The effect of silibinin (Legalon) on the the free radical scavenger mechanisms of human erythrocytes in vitro.

The effect of Legalon was investigated parallel with that of Adriblastina (doxorubicin) and paracetamol on some parameters characterizing the free radical scavenger mechanisms of human erythrocytes in vitro and on the time of acid hemolysis performed in aggregometer. Observations suggest that Adriblastina enhances the lipid peroxidation of the membrane of red blood cells, while paracetamol causes significant depletion of intracellular glutathione level, thus decreasing the free radical eliminating capacity of the glutathione peroxidase system. Legalon on the other hand, is able to increase the activity of both superoxide dismutase and glutathione peroxidase, which may explain the protective effect of the drug against free radicals and also the stabilizing effect on the red blood cell membrane, shown by the increase of the time of full haemolysis.

Lipid peroxidation and superoxide dismutase activity in muscle and erythrocytes in adult muscular dystrophies and neurogenic atrophies.

Lipid peroxidation (LP) and superoxide dismutase (SOD) activity were determined in erythrocytes and skeletal muscle obtained from patients with limb-girdle and facioscapulohumeral muscular dystrophies, neurogenic atrophies and from age-matched control subjects. Neither lipid peroxidation nor SOD activity in erythrocytes of patients differed from control values. SOD activity and LP in muscle specimens were also normal in types of neurogenic atrophy. Lipid peroxidation in the muscle from patients with adult types of muscular dystrophy had a tendency to be increased. The values were widely scattered, the highest being obtained in the older patients with long duration of disease.

Effect of 2-mercaptoethanol on some metabolic indices of ageing of CBA/Ca inbred mice.

It has been shown in several studies that 2-mercaptoethanol (2-ME) improves the life span and aspects of life performance of laboratory animals. To obtain further details on the beneficial effects of 2-ME, a long-term study has been performed on male CBA/Ca inbred mice treated with this antioxidant. Four month-old mice were each given 4 micrograms of 2-ME in physiological saline via intraperitoneal (i.p.) injection 3 x per week. Measurements were made of the following: cold tolerance (heat performance), apparent total body protein turnover (T1/2), changes in the major lipid and fatty acid compositions of the liver, superoxide dismutase activity and formation of malondialdehyde and observations on a range of pathological changes. It was found that the basal rectal temperatures of the treated mice were higher and in the oldest group, heat performance capacity was better than those of the controls. After about 1 year of age the apparent biological half-life time of total body protein (T1/2) was observed to be shorter in the treated mice. Significant increases were observed to occur in the proportions of the polyunsaturated fatty acids in the lipids of the liver in the mice injected with 2-ME. Although no differences were observed in the superoxide dismutase activities, malondialdehyde concentrations in the livers of the experimental mice were significantly increased. Autopsy data showed that Dunn-sarcomas associated with amyloidoses occurred more frequently in the untreated mice.

Comparative study on the metabolism and filtrability of red blood cells of the calf and adult cattle.

Filtrability of calf and adult cattle red blood cells was studied to characterize their durability; to compare their metabolism, lactate dehydrogenase, glucose-6-phosphate dehydrogenase and glutamate-oxalacetate transaminase activity and ATP-content were determined. The activity of the enzymes declined gradually with age. The most pronounced decline of activity, particularly in the case of lactate dehydrogenase, was observed 6--8 weeks after birth. ATP content of calf red blood cells was ten times higher than that of the adult cattle, however, it proved to be less stable. Filtrability of fresh calf and adult cattle red blood cells was identical, while following incubation for 15--24 hours the decrease of filtrability was more pronounced in calf red blood cells indicating a lower filtrability and, consequently a shorter expected lifetime.

The effect of dbcAMP on the lysolecithin induced hemolysis of calf and adult cattle erythrocytes.

The calf erythrocytes have an increased sensitivity against lysolecithin as compared to their adult counterparts. 10(-3)M dbcAMP increases the hemolysis induced by 5 microgram of lysolecithin in 0.15 M NaCl containing 10 mM phosphate buffer (pH 7.4). By increasing the level of phosphate buffer (75 mM) in the incubation mixture, 10(-3)M dbcAMP decreases the hemolysis induced by 5 microgram of lysolecithin. These data suggest a dual effect exerted by dcbAMP: the relatively labilizing or stabilizing effect prevails as a function of exogenous inorganic phosphate level. 10(-6)M dcbAMP also has a relative protective effect against lysolecithin. The combined addition of cAMP (10(-3)) and theophyllin (10(-4)M) does not stabilize the membrane. By increasing the level of lysolecithin to 20 microgram/ml the stabilizing effect of dcbAMP disappears. DbcAMP (10(-3)) as well as cAMP (10(-3)M) and theophyllin (10(-4)M) have a minimum increasing effect on hemolysis in the absence of lysolecithin, too.

Age Factors

A mofidied method for measuring the filtrability of erythrocytes.

A modified method based on Teitel's original procedure for measuring the filtrability of erythrocytes was described. The filter paper was not placed into a funnel but to the exit at the bottom of a 2 ml glass column. The filtration of the red cell suspension was quantitated by measuring the decrease of the volume in the column.

Effect of osmotic pressure, ionic strength and dibutyryl cyclic adenosine monophosphate on the adhesion of hen erythrocytes.

In the presence of lysolecithin at physiological pH it was found that the increase of ionic strength facilitates the adhesion of hen erythrocytes. In this medium, dibutyryl cyclic adenosine monophosphate (DBcAMP) increases the adhesion index of the cells. If the osmotic pressure is elevated without a proper increase of ionic strength, the lysolecithin induced hemolysis and adhesion are found to be lacking.

[Effect of leukeran on the local Shwartzman phenomenon].

A study has been made to investigate the effect of treatment with Leukeran (Chorambucil) on the development of what is known as a loval Shwartzman phenomenon. The experimental results show that it is only in those cases where the leukopoietic capacity of the organism has been fully exhausted that it is possible for the development of this particular phenomenon to be reliably prevented by immunsuppression brought about by treatment with Leukeran.

[Muscular clearance after administration of phenoxybenzamine in shock].

The authors made an experimental investigation in the dog concerning the evolution of the muscular clearance of phenoxybenzamine in the course of de-shocking procedures. The animals were shocked by hemorrhage and were treated by perfusion of volemic mass and administration of phenoxybenzamine. This substance was flowed in paralell with the hemodynamic parameters and the rhythm of the diuresis. It was noted that the direction and the amplitude of changes concerning the half-life of Na131I do not depend on the blood pressure.
