Cellular retinoic acid binding protein is associated with mitochondria.

We report that immunohistochemical staining for cellular retinoic acid-binding protein (CRABP) was restricted to the cytoplasm of cortical cells in bovine adrenal. In contrast, staining for the similar protein, cellular retinol-binding protein (CRBP), was found throughout these cells. After transfections of CRABP and CRBP into cultured cells, immunofluorescence analyses again revealed cytoplasmic restriction only for CRABP, with a pronounced punctate appearance. Use of organelle-specific fluorochromes indicated that CRABP immunofluorescence overlaid exactly with the pattern of the mitochondrial-specific fluorochrome. Confirmation of this association came with subcellular fractionation of the adrenal cortex. CRABP, but not CRBP, co-sedimented with the mitochondria, a novel finding for a member of this superfamily of cellular lipid-binding proteins.

Peroxovanadate induces tyrosine phosphorylation of multiple signaling proteins in mouse liver and kidney.

The intraperitoneal injection of a vanadate/H2O2 mixture (peroxovanadate) into mice resulted within minutes in the appearance of numerous tyrosine-phosphorylated proteins in the liver and kidney. These effects are presumably due to the inhibition of phosphotyrosine phosphatase activity. Three of the tyrosine-phosphorylated proteins have been identified as the receptors for epidermal growth factor, insulin, and hepatocyte growth factor. The injection of peroxovanadate also enhanced the tyrosine phosphorylation of many of the proteins known to function downstream of these receptors, including SHC, signal transducer and activator of transcription (Stat) 1alpha,beta, Stat 3, Stat 5, phospholipase C-gamma, insulin receptor substrate 1, GTPase-activating protein, beta-catenin, gamma-catenin, p120cas, SHP-1, and SHP-2. The administration of peroxovanadate also induced nuclear translocation of a number of tyrosine-phosphorylated Stat proteins. In addition, the global effects on tyrosine phosphorylation permitted the detection of a number of novel intracellular protein interactions, including an association of Tyk2 with beta-catenin. The in situ administration of peroxovanadate may prove useful in the search for novel tyrosine-phosphorylated proteins and the identification of new interactions between previously identified tyrosine-phosphorylated substrates.

Induction and activation of Stat 5 in the ovaries of pseudopregnant rats.

PRL acts in the ovary to promote the maintenance and function of the corpus luteum. However, the cellular signals that induce these responses have not been clearly defined. In the present report we demonstrate that Stat 5, previously identified as a transcription factor activated by PRL in the mammary gland, is also activated by PRL in the ovaries of pseudopregnant rats. Intraperitoneal injection of PRL into pseudopregnant rats results in the tyrosine phosphorylation and nuclear translocation of Stat 5. This activated Stat 5 possesses DNA-binding activity for a sequence containing the PRL-inducible element. In addition, we report that luteinization of the PMSG-primed ovary by the administration of hCG is accompanied by an induction of Stat 5 protein.

Magnetic resonance imaging versus angiography in the preoperative assessment of abdominal aortic aneurysms.

Magnetic resonance is a cost-effective imaging tool in the preoperative evaluation of abdominal aortic aneurysm, and yields more clinically relevant information than ultrasonography and computerized tomography. Magnetic resonance imaging has advantages over angiography in that is costs substantially less, is noninvasive, and reconstructs images in multiple planes. By utilizing multiplanar imaging, magnetic resonance imaging yields more information than angiography. The position of the left renal vein is imaged only by magnetic resonance, as is the origin of the superior mesenteric artery. Angiography remains important in patients with associated occlusive disease and claudication. The use of magnetic resonance in the emergent setting is limited at this time.

Hangman's fracture in an infant.

A case is reported of fracture of the pedicles of the axis vertebra in an infant. Non-operative management in a Minerva cast resulted in union and full recovery without complications.

A reappraisal of acute traumatic central cord syndrome.

A review of 77 patients with traumatic central cord syndrome revealed that atypical variations are more common than the existing literature suggests and that these may be seen in a wide variety of acute injuries to the cervical spine. In general the outcome is good; a favourable prognosis on admission is suggested by good hand function, hyperpathia, Lhermitte's sign and normal perianal sensation. The study has highlighted the value of regular muscle charting and has cast doubt on previous neuroanatomical assumptions about the syndrome.

Hormone receptors in male breast carcinoma.

It is well established that approximately 60% of female breast cancers contain estrogen receptors (ER+). The hormonal receptor status of male breast cancers remains relatively unknown. Tumor tissue from 14 patients has been reviewed at the University of Oregon Health Sciences Center. Specimens of all 14 patients were ER+ with a range of 5.5 to 374 fmol/mg protein. Eight of 12 patients had tumors which also contained progesterone receptors (PR+) with a range of 42 to 1852 fmol/mg protein. Nine patients were treated by modified radical mastectomy, two by radical mastectomy, one by simple mastectomy, and two by biopsies only. Twelve patients remain free of disease with a mean follow-up of less than 2 years. The two patients who developed metastases both responded to endocrine ablation by orchiectomy and subsequent adrenalectomy. These data suggest that male breast cancer is highly endocrine sensitive and that endocrine ablation can play an integral part in those men unfortunate enough to develop disseminated disease.

Pediatric diaphragmatic hernias. An 11 year experience.

A retrospective study of 36 infants and children with diaphragmatic hernia was carried out. Mortality was confined to the group of patients identified between birth and 6 hours of life. Survivors appeared to be separable early in life from those who died on the basis of clinical status at birth measured by the Apgar, by ventilatory capacity of the lung reflected by carbon dioxide pressure and by acid base balance. Oxygenation, time from birth surgery, maternal factors, and labor and delivery appeared to play no role in survival. Hypoplasia of the lung reflected in low lung weights in those who died did not correlate with initial clinical status or blood gas data. Surgical adjuncts to reduction and closure of the hernia did not appear to have an effect on survival. Definition of patients at high risk of dying will permit critical application an evaluation of modes of treatment beyond those now used regularly in the care of these desperately ill infants.
