[Systemic breeding of the new variety of Biantiao ginseng (Panax ginseng)].

OBJECTIVE: To breed new varieties of Biantiao ginseng for high yield and fine quality. METHODS: Systemic breeding methods were applied. About 3,000 outstanding Biantiao ginseng roots were selected and planted in breeding field, and self-crossed for four generations. During the course, inferior lines or plants were rejected. Then strain comparison, identification of resistance to black-speck disease, and analysis of active compositions were carried out. RESULTS: "Biantiao 1" (BT1), the first new variety of Biantiao ginseng, with green stems and thick, long, elegant roots and median resistance to black-speck disease, has been harvested since 20 years. The percentage of Biantiao ginseng roots and yield were 15% and 30% higher than the control's respectively. The content of total ginsenosides and the main monomers was 1.8%-2.5% higher than the control's. The characteristics of overground part and root of BT1 were uniform and stable. CONCLUSIONS: BT1, a new excellent ginseng variety, has a good potential value to be generalized in ginseng production.

[Regulation on growth and accumulation of total alkaloid in Leonurus artemisia (Lour.)S.Y. Hu by various microfertilizers].

OBJECTIVE: To trace the effects of various microfertilizers on the growth and accumulation of total alkaloid in Leonurus artemisia. METHOD: The total alkaloid was determined by spectrophotometry. RESULT: Microfertilizers of different concentrations could facilitate the growth of L. artemisia in various degrees and Mn-fertilizer of standard concentration and Cu-fertilizer of excessive concentration could help raise the output of the plant markedly by up to 43.6% and 55.2% respectively. Microfertilizers of different concentrations could improve the content of total alkaloid in L. artemisia: Mn-fertilizer of standard concentration and B-fertilizer of excessive concentration could enhance the contents of total alkaloid up to 2.15% and 2.09% respectively. Mn-fertilizer and Cu-fertilizer of standard concentration have been proved very useful in improving the resistibility and ability of the plant to survive summer safely, and Fe-fertilizer of standard concentration could bring the plant an obvious increase of output after surviving summer. CONCLUSION: Application of Mn-fertilizer of standard concentration is very important for cultivating L. artemisia of excellent quality and high output.

High resolution surface structure of E. coli GroES oligomer by atomic force microscopy.

Using atomic force microscopy (AFM) in aqueous solution, we show that the surface structure of the oligomeric GroES can be obtained up to 10 angstroms resolution. The seven subunits of the heptamer were well resolved without image averaging. The overall dimension of the GroES heptamer was 8.4 +/- 0.4 nm in diameter and 3.0 +/- 0.3 nm high. However, the AFM images further suggest that there is a central protrusion of 0.8 +/- 0.2 nm high and 4.5 +/- 0.4 nm in diameter on one side of GroES which displays a profound seven-fold symmetry. It was found that GroEL could not bind to the adsorbed GroES in the presence of AMP-PNP and Mg2+, suggesting that the side of GroES with the central protrusion faces away from the GroEL lumen, because only one side of GroES was observed under these conditions. Based on the results from both electron and atomic force microscopy, a surface model for the GroES is proposed.
