionein (MT) epitope to a monoclonal anti-rat Zn-MT-II antibody (MT 189-14-7), N-terminal peptides of various lengths of mammalian metallothioneins (MTs) were synthesized by a conventional solution method using the newly developed beta-2-adamantylaspartate, and their immunological properties were examined. It was found that the N-terminal acetyl group was indispensable for the reaction with the monoclonal antibody and the N-terminally acetylated pentapeptide, Ac-Met-Asp-Pro-Asn-Cys-OH, was the smallest peptide which exhibited a significant reactivity with the antibody.

Development of a highly cardioselective ultra short-acting beta-blocker, ONO-1101.

A novel, highly cardioselective ultra short-acting beta-blocker, ONO-1101, has been developed for application in the emergency treatment of tachycardia and better control of heart rate in surgery. This agent is approximately nine times more potent in beta-blocking activity in vivo and eight times more cardioselective in vitro than esmolol. This beta-blocking drug has a short duration of activity, enabling rapid recovery after cessation of administration if side effects occur. It can be used safely in patients suffering from acute heart disease and represents a major therapeutic advance in the treatment of heart disease.

Targeting of mitomycin C to the liver by the use of asialofetuin as a carrier.

Desialylated fetuin (asialofetuin) was adopted as a carrier for introducing drugs in parenchymal liver cells. Mitomycin C, as a model guest-compound, was covalently attached to asialofetuin through a spacer of the succinyl group. The asialofetuin-mitomycin C conjugate contained 4.4 w/w% of mitomycin C and liberated it gradually at physiological conditions (t1/2 = 37 h). The survival time of the conjugate in rat blood circulation was significantly smaller than that of the non-desialylated fetuin conjugate; the elimination half-life was 7.3 min after intravenous injection. At 30 min after injection of the conjugate in rats, 40% of the dose was present in the liver. Parenchymal cells in the liver selectively took up the conjugate, which was highly distributed to the lysosomal fraction in the cell. The greater uptake of the conjugate by hepatocytes reflected the increased excretion in the bile; totally 10.4% of the dose was recovered.

Antigenic determinants on rat metallothionein: fine epitope mapping for a murine monoclonal antibody and rabbit polyclonal antisera.

In order to determine the epitope of metallothionein (MT) to a murine monoclonal antibody (MT 189-14-7) which had been produced by immunization with rat MT 2 (Kikuchi et al. (1988) Mol. Immunol. 25, 1033-1036), various lengths of synthetic oligopeptides were tested for their inhibitory activities in competitive radioimmunoassay (RIA). The amino-terminal acetylated pentapeptide, AcMDPNC, exhibited an inhibitory activity comparable to that of native MTs, whereas the acetylated tetrapeptide, AcMDPN, and the deacetylated heptapeptide, MDPNCSC, were much less inhibitory. The results suggest that the major part of the epitope structure of MT to the MT 189-14-7 monoclonal antibody is located within the amino-terminal acetylated pentapeptide, AcMDPNC. The specificities of polyclonal rabbit anti-MT antisera raised against the same immunogen were also determined by using various animal MTs and synthetic peptides as inhibitors in the RIA. Among three antisera tested, two reacted with several amino-terminal oligopeptides similarly to the MT 189-14-7 antibody. The major epitope structures to these polyclonal antibodies were shown to be located within the acetylated tetrapeptide, AcMDPN. Another antiserum contained at least two different populations of antibodies: one consisted of antibodies reactive with the amino-terminal synthetic peptides, while the other was not reactive with them. These results suggest that, in the rabbit also, the amino-terminal region common to various animal MTs can be an epitope to antibodies raised against rat MT, as shown in the mouse. Moreover, the results indicate that the synthetic amino-terminal peptides are useful for determination of the specificity of polyclonal rabbit anti-MT antibodies, which have been widely used for the quantification of MTs.

Preparation and properties of a mitomycin C-albumin conjugate.

Mitomycin C, an anti-neoplastic agent, was covalently attached to bovine serum albumin through various kinds of spacers such as glutaryl, succinyl, trans-aconityl, methylsuccinyl and the trimellityl group. The prior acylation of albumin not only prevented protein polymerization in the presence of carbodiimide, but also increased the extent of conjugation of the drug. The conjugate of mitomycin C-glutarylated albumin showed the best properties among the conjugates prepared in meeting the requirements for a high yield of nonpolymerized product with an adequately high mitomycin C content and stability as a macromolecular prodrug.

