Production of adenocarcinomas of the small intestines of Wistar rats fed panfuran-S containing 3-di(hydroxymethyl)amino-6-(5-nitro-2-furylethenyl)-1,2,4-triazine.

Panfuran-S in the diet produced a high incidence of invasive and metastatic adenocarcinomas of the duodenum and the jejunum in outbred Wistar rats. Depressed growth and incidence of cancer production were dose-dependent. Adenocarcinomas of the duodenum were present in 2 of 18 (11%) rats that received 1,750 ppm 3-di(hydroxymethyl)amino-6-(5-nitro-2-furylethenyl)-1,2,4-triazine (DHNT) in the diet and in 11 of 19 (58%) rats that received 3,500 ppm DHNT. Adenocarcinomas of the jejunum were present in 10 of 18 (56%) and 16 of 19 (84%) rats that received 1,750 and 3,500 ppm DHNT, respectively, for 35-37 weeks. The only other tumors observed were squamous cell papillomas of the forestomach. Histologically, most of the adenocarcinomas appeared tubular or papillary and were similar to adenocarcinomas in the small intestine of man and also to human intestinal-type adenocarcinoma of the stomach. This system provided a useful experimental model for the study of the pathogenesis of carcinoma of the small intestine.

Neural changes in recurrent infection of infectious bovine rhinotracheitis virus in calves treated with dexamethasone.

Recurrent infection by infectious bovine rhinotracheitis (IBR) virus was induced in calves by dexamethasone (DM) treatment (given 5 days) at 5 months after primary infection. The virus appeared in nasal secretions of the calves on the 4th day after initiation of DM treatment and continued until the 9th day. The calves were killed on the 1st, 3rd, 4th, 5th, 6th, 7th, 8th, 10th, and 11th days after DM treatment was started for examination by histopathologic and immunofluorescent antibody techniques. The most significant neural change was trigeminal ganglionitis with neuronophagia, which was observed from the 3rd to the 11th day. Significantly, the extent of changes in the trigeminal ganglion and medulla oblongata corresponded to the amount of DM treatment administered. The IBR virus antigen was first observed in the trigeminal ganglion cells, and thereafter, it was detected in the Schwann cells, satellite cells, neuroglia cells, and nasal mucosa until the 10th day. These observations indicate that the IBR virus is capalbe of producing a persistent infection in the trigeminal ganglion and that trigeminal ganglionitis may be a characteristic lesion for inducing the reactivation of lagent IBR virus.

Bovine virus diarrhea-mucosal disease. II. Isolation and characterization of a cytopathogenic virus and experimental production of the disease.

A disease broke out in calves in the Tokachi district of Hokkaido. It induced pyrexia, respiratory symptoms, diarrhea, bloody feces, leukopenia, and sometimes erosion of the oral mucous membrane and muzzle. Its morbidity rate was 90% and its fatality rate 50%. Bovine virus diarrhea (BVD) virus was isolated from organs of dead calves and blood and feces of affected calves. It exhibited a cytopathic effect on calf kidney cell culture. Antisera against the Nose and the Oregon C24V strains of BVD virus showed an antibody titer of the same order against the homologous virus and the isolated strain. Antiserum against the isolated strain, however, showed much lower antibody titers against the Nose and the Oregon C24V strains than against the homologous virus. When inoculated with the isolated virus, two calves manifested acute symptoms, but recovered at any rate. One of them, however, suffered again from clinical infection and died eventually 37 days after inoculation. It presented pathological changes closely resembling those of the case of spontaneous infection. Virus was recovered from its principal organs, intestinal canal, and lymph nodes of various regions of the body.

Organotropic effect of N-bis (2-hydroxypropyl)nitrosamine: production of lung and liver tumors by its oral administration in mice.

