A structural model of cholinergic synaptic vesicles from the electric organ of Torpedo marmorata deduced from density measurements at different osmotic pressures.

Density measurements made on cholinergic synaptic vesicles from the electric organs of Torpedo marmorata at different osmotic pressures are consistent with the following structural model of the vesicle. The particle behaves like a sphere 80-100 nm in diameter bounded by a semi-permeable membrane. The bulk of its soluble constituents are in true solution at physiological osmolalities. The limiting membrane is approximately 4-5 nm thick, suggesting that it contains large areas of phospholipid bilayer exposed to its bathing medium. The limiting membrane takes up about 26% (v/v) of the particle, a further 34% (v/v) of which is osmotically active water and 31% (v/v) hydrated core material at 800 mosmol/1. The buoyant density of the membrane is 1.132 g . cm-3. The density of the hydrated core material is approximately 1.05 g . cm-3. The membrane is selectively permeable to small molecules when subjected to hypo-osmotic stress. It is proposed that this occurs by the formation of small transient pores in the lipid bilayer of the membrane, which are induced by stretching caused by the osmotic pressure change.

Duodenal leiomyoma as a cause of gastrointestinal hemorrhage.

A case report and review of the problem of duodenal leiomyoma are presented. Although these lesions are rare and usually asymptomatic, they can present with symptoms of hemorrhage, obstruction, pain, and perforation. Barium roentgenographic examination may show filling defects of compressive lesions. Angiography shows a hypervascular, encapsulated lesion. Endoscopy may reveal a submucosal mass with central depression. At operation, it is important to distinguish the benign leiomyoma from leiomyosarcoma.

Colonoscopic evaluation of rectal bleeding: a study of 304 patients.

We studied 258 patients with rectal bleeding and 46 patients with anemia and occult blood in the stool. All 304 patients had negative proctosigmoidoscopies, single-contrast barium studies that were negative or showed diverticula only, and colonoscopic evaluation. In the 258 patients, the overall incidence of finding significant lesions by colonoscopy was 41.5%. Twenty-nine patients (11.2%) had carcinoma and 17 patients (6.6%) had cecal telangiectasia. In the 46 patients, the overall incidence of finding significant lesions was 19.6%. Three patients with carcinoma were found in this group. A significant number of both benign and malignant lesions were detected by colonoscopy proximal to the splenic flexure. Colonoscopy should be done in patients with rectal bleeding or anemia and occult blood in the stool who have had negative proctosigmoidoscopies and single-contrast barium studies interpreted as normal or showing diverticula.

Disulfiram hepatitis.

A case of an adult with disulfiram-induced hepatitis is presented. An etiological role for the drug was proved when the patient underwent a provocative challenge and developed evidence of hepatocellular damage, reversible on withdrawal of the drug. Disulfiram-induced liver injury is difficult to detect in a population with a high incidence of alcohol-related hepatic disease. Liver chemistries should be screened before beginning disulfiram therapy and periodically during treatment.

Rapid diagnosis of obstructive jaundice due to pancreatic abscess with pancreaticobiliary fistula.

A case of pancreatic abscess with pancreaticobiliary fistula manifesting as obstructive jaundice of occult etiology is presented. Diagnosis was made preoperatively by skinny needle percutaneous transhepatic cholangiography. In addition, a communication between the biliary tree and the pancreatic abscess cavity was demonstrated in the absence of previous biliary surgery or primary biliary tract disease. We feel this is the procedure of choice for emergency visualization of the biliary tree in a jaundiced patient where a surgically approachable lesion is suspected.

Adenosine triphosphatase activity associated with purified cholinergic synaptic vesicles of Torpedo marmorata.

A rapid method for purifying Torpedo electric organ vesicles is described, which employs an isoosmotic continuous sucrose-glycine gradient followed by chromagography on CPG-10-3000 porous glass beads. The synaptic vesicles have a buoyant density of 1.057 g/ml. The purified vesicles are free of cholinesterase, lactate dehydrogenase and Na+, K+-stimulated ATPase activity. They contain a ouabaininsensitive, Na+, K+-inhibited, Mg2+, Ca2+-stimulated ATPase activity. This is further stimulated by acetylcholine but not by choline.

Demonstration of binding sites for divalent and trivalent ions on the outer surface of chromaffin-granule membranes.

1. Trivalent ions Tb3+, Eu3+ and La3+ aggregate chromaffin granules and produce structural changes in the core material. These ions also stain the outer (cytoplasmic) surface of the granule membrane in the presence of PO43+ ions and inhibit OSO4 staining. The electron-dense patches of TbPO4 complex are distributed in a non-random fashion. 2. Tb3+ also functions as a fluorescent membrane probe for divalent ion binding sites on the granule membrane. Using the enhancement of Tb3+ fluorescence upon binding, a Kd for Tb3+ of approximately 15 micronM was measured. Ca2+ and Mg2+ are competitive inhibitors of this binding while Na+ and K+ had no effect. Results suggest that the fluorescent Tb3+ -binding site is a protein. 3. Tb3+ also binds to mitochondria and other contaminants as judged by electron microscopy. However purified mitochondria show qualitatively different binding of Tb3+ by fluorescence. 4. A model for the location of divalent ion binding sites on the granule membrane and the results are discussed in terms of requirements for the participation of these sites in granule exocytosis in vivo.

Calcium-induced displacement of membrane-associated particles upon aggregation of chromaffin granules.

Isolated chromaffin granules incubated in 10 millimolar calcium chloride aggregated, forming contact sites with a pentalaminar membrane structure. These circular attachment sites were free of membrane-associated particles, which accumulated at the periphery. Incubation in 20 millimolar ethylenediaminetetraacetic acid reversed these changes, which are regarded as initial events in the membrane fusion reaction.

Some physical properties of adrenal medulla chromaffin granules isolated by a new continuous iso-osmotic density gradient method.

Centrifugation of crude preparations of chromaffin granules on iso-osmotic continuous gradients of sucrose and metrizamide produce granule fractions of high purity without pelleting. These granules have a buoyant density of 1.123 at 300 mosM, which increases to 1.222 at 1800 mosM. Exchange of water for 2H2O greatly increases the density of the granules. An apparent water space for the granules can be calculated from the difference in density in 2H2O and water. The water space of 0.63 at 300 mosM diminishes as the osmolality of the suspension medium is increased, suggesting that the granule membrane is a good osmometer and that the core contains a large osmotically active water space. Other separation methods for chromaffin granules are discussed in terms of these results.