Intrahepatic delivery of glutathione by conjugation to dextran.

Glutathione was covalently attached to dextran (T-40) by the CNBr activation method. The compound obtained was a water-soluble powder containing 10 (w/w%) glutathione, which was gradually released from the conjugate in aqueous media. Mice depleted of glutathione by treatment with buthionine sulfoximine, a potent inhibitor of gamma-glutamylcysteine synthetase, exhibited a significant increase in hepatic glutathione level after intravenous injection of the conjugate. In mice given a lethal dose of acetaminophen, the survival rate increased progressively with coadministration of the conjugate, whereas little improvement was found when free glutathione was given. The conjugate maintained the serum transaminase activities at lower level after acetaminophen administration. These findings suggest that the dextran conjugate of glutathione is transported into hepatic cells and is intracellularly hydrolyzed to free form, which protects mice from hepatotoxicity due to acetaminophen.

Effects of glutathione, as the dextran conjugate, on acetaminophen-induced hepatotoxicity.

Glutathione was covalently attached to dextran (T-40) by the CNBr activation method. In mice given a lethal dose of acetaminophen, the 30-d survival rate increased progressively with coadministration of the conjugate, whereas little improvement was found when free glutathione was given. The dextran conjugate of glutathione maintained the serum transaminase activities at lower levels after acetaminophen administration, giving effective protection against acetaminophen hepatotoxicity.

Amino acids and peptides. XXV. Application of newly developed beta-1- and beta-2-adamantylaspartates to peptide synthesis by solid phase and conventional solution methods.

Newly developed beta-1- and beta-2-adamantylaspartates [H-Asp(O-1-Ada)-OH and H-Asp(O-2-Ada)-OH] were applied to the synthesis of a C-terminal octapeptide of the beta-subunit of human chorionic gonadotropin (hCG) by a conventional solution method and a hexacosapeptide of the alpha-subunit of insulin receptor (30-55) by a solid-phase method with the objective of suppressing aspartimide formation during the synthesis of aspartylpeptides. The 1-Ada and 2-Ada groups were confirmed to be useful protecting groups for the beta-carboxyl function of the Asp residue.

[Stress-generated potential (SGP) on the surface of fluid-filled monkey mandible].

Previous studies have shown that stress generated potential (SGP) with bone deformation caused by orthopedic force have an important role in regulation on bone remodeling. The purpose of this study was to evaluate the effects of the chin cap on bone remodeling by elucidating the characteristics and distribution of SGP at various parts on the mandible. Experiment was carried out using a mandible of macaca irus monkey which was kept in moisture with 0.9% saline. SGP was recorded using Ag-AgCl electrode from the 32 sections on the surface of the mandible. The force (3 kg) was applied to the chin with six load conditions. It simulated the force generated by chin cap. Temporomandibular joints and the soft tissue except masseter muscle were kept intact. The results were summarized as follows. 1. SGP was generated only when the load was applied or removed. No SGP was generated when the load was steady, even the absolute value was large. 2. When the interval between "on" and "off" of the load was shorted, generated SGP with "off" timing decreased, while that with "on" timing unchanged. 3. When the load was applied in the low pull direction under the centric occlusion, rather larger negative SGP was recorded at the chin, posterior sections of mandibular body and inferior border of the mandible. 4. When jaw was in closing position, high pull loading generated larger SGP at the alveolar bone and the mandibular body than that with low pull loading, although their electric sign was the same at each recorded sections. 5. When jaw was in opening position, low pull loading generated larger SGP at the posterior parts of the alveolar bone and the mandibular body than that in closing position. 6. When a bite block was placed between the upper and lower teeth, larger amplitude in SGP was the evidence in all the recorded sections especially around the bite block. 7. SGP recorded at the posterior part of the mandible increased with the sliding of the location of the bite block towards posterior portion. 8. The electric sign and the amplitude in SGP and that of the stress analyzed by a finite element method were similar at the each sections on the mandible. This suggests SGP is related with the stress generated in the bone. The results suggested that effects of chin-cap on the bone remodeling might be dependent on the direction of the load, tooth contact, existence of bite block, and its location. Also dynamic loading was suggested to be more effective.