The organotropic effect of N-bis(2-hydroxypropyl)nitrosamine (DHPN) orally administered to mice was studied. The main targets for the carcinogenic effect of DHPN were the lung and liver, with tumors induced by doses greater than 100 ppm DHPN for 16 weeks. The highest incidence was seen in liver vascular tumors including hemangioma, hemangioendothelioma, and hemangioendothelial sarcoma in mice treated with 1,000 ppm DHPN. Lung tumors were alveolar adenoma and adenocarcinoma. No pancreatic tumors were found. The present results suggest that the carcinogenic response in mice to DHPN was similar to that in rats and guinea pigs but different from that in hamsters.

Differential effects of 4-hydroxyaminoquinoline-1-oxide on pancreatic and liver DNA synthesis in rats.

The effect of 4-hydroxyaminoquinoline-1-oxide (4-HAQO) on DNA synthesis in the pancreas and liver, target and non-target organs for 4-HAQO carcinogenesis, respectively, were compared. Pancreatic and liver DNA synthesis were simultaneously induced in rats fed a protein deficient diet containing 0.5% DL-ethionine for 18 days, and DNA synthesis in both tissues was inhibited by hydroxyurea. A single i.v. injection of 4-HAQO at a dose of 7 mg/kg body weight also inhibited DNA synthesis in both tissues within 4 h. In the pancreas the inhibition was maximum at a dose of 7 mg/kg, and DNA synthesis was less than in the pancreas of rats fed a control grain diet. This inhibition continued for the subsequent 5 days which were tested. In the liver, the degree of inhibition was less than in pancreas but the value remained higher than in rats fed control diet. The inhibition of liver DNA synthesis at a dose of 7 mg/kg completely recovered within 1 day. These results suggest that the lesions of DNA induced by 4-HAQO and its repair might be different between the pancreas and the liver. A pancreatic chemical carcinogen, 4-HAQO, might thus have the same cytotoxic effect that liver carcinogens have toward the liver resulting in failure to respond to mitotic stimuli. This might be causally related to the organotropism of 4-HAQO toward the pancreas.

The first seven cases of chronic beryllium disease in ceramic factory workers in Japan.

In the present paper the first 7 cases in Japan of chronic beryllium disease found in workers employed in a ceramic factory utilizing beryllium have been described. Immunological examinations of these cases showed changes similar to those observed in sarcoidosis, that is, negative tuberculin test and increase in serum gamma globulin and immunoglobulins. The fact that a considerable number of workers in the same factories as the patients showed negative tuberculin reaction may suggest that there may be further cases of chronic beryllium disease among them that are still in a latent period.

Eimeria leuckarti infection in foals.

This is the first report on Eimeria leuckarti infection in foals in Japan. Seven Thoroughbred or Angloarabian foals 2 to 7 months of age raised in the Hidaka district, Hokkaido, were infected with E. leuckarti. They died of severe alterations caused by the larval migration of Strongylus vulgaris, and were examined over a period from 1970 to 1973. Protozoa of this species were observed in the small intestine in all the foals. They were found mostly in vacuoles of the cytoplasm of monoclear cells in the lamina propria at or near the tip of villi. Various stages of gametocytes, oocysts, and microgametocytes were found. In infected cells the nucleus was markedly enlarged, but the cellular response was very slight to the parasite.

Histopathological changes of the brain in swine fetuses naturally infected with procine parvovirus.

During a period from 1971 to 1972, some cases of stillbirth and abortion associated with porcine parvovirus infection were recognized in swine herds in Japan. The brain was examined histopathologically in five stillborn piglets and four dead fetuses from which the parvoviruses had been isolated. Similar histological changes were observed in all the piglets and fetuses, except one piglet which was free from recognizable lesions. The brain lesions were considered to belong to the category of meningoencephalitis consisting of perivascular cuffing of proliferating adventitial cells and a few plasma cells. They were localized in the cerebral gray and white matter and leptomeninges, but not in the cerebellum. These histological lesions were thought to be pathognomonic to porcine parvovirus infection. The difference in brain lesions between porcine parvovirus infection and Japanese encephalitis virus infection was discussed.
